Abstract: Juglone (5-hydroxy-1,4-naphthoquinone) and plumbagin (5-hydroxy-3-methyl-1,4-naphthoquinone) are yellow pigments found in black walnut (Juglans regia). Herbal preparations derived from black walnut have been used as hair dyes and skin colorants in addition to being applied topically for the treatment of acne, inflammatory diseases, ringworm, and fungal, bacterial, or viral infections. We have studied the cytotoxicity of these quinones to HaCaT keratinocytes. Exposure to juglone or plumbagin (1−20 μM) resulted in a concentration-dependent decrease in cell viability. The cytotoxicity of these quinones is due to two different mechanisms, namely, redox cycling and reaction with glutathione (GSH). Redox cycling results in the generation of the corresponding semiquinone radicals, which were detected by electron paramagnetic resonance. Incubation of keratinocytes with the quinones generated hydrogen peroxide (H2O2) and resulted in the oxidation of GSH to GSSG. Depletion of GSH by buthionine sulfoximine enhanced ...

Abstract: Children and adolescents with obsessive compulsive disorder were studied in an 8-week, multicenter, double-blind, parallel groups trial of clomipramine hydrochloride (CMI) versus placebo. Efficacy assessments included the child version of the Yale-Brown Obsessive Compulsive Scale and the National Institute of Mental Health Global rating scale. At the end of 8 weeks, CMI-treated patients showed a mean reduction in Yale-Brown Obsessive Compulsive Scale score of 37% compared to 8% in the placebo group. Side effects were typical of tricyclic antidepressants. In a 1-year open label treatment, CMI continued to be effective and well tolerated.

Abstract: A four-dimensional data assimilation (FDDA) scheme based on Newtonian relaxation or nudging has been developed and evaluated in the Pennsylvania State University/National Center for Atmospheric Research (PSU/NCAR) Limited-Area Mesoscale Model. It was shown in Part I of this study that continuous assimilation of standard-resolution rawinsonde observations throughout a model integration, rather than at only the initial time, can successfully limit large model error growth (amplitude and phase errors) while the model maintains intervariable consistency and generates realistic mesoscale structures not resolved by the data. The purpose of this paper is to further refine the previously reported FDDA strategy used to produce “dynamic analyses” of the atmosphere by investigating the effects of data assimilation within the planetary boundary layer (PBL).
