Abstract: Misfolded endoplasmic reticulum proteins are retro-translocated through the membrane into the cytosol, where they are poly-ubiquitinated, extracted from the membrane, and degraded by the proteasome-a pathway termed endoplasmic reticulum-associated protein degradation (ERAD). Proteins with misfolded domains in the endoplasmic reticulum lumen or membrane are discarded through the ERAD-L and ERAD-M pathways, respectively. In Saccharomyces cerevisiae, both pathways require the ubiquitin ligase Hrd1, a multi-spanning membrane protein with a cytosolic RING finger domain. Hrd1 is the crucial membrane component for retro-translocation, but it is unclear whether it forms a protein-conducting channel. Here we present a cryo-electron microscopy structure of S. cerevisiae Hrd1 in complex with its endoplasmic reticulum luminal binding partner, Hrd3. Hrd1 forms a dimer within the membrane with one or two Hrd3 molecules associated at its luminal side. Each Hrd1 molecule has eight transmembrane segments, five of which form an aqueous cavity extending from the cytosol almost to the endoplasmic reticulum lumen, while a segment of the neighbouring Hrd1 molecule forms a lateral seal. The aqueous cavity and lateral gate are reminiscent of features of protein-conducting conduits that facilitate polypeptide movement in the opposite direction-from the cytosol into or across membranes. Our results suggest that Hrd1 forms a retro-translocation channel for the movement of misfolded polypeptides through the endoplasmic reticulum membrane.

Abstract: Hyperspectral images exhibit strong dependencies across spatial and spectral neighbors, which have been proved to be very useful for hyperspectral image classification. State-of-the-art hyperspectral image classification algorithms use the dependencies in a heuristic way or in probabilistic frameworks but impose unreasonable assumptions on observed data. In this paper, we formulate a conditional random field (CRF) to replace such heuristics and unreasonable assumptions for the classification of hyperspectral images. Moreover, because of avoiding explicit modeling of the observed data, the proposed method can incorporate the classification of hyperspectral images with different statistics characteristics into a unified probabilistic framework. Since the usual classification task for hyperspectral images needs the proposed CRF to be trained on local samples, available global training methods cannot be directly used. Under piecewise training framework, this paper develops an efficient local method to train the CRF. It is efficiently implemented through separated trainings of simple classifiers defined by corresponding potentials. However, the independent classifier trainings may lead to over-counting problems during inference. So we further propose a strategy to combine the independently trained models to obtain final CRF model. Experiments on real-world hyperspectral data show that our algorithm is competitive with the most recent results in hyperspectral image classification.

Abstract: Electron-positron pair creation by multiphoton absorption in the collision of a relativistic electron with a strong laser beam is calculated within laser-dressed quantum electrodynamics. Total production rates, positron spectra, and relative contributions of different reaction channels are obtained in various interaction regimes. We study the process in a manifestly nonperturbative domain which is shown accessible to future experiments utilizing the electron beam lines at novel x-ray laser facilities or all-optical setups based on laser acceleration. Our theory moreover allows us to add further insights into the experimental data from SLAC [D. Burke et al., Phys. Rev. Lett. 79, 1626 (1997).].

Abstract: The present work investigated combustion instabilities inside an ethylene-fueled scramjet combustor mounted on a Mach 2.1 direct-connect test facility with an inflow stagnation temperature of 846 K. Effects of fueling schemes on the combustion stability characteristics were examined. The experimental results suggest that the oscillation modes correlate with mixing status closely. For the cases with a quasi-steady thermal throat or stable shock trains, flame fluctuation exists in a mode of thermo-acoustic type oscillation with a broad frequency range. For the cases with a transient thermal throat, if a fuel/air premixed region from the injection to the cavity flameholder exists, the cavity pilot flame could reignite the fuel/air mixture and undergo a process similar to deflagration–detonation transition (DDT). This process couples with the flame quenching upstream of the injection location, and a DDT-type low frequency oscillation can be formed.
