Abstract: This study instituted a unique approach to bone tissue engineering by combining effects of mechanical stimulation in the form of fluid shear stresses and the presence of bone-like extracellular matrix (ECM) on osteodifferentiation. Rat marrow stromal cells (MSCs) harvested from bone marrow were cultured on titanium (Ti) fiber mesh discs for 12 days in a flow perfusion system to generate constructs containing bone-like ECM. To observe osteodifferentiation and bone-like matrix deposition, these decellularized constructs and plain Ti fiber meshes were seeded with MSCs (Ti/ECM and Ti, respectively) and cultured in the presence of fluid shear stresses either with or without the osteogenic culture supplement dexamethasone. The calcium content, alkaline phosphatase activity, and osteopontin secretion were monitored as indicators of MSC differentiation. Ti/ECM constructs demonstrated a 75-fold increase in calcium content compared with their Ti counterparts after 16 days of culture. After 16 days, the presence of dexamethasone enhanced the effects of fluid shear stress and the bone-like ECM by increasing mineralization 50-fold for Ti/ECM constructs; even in the absence of dexamethasone, the Ti/ECM constructs exhibited approximately a 40-fold increase in mineralization compared with their Ti counterparts. Additionally, denatured Ti/ECM* constructs demonstrated a 60-fold decrease in calcium content compared with Ti/ECM constructs after 4 days of culture. These results indicate that the inherent osteoinductive potential of bone-like ECM along with fluid shear stresses synergistically enhance the osteodifferentiation of MSCs with profound implications on bone-tissue-engineering applications.

Abstract: Context. Several fast radio bursts have been discovered recently, showing a bright, highly dispersed millisecond radio pulse. The pulses do not repeat and are not associated with a known pulsar or gamma-ray burst. The high dispersion suggests sources at cosmological distances, hence implying an extremely high radio luminosity, far larger than the power of single pulses from a pulsar. Aims. We suggest that a fast radio burst represents the final signal of a supramassive rotating neutron star that collapses to a black hole due to magnetic braking. The neutron star is initially above the critical mass for non-rotating models and is supported by rapid rotation. As magnetic braking constantly reduces the spin, the neutron star will suddenly collapse to a black hole several thousand to million years after its birth. Methods. We discuss several formation scenarios for supramassive neutron stars and estimate the possible observational signatures making use of the results of recent numerical general-relativistic calculations. Results. While the collapse will hide the stellar surface behind an event horizon, the magnetic-field lines will snap violently. This can turn an almost ordinary pulsar into a bright radio “blitzar”: Accelerated electrons from the travelling magnetic shock dissipate a significant fraction of the magnetosphere and produce a massive radio burst that is observable out to z > 0:7. Only a few percent of the neutron stars needs to be supramassive in order to explain the observed rate. Conclusions. We suggest the intriguing possibility that fast radio bursts might trace the solitary and almost silent formation of stellar mass black holes at high redshifts. These bursts could be an electromagnetic complement to gravitational-wave emission and reveal a new formation and evolutionary channel for black holes and neutron stars that are not seen as gamma-ray bursts. If supramassive neutron stars are formed at birth and not by accretion, radio observations of these bursts could trace the core-collapse supernova rate throughout the universe.

Abstract: The diagnosis of rheumatoid arthritis (RA) is primarily based on clinical symptoms, so it is often difficult to diagnose RA in very early stages of the disease. A disease-specific autoantibody that could be used as a serological marker would therefore be very useful. Most autoimmune diseases are characterized by a polyclonal B-cell response targeting multiple autoantigens. These immune responses are often not specific for a single disease. In this review, the most important autoantibody/autoantigen systems associated with RA are described and their utility as a diagnostic and prognostic tool, including their specificity, sensitivity and practical application, is discussed. We conclude that, at present, the antibody response directed to citrullinated antigens has the most valuable diagnostic and prognostic potential for RA.

Abstract: Because the human visual system is continually being bombarded with inputs, it is necessary to have effective mechanisms for filtering out irrelevant information. This is partly achieved by the allocation of attention, allowing the visual system to process relevant input while blocking out irrelevant input. What is the physiological substrate of attentional allocation? It has been proposed that alpha activity reflects functional inhibition. Here we asked if inhibition by alpha oscillations has behavioral consequences for suppressing the perception of unattended input. To this end, we investigated the influence of alpha activity on motion processing in two attentional conditions using magneto-encephalography. The visual stimuli used consisted of two random-dot kinematograms presented simultaneously to the left and right visual hemifields. Subjects were cued to covertly attend the left or right kinematogram. After 1.5 sec, a second cue tested whether subjects could report the direction of coherent motion in the attended (80%) or unattended hemifield (20%). Occipital alpha power was higher contralateral to the unattended side than to the attended side, thus suggesting inhibition of the unattended hemifield. Our key finding is that this alpha lateralization in the 20% invalidly cued trials did correlate with the perception of motion direction: Subjects with pronounced alpha lateralization were worse at detecting motion direction in the unattended hemifield. In contrast, lateralization did not correlate with visual discrimination in the attended visual hemifield. Our findings emphasize the suppressive nature of alpha oscillations and suggest that processing of inputs outside the field of attention is weakened by means of increased alpha activity.
