Abstract: Aurora-A protein kinase, which is the product of an oncogene, is required for the assembly of a functional mitotic apparatus and the regulation of cell ploidy. Overexpression of Aurora-A in tumour cells has been correlated with cancer susceptibility and poor prognosis. Aurora-A activity is required for the recruitment of CDK1-cyclin B1 to the centrosome prior to its activation and the commitment of the cell to mitosis. In this report, we demonstrate that the CDC25B phosphatase, an activator of cyclin dependent kinases at mitosis, is phosphorylated both in vitro and in vivo by Aurora-A on serine 353 and that this phosphorylated form of CDC25B is located at the centrosome during mitosis. Knockdown experiments by RNAi confirm that the centrosome phosphorylation of CDC25B on S353 depends on Aurora-A kinase. Microinjection of antibodies against phosphorylated S353 results in a mitotic delay whilst overexpression of a S353 phosphomimetic mutant enhances the mitotic inducing effect of CDC25B. Our results demonstrate that Aurora-A phosphorylates CDC25B in vivo at the centrosome during mitosis. This phosphorylation might locally participate in the control of the onset of mitosis. These findings re-emphasise the role of the centrosome as a functional integrator of the pathways contributing to the triggering of mitosis.

Abstract: Aim One of the main gaps in the assessment of biodiversity is the lack of a unified framework for measuring its taxonomic and functional facets and for unveiling the underlying patterns. Location Europe, 25 large river basins. Methods Here, we develop a decomposition of functional β-diversity, i.e. the dissimilarity in functional composition between communities, into a functional turnover and a functional nestedness-resultant component. Results We found that functional β-diversity was lower than taxonomic β-diversity. This difference was driven by a lower functional turnover compared with taxonomic turnover while the nestedness-resultant component was similar for taxonomic and functional β-diversity. Main conclusions Fish faunas with different species tend to share the same functional attributes. The framework presented in this paper will help to analyse biogeographical patterns as well as to measure the impact of human activities on the functional facets of biodiversity.

Abstract: In classical Constraint Satisfaction Problems (CSPs) knowledge is embedded in a set of hard constraints, each one restricting the possible values of a set of variables. However constraints in real world problems are seldom hard, and CSP's are often idealizations that do not account for the preference among feasible solutions. Moreover some constraints may have priority over others. Lastly, constraints may involve uncertain parameters. This paper advocates the use of fuzzy sets and possibility theory as a realistic approach for the representation of these three aspects. Fuzzy constraints encompass both preference relations among possible instantiations and priorities among constraints. In a Fuzzy Constraint Satisfaction Problem (FCSP), a constraint is satisfied to a degree (rather than satisfied or not satisfied) and the acceptability of a potential solution becomes a gradual notion. Even if the FCSP is partially inconsistent, best instantiations are provided owing to the relaxation of some constraints. Fuzzy constraints are thus flexible. CSP notions of consistency and k-consistency can be extended to this framework and the classical algorithms used in CSP resolution (e.g., tree search and filtering) can be adapted without losing much of their efficiency. Most classical theoretical results remain applicable to FCSPs. In the paper, various types of constraints are modelled in the same framework. The handling of uncertain parameters is carried out in the same setting because possibility theory can account for both preference and uncertainty. The presence of uncertain parameters leads to ill-defined CSPs, where the set of constraints which defines the problem is not precisely known.

Abstract: Our facial expressions give others the opportunity to access our feelings, and constitute an important nonverbal tool for communication. Many recent studies have investigated emotional perception in adults, and our knowledge of neural processes involved in emotions is increasingly precise. Young children also use faces to express their internal states and perceive emotions in others, but little is known about the neurodevelopment of expression recognition. The goal of the current study was to determine the normal development of facial emotion perception. We recorded ERPs in 82 children 4 to 15 years of age during an implicit processing task with emotional faces. Task and stimuli were the same as those used and validated in an adult study; we focused on the components that showed sensitivity to emotions in adults (P1, N170 and frontal slow wave). An effect of the emotion expressed by faces was seen on the P1 in the youngest children. With increasing age this effect disappeared while an emotional sensitivity emerged on N170. Early emotional processing in young children differed from that observed in the adolescents, who approached adults. In contrast, the later frontal slow wave, although showing typical age effects, was more positive for neutral and happy faces across age groups. Thus, despite the precocious utilization of facial emotions, the neural processing involved in the perception of emotional faces develops in a staggered fashion throughout childhood, with the adult pattern appearing only late in adolescence.
