Abstract: The cytosine-guanine (CpG) dinucleotide has long been known to be a hotspot for pathological mutation in the human genome. This hypermutability is related to its role as the major site of cytosine methylation with the attendant risk of spontaneous deamination of 5-methylcytosine (5mC) to yield thymine. Cytosine methylation, however, also occurs in the context of CpNpG sites in the human genome, an unsurprising finding since the intrinsic symmetry of CpNpG renders it capable of supporting a semi-conservative model of replication of the methylation pattern. Recently, it has become clear that significant DNA methylation occurs in a CpHpG context (where H = A, C or T) in a variety of human somatic tissues. If we assume that CpHpG methylation also occurs in the germline, and that 5mC deamination can occur within a CpHpG context, then we might surmise that methylated CpHpG sites could also constitute mutation hotspots causing human genetic disease. To test this postulate, 54,625 missense and nonsense mutations from 2,113 genes causing inherited disease were retrieved from the Human Gene Mutation Database http://www.hgmd.org. Some 18.2 per cent of these pathological lesions were found to be C → T and G → A transitions located in CpG dinucleotides (compatible with a model of methylation-mediated deamination of 5mC), an approximately ten-fold higher proportion than would have been expected by chance alone. The corresponding proportion for the CpHpG trinucleotide was 9.9 per cent, an approximately two-fold higher proportion than would have been expected by chance. We therefore estimate that ~5 per cent of missense/nonsense mutations causing human inherited disease may be attributable to methylation-mediated deamination of 5mC within a CpHpG context.

Abstract: This paper is the first of two papers which will introduce a research approach where groups of directors and senior managers have, not only been observed, but also captured interacting with each other on audio tape recordings. It represents a move from asking board members questions during interviews to seeing and hearing them interactively perform in the boardroom (and elsewhere) over a period of time. When we undertake such ethnographic research what we primarily see is directors and senior managers talking to each other. We suggest that through a close study of their talk-based interpersonal routines, a detailed account of their skills and how factors such as knowledge or know-how and experience are deployed to influence boardroom process is possible. In this first paper our objective is limited to: making a case for a focus upon talk-based interpersonal routines in the boardroom/top management team (TMT); an introductory outline of our theoretical and analytical infrastructure drawn from sociology and; reproducing one illustrative extract of directors’ talk to show what this data ‘looks like’. We conclude by outlining an emerging alternative avenue for developing boards/TMTs in a grounded and reflective fashion.

Abstract: The definitive goal of this research is to develop protein-based scaffolds for use in soft tissue regeneration, particularly in the field of dermal healing. The premise of this investigation was to characterize the mechanical properties of gelatin cross-linked with microbial transglutaminase (mTGase) and to investigate the cytocompatibility of mTGase cross-linked gelatin. Dynamic rheological analysis revealed a significant increase in the storage modulus and thermal stability of gelatin after cross-linking with mTGase. Static, unconfined compression tests showed an increase in Young's modulus of gelatin gels after mTGase cross-linking. A comparable increase in gel strength was observed with 0.03% mTGase and 0.25% glutaraldehyde cross-linked gelatin gels. In vitro studies using 3T3 fibroblasts indicated cytotoxicity at a concentration of 0.05% mTGase after 72 h. However, no significant inhibition of cell proliferation was seen with cells grown on lower concentrations of mTGase cross-linked gelatin substrates. The mechanical improvement and cytocompatibility of mTGase cross-linked gelatin suggests mTGase has potential for use in stabilizing gelatin gels for tissue-engineering applications.

Abstract: Covalent organic frameworks (COFs) have aroused immense scientific interest as an exhilarating class of porous materials due to their structure tunability and diverse properties. However, understanding of their response towards laser induced nonlinear optical (NLO) applications is in its infancy and demands prompt attention. Herein, we report three novel regioregular porphyrin based porous COFs, Por‐COF‐HH and its dual metalated congeners (Por‐COF‐ZnCu and Por‐COF‐ZnNi) with excellent NLO properties. Notably, intensity dependent NLO switching behavior was observed for these Por‐COFs, which is highly desirable for optical switching and optical limiting devices. Moreover, the efficient π‐conjugation and charge transfer transition in ZnCu‐Por‐COF enable a high nonlinear absorption coefficient (β=4470 cm/GW) and figure of merit (FOM = σ1/σo, 3565) values compared to other state‐of‐art materials including molecular porphyrins (β=~100‐400 cm/GW), metal‐organic frameworks (MOFs; β=~0.3‐0.5 cm/GW) and graphene (β=900 cm/GW).
