Abstract: Inference of how evolutionary forces have shaped extant genetic diversity is a cornerstone of modern comparative sequence analysis. Advances in sequence generation and increased statistical sophistication of relevant methods now allow researchers to extract ever more evolutionary signal from the data, albeit at an increased computational cost. Here, we announce the release of Datamonkey 2.0, a completely re-engineered version of the Datamonkey web-server for analyzing evolutionary signatures in sequence data. For this endeavor, we leveraged recent developments in open-source libraries that facilitate interactive, robust, and scalable web application development. Datamonkey 2.0 provides a carefully curated collection of methods for interrogating coding-sequence alignments for imprints of natural selection, packaged as a responsive (i.e. can be viewed on tablet and mobile devices), fully interactive, and API-enabled web application. To complement Datamonkey 2.0, we additionally release HyPhy Vision, an accompanying JavaScript application for visualizing analysis results. HyPhy Vision can also be used separately from Datamonkey 2.0 to visualize locally executed HyPhy analyses. Together, Datamonkey 2.0 and HyPhy Vision showcase how scientific software development can benefit from general-purpose open-source frameworks. Abstract: CRISPR-Cas9 represents a promising platform for genome editing, yet means for its safe and efficient delivery remain to be fully realized. A novel vehicle that simultaneously delivers the Cas9 protein and single guide RNA (sgRNA) is based on DNA nanoclews, yarn-like DNA nanoparticles that are synthesized by rolling circle amplification. The biologically inspired vehicles were efficiently loaded with Cas9/sgRNA complexes and delivered the complexes to the nuclei of human cells, thus enabling targeted gene disruption while maintaining cell viability. Editing was most efficient when the DNA nanoclew sequence and the sgRNA guide sequence were partially complementary, offering a design rule for enhancing delivery. Overall, this strategy provides a versatile method that could be adapted for delivering other DNA-binding proteins or functional nucleic acids.

Abstract: We report data from the first representative national phone survey of Americans' perceptions about nanotechnology (N =1536). Public opinion about nanotechnology is in its infancy, and knowledge about it is quite limited. Yet, Americans' initial reaction to nanotechnology is thus far generally positive, probably rooted in a generally positive view of science overall. Survey respondents expected benefits of nanotechnology to be more prevalent than risks, and they reported feeling hopeful about nanotechnology rather than worried. Their most preferred potential benefit of nanotechnology is “new and better ways to detect and treat human diseases,” and they identified “losing personal privacy to tiny new surveillance devices” as the most important potential risk to avoid. The most discouraging aspect to the data is respondents' lack of trust in business leaders to minimize nanotechnology risks to human health. Overall, these data indicate that while Americans do not necessarily presume benefits and the absence of risks, their outlook is much more positive than not.
