Abstract: The ionotropic α-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) receptor is densely distributed in the mammalian brain and is primarily involved in mediating fast excitatory synaptic transmission Recent studies in both heterologous expression systems and cultured neurons have shown that the AMPA receptor can be phosphorylated on their subunits (GluR1, GluR2, and GluR4) All phosphorylation sites reside at serine, threonine, or tyrosine on the intracellular C-terminal domain Several key protein kinases, such as protein kinase A, protein kinase C, Ca2+/calmodulin-dependent protein kinase II, and tyrosine kinases (Trks; receptor or nonreceptor family Trks) are involved in the site-specific regulation of the AMPA receptor phosphorylation Other glutamate receptors (N-methyl-d-aspartate receptors and metabotropic glutamate receptors) also regulate AMPA receptors through a protein phosphorylation mechanism Emerging evidence shows that as a rapid and short-term mechanism, the dynamic protein phosphorylation directly modulates the electrophysiological, morphological (externalization and internalization trafficking and clustering), and biochemical (synthesis and subunit composition) properties of the AMPA receptor, as well as protein-protein interactions between the AMPA receptor subunits and various intracellular interacting proteins These modulations underlie the major molecular mechanisms that ultimately affect many forms of synaptic plasticity

Abstract: This study examined the protective effects of cilostazol on cerebral infarcts produced by subjecting rats to 2-h occlusion of the left middle cerebral artery followed by 24-h reperfusion. The ischemic cerebral infarct consistently involved the cortex and striatum. The infarct size was significantly reduced, when rats received 10 mg/kg cilostazol intravenously 5 min or 1 h after the completion of 2-h ischemia. Cyclic AMP level was significantly elevated in the cortex of 4- and 12-h reperfusion (P < 0.01) following treatment with cilostazol (10 mg/kg, 5 min after 2-h ischemia) accompanied by decreased tumor necrosis factor-alpha level. Samples from the regions corresponding to the penumbra showed markedly reduced Bcl-2 protein level and, in contrast, high levels of Bax protein and cytochrome c release. Cilostazol decreased Bax protein and cytochrome c release and increased the levels of Bcl-2 protein. Cilostazol (10(-7)-10(-5) M) potently and concentration dependently scavenged hydroxyl and peroxyl radicals. In conclusion, cilostazol treatment decreases ischemic brain infarction in association with inhibition of apoptotic and oxidative cell death.

Abstract: A truck scheduling problem for container transportation in a local area with multiple depots and multiple terminals including containers as a resource for transportation is addressed. Four types of movements of containers as inbound full, outbound full, inbound empty and outbound empty movements as well as the time windows at both the origin and the destination are considered. The total operating time of all trucks in operation is taken as the optimization criterion that has to be minimized. The problem is mathematically modeled based on a preparative graph formulation and falls into an extension of the multiple traveling salesman problem with time windows (m-TSPTW). The window partition based solution method for the m-TSPTW in Wang and Regan (Transp Res Part B: Methodol 36:97–112, 2002) is modified so that its computation time is reduced greatly. The experiments based on a number of randomly generated instances indicate that the modified method is quite fast and the quality of solutions is relatively high for the m-TSPTW. These experiments also demonstrate that our approach is able to generate high-quality results for the equivalent truck scheduling and inland container movement problem in container drayage operations.
