Abstract: To meet challenges in terms of throughput and turnaround time, many diagnostic laboratories are shifting from Sanger sequencing to higher throughput next-generation sequencing (NGS) platforms. Bearing in mind that the performance and quality criteria expected from NGS in diagnostic or research settings are strikingly different, we have developed an Ion Torrent's PGM-based routine diagnostic procedure for BRCA1/2 sequencing. The procedure was first tested on a training set of 62 control samples, and then blindly validated on 77 samples in parallel with our routine technique. The training set was composed of difficult cases, for example, insertions and/or deletions of various sizes, large-scale rearrangements and, obviously, mutations occurring in homopolymer regions. We also compared two bioinformatic solutions in this diagnostic context, an in-house academic pipeline and the commercially available NextGene software (Softgenetics). NextGene analysis provided higher sensitivity, as four previously undetected single-nucleotide variations were found. Regarding specificity, an average of 1.5 confirmatory Sanger sequencings per patient was needed for complete BRCA1/2 screening. Large-scale rearrangements were identified by two distinct analyses, that is, bioinformatics and fragment analysis with electrophoresis profile comparison. Turnaround time was enhanced, as a series of 30 patients were sequenced by one technician, making the results available for the clinician in 10 working days following blood sampling. BRCA1/2 genes are a good model, representative of the difficulties commonly encountered in diagnostic settings, which is why we believe our findings are of interest for the whole community, and the pipeline described can be adapted by any user of PGM for diagnostic purposes.

Abstract: In this work, numerical simulations for predicting uni- and multi-axial ratchetting behaviors are carried out, using a polycrystal plasticity model. In this multi-scale modeling, the single crystal behavior is based on crystallographic slip (intragranular scale), whereas the polycrystal behavior is obtained from an explicit transition rule to calculate the local stresses and strains (intergranular scale). A systematic study is performed to show the effect of intergranular and intragranular hardening on the ratchetting behavior. For illustrative purposes, two examples are presented: the model is applied to simulate the experimental results from the literature for a 316 austenitic stainless steel and for a 1026 carbon steel. It was demonstrated that the polycrystalline model was successful in describing the inelastic behavior of the two considered materials adequately.

Abstract: Quasi-periodic motions and their stability are addressed from the point of view of different harmonic balance-based approaches. Two numerical methods are used: a generalized multidimensional version of harmonic balance and a modification of a classical solution by harmonic balance. The application to the case of a nonlinear response of a Duffing oscillator under a bi-periodic excitation has allowed a comparison of computational costs and stability evaluation results. The solutions issued from both methods are close to one another and time marching tests showing a good agreement with the harmonic balance results confirm these nonlinear responses. Besides the overall adequacy verification, the observation comparisons would underline the fact that while the 2D approach features better performance in resolution cost, the stability computation turns out to be of more interest to be conducted by the modified 1D approach.

Abstract: The redundancy of multiphase drives provides an inherent fault-tolerant capability that is appreciated in applications with a complicated corrective maintenance or safety-critical requirements. Fault restrictions, however, force the system to be reconfigured to operate in a smooth and efficient manner. Previous works have been focused on the optimization of current waveforms to generate an undisturbed operation but still maintaining the prefault rated flux settings. This study shows that efficient controllers can improve the postfault performance in six-phase induction machines supplied by parallel-connected converters if offline optimization is used to obtain a variable reference flux. Theoretical and experimental results confirm that the proposed flux adaptation method provides higher torque/power capability, lower degree of imbalance in the current sharing between windings and efficiency improvement.
