Abstract: Confluent cell monolayers in tissue culture are fragile and can easily be mechanically disrupted, often leaving an area devoid of cells. This opening in the cell sheet is then repopulated, because the cells on the fringe of the damage, which are no longer contact-inhibited, move into the available space. This mechanical disruption is often done deliberately in a "wound-healing" assay as a means to assess the migration of the cells. In such assays, a scrape is made in the cell layer followed by microscopy to monitor the advance of the cells into the wound. We have found that these types of assays can also be accomplished electrically. In this approach, cells growing on small electrodes and monitored by using electric cell-substrate impedance sensing are subjected to currents, resulting in severe electroporation and subsequent cell death. After this invasive treatment, the electrode's impedance is again monitored to chart the migration and ultimate healing of the wound. We report here that this procedure to study cell behavior is both highly reproducible, quantitative, and provides data similar to that acquired with traditional measurements.

Abstract: Numerous cannabinoids have been synthesized that are extremely potent in all of the behavioral assays conducted in our laboratory. An important feature in increasing potency has been the substitution of a dimethylheptyl (DMH) side chain for the pentyl side chain. Our previous studies have shown that (−)-11-OH-Δ8-THC-dimethylheptyl was 80–1150 times more potent than Δ9-THC. Stereospecificity was demonstrated by its (+)- enantiomer which was more than 1400–7500 times less potent. A related series of DMH cannabinoid analogs has recently been synthesized and preliminary evaluations reported here. (−)-11-OH-Δ9-THC-DMH was found to be equipotent with (−)-11-OH-Δ8-THC-DMH. The aldehyde (−)-11-oxo-Δ9-THC-DMH was 15–50 times more potent than Δ9-THC. Surprisingly, (−)-11-carboxy-Δ9-THC-DMH was also active, being slightly more potent than Δ9-THC. In the bicyclic cannabinoid series, the length and bulk of the side chain were found to be equally important. Aminoalkylindoles, which are structurally dissimilar from classical cannabinoids, have been found to exhibit a pharmacological profile similar to Δ9-THC. Though not extremely potent in vivo, they appear to represent an entirely new approach to studying the actions of the cannabinoids. The structural diversity and wide-ranging potencies of the analogs described herein provide the opportunity to develop a pharmacophore for the cannabinoids using molecular modeling techniques.

Abstract: The feasibility of using nitrogen and oxygen isotope ratios of nitrate (NO 3 − ) for elucidating sources and transformations of riverine nitrate was evaluated in a comparative study of 16 watersheds in the northeastern U.S.A. Stream water was sampled repeatedly at the outlets of the watersheds between January and December 1999 for determining concentrations, δ 15N values, and δ 180 values of riverine nitrate.

Abstract: We report the experimental results and theoretical analysis of broadband detection of terahertz (THz) waves via electric-field-induced second-harmonic generation in laser-induced air plasma with ultrashort laser pulses. By introducing the second-harmonic component of the white light in the laser-induced plasma as a local oscillator, coherent detection of broadband THz waves with ambient air is demonstrated for the first time. Our results show that, depending on the probe intensity, detection of THz waves in air can be categorized as incoherent, hybrid, and coherent detection. Coherent detection is achieved only when the tunnel ionization process dominates in gases.
