TL;DR: Details of more than 100 new subtilases discovered in the past five years are summarized, and amino acid sequences of their catalytic domains are compared in a multiple sequence alignment.
Abstract: Subtilases are members of the clan (or superfamily) of subtilisin-like serine proteases. Over 200 subtilases are presently known, more than 170 of which with their complete amino acid sequence. In this update of our previous overview (Siezen RJ, de Vos WM, Leunissen JAM, Dijkstra BW, 1991, Protein Eng 4:719-731), details of more than 100 new subtilases discovered in the past five years are summarized, and amino acid sequences of their catalytic domains are compared in a multiple sequence alignment. Based on sequence homology, a subdivision into six families is proposed. Highly conserved residues of the catalytic domain are identified, as are large or unusual deletions and insertions. Predictions have been updated for Ca(2+)-binding sites, disulfide bonds, and substrate specificity, based on both sequence alignment and three-dimensional homology modeling.

TL;DR: Among patients with stage III epithelial ovarian cancer, the addition of hyperthermic intraperitoneal chemotherapy (HIPEC) to interval cytoreductive surgery resulted in longer recurrence‐free survival and overall survival than surgery alone and did not result in higher rates of side effects.
Abstract: Background Treatment of newly diagnosed advanced-stage ovarian cancer typically involves cytoreductive surgery and systemic chemotherapy. We conducted a trial to investigate whether the addition of hyperthermic intraperitoneal chemotherapy (HIPEC) to interval cytoreductive surgery would improve outcomes among patients who were receiving neoadjuvant chemotherapy for stage III epithelial ovarian cancer. Methods In a multicenter, open-label, phase 3 trial, we randomly assigned 245 patients who had at least stable disease after three cycles of carboplatin (area under the curve of 5 to 6 mg per milliliter per minute) and paclitaxel (175 mg per square meter of body-surface area) to undergo interval cytoreductive surgery either with or without administration of HIPEC with cisplatin (100 mg per square meter). Randomization was performed at the time of surgery in cases in which surgery that would result in no visible disease (complete cytoreduction) or surgery after which one or more residual tumors measu...

31 Jul 2013TL;DR: The Lambda Calculus has been extended with types and used in functional programming (Haskell, Clean) and proof assistants (Coq, Isabelle, HOL), used in designing and verifying IT products and mathematical proofs.
Abstract: This handbook with exercises reveals in formalisms, hitherto mainly used for hardware and software design and verification, unexpected mathematical beauty. The lambda calculus forms a prototype universal programming language, which in its untyped version is related to Lisp, and was treated in the first author's classic The Lambda Calculus (1984). The formalism has since been extended with types and used in functional programming (Haskell, Clean) and proof assistants (Coq, Isabelle, HOL), used in designing and verifying IT products and mathematical proofs. In this book, the authors focus on three classes of typing for lambda terms: simple types, recursive types and intersection types. It is in these three formalisms of terms and types that the unexpected mathematical beauty is revealed. The treatment is authoritative and comprehensive, complemented by an exhaustive bibliography, and numerous exercises are provided to deepen the readers' understanding and increase their confidence using types.

TL;DR: Clinical evidence shows that filgrastim, lenograstim and pegfilgrastIM have clinical efficacy and the use of any of these agents to prevent FN and FN-related complications where indicated and prophylactic G-CSF support is recommended.

TL;DR: The existence of PAD in non‐mammalian vertebrates and the existence of a fifth mammalian PAD are described and tissue‐specific expression, genomic organization and evolutionary conservation of the different PAD isotypes will be discussed in detail.
Abstract: Peptidylarginine deiminase (PAD, EC 3.5.3.15) enzymes catalyze the conversion of protein-bound arginine to citrulline. This post-translational modification may have a big impact on the structure and function of the target protein. In this review, we will discuss the effects of citrullination and its involvement in several human diseases, including rheumatoid arthritis and multiple sclerosis. So far, four isotypes of PAD have been described in mammals. We describe the existence of PAD in non-mammalian vertebrates and the existence of a fifth mammalian PAD. In addition, tissue-specific expression, genomic organization and evolutionary conservation of the different PAD isotypes will be discussed in detail.
