Abstract: This paper describes the first major attempt to compare seven different inverse approaches for identifying aquifer transmissivity. The ultimate objective was to determine which of several geostatistical inverse techniques is better suited for making probabilistic forecasts of the potential transport of solutes in an aquifer where spatial variability and uncertainty in hydrogeologic properties are significant. Seven geostatistical methods (fast Fourier transform (FF), fractal simulation (FS), linearized cokriging (LC), linearized semianalytical )LS), maximum likelihood (ML), pilot point (PP), and sequential self-calibration (SS)) were compared on four synthetic data sets. Each data set had specific features meeting (or not) classical assumptions about stationarity, amenability to a geostatistical description, etc. The comparison of the outcome of the methods is based on the prediction of travel times and travel paths taken by conservative solutes migrating in the aquifer for a distance of 5 km. Four of the methods, LS, ML, PP, and SS, were identified as being approximately equivalent for the specific problems considered. The magnitude of the variance of the transmissivity fields, which went as high as 10 times the generally accepted range for linearized approaches, was not a problem for the linearized methods when applied to stationary fields; that is, their inverse solutions and travel time predictions were as accurate as those of the nonlinear methods. Nonstationarity of the “true” transmissivity field, or the presence of “anomalies” such as high-permeability fracture zones was, however, more of a problem for the linearized methods. The importance of the proper selection of the semivariogram of the log10 (T) field (or the ability of the method to optimize this variogram iteratively) was found to have a significant impact on the accuracy and precision of the travel time predictions. Use of additional transient information from pumping tests did not result in major changes in the outcome. While the methods differ in their underlying theory, and the codes developed to implement the theories were limited to varying degrees, the most important factor for achieving a successful solution was the time and experience devoted by the user of the method.

Abstract: Mycobacterium abscessus is an emerging rapidly growing mycobacterium (RGM) causing a pseudotuberculous lung disease to which patients with cystic fibrosis (CF) are particularly susceptible. We report here its complete genome sequence. The genome of M. abscessus (CIP 104536T) consists of a 5,067,172-bp circular chromosome including 4920 predicted coding sequences (CDS), an 81-kb full-length prophage and 5 IS elements, and a 23-kb mercury resistance plasmid almost identical to pMM23 from Mycobacterium marinum. The chromosome encodes many virulence proteins and virulence protein families absent or present in only small numbers in the model RGM species Mycobacterium smegmatis. Many of these proteins are encoded by genes belonging to a “mycobacterial” gene pool (e.g. PE and PPE proteins, MCE and YrbE proteins, lipoprotein LpqH precursors). However, many others (e.g. phospholipase C, MgtC, MsrA, ABC Fe(3+) transporter) appear to have been horizontally acquired from distantly related environmental bacteria with a high G+C content, mostly actinobacteria (e.g. Rhodococcus sp., Streptomyces sp.) and pseudomonads. We also identified several metabolic regions acquired from actinobacteria and pseudomonads (relating to phenazine biosynthesis, homogentisate catabolism, phenylacetic acid degradation, DNA degradation) not present in the M. smegmatis genome. Many of the “non mycobacterial” factors detected in M. abscessus are also present in two of the pathogens most frequently isolated from CF patients, Pseudomonas aeruginosa and Burkholderia cepacia. This study elucidates the genetic basis of the unique pathogenicity of M. abscessus among RGM, and raises the question of similar mechanisms of pathogenicity shared by unrelated organisms in CF patients.

Abstract: The position and intensity of the southern westerly wind belt varies seasonally as a consequence of changes in sea surface temperature. During the austral winter, the belt expands northward and the wind intensity in the core decreases. Conversely, during the summer, the belt contracts, and the intensity within the core is strengthened. Reconstructions of the westerly winds since the last glacial maximum, however, have suggested that changes at a single site reflected shifts throughout the entire southern wind belt 1‐4 . Here we use sedimentological and pollen records to reconstruct precipitation patterns over the past 12,500 yr from sites along the windward side of the Andes. Precipitation at the sites, located in the present core and northern margin of the westerlies, is driven almost entirely by the wind belt 5 , and can be used to reconstruct its intensity. Rather than varying coherently throughout the Holocene epoch, we find a distinct anti-phasing of wind strength between the core and northern margin over multi-millennial timescales. During the early Holocene, the core westerlies were strong whereas the northern margin westerlies were weak. We observe the opposite pattern in the late Holocene. As this variation resembles modern seasonal variability, we suggest that our observed changes in westerly wind strength can best be explained by variations in sea surface temperature in the eastern South Pacific Ocean. Abstract: Post-transplant lymphoproliferative disorders (PTLDs) are a group of conditions that involve uncontrolled proliferation of lymphoid cells as a consequence of extrinsic immunosuppression after organ or haematopoietic stem cell transplant. PTLDs show some similarities to classic lymphomas in the non-immunosuppressed general population. The oncogenic Epstein-Barr virus (EBV) is a key pathogenic driver in many early-onset cases, through multiple mechanisms. The incidence of PTLD varies with the type of transplant; a clear distinction should therefore be made between the conditions after solid organ transplant and after haematopoietic stem cell transplant. Recipient EBV seronegativity and the intensity of immunosuppression are among key risk factors. Symptoms and signs depend on the localization of the lymphoid masses. Diagnosis requires histopathology, although imaging techniques can provide additional supportive evidence. Pre-emptive intervention based on monitoring EBV levels in blood has emerged as the preferred strategy for PTLD prevention. Treatment of established disease includes reduction of immunosuppression and/or administration of rituximab (a B cell-specific antibody against CD20), chemotherapy and EBV-specific cytotoxic T cells. Despite these strategies, the mortality and morbidity remains considerable. Patient outcome is influenced by the severity of presentation, treatment-related complications and risk of allograft loss. New innovative treatment options hold promise for changing the outlook in the future.
