Abstract: Temozolomide, an alkylating agent, has shown promise in treating primary central nervous system lymphoma (PCNSL). The enzyme O(6)-methylguanine-DNA methyltransferase (MGMT) repairs alkylating damage, such as that induced by temozolomide. We hypothesized that MGMT immunohistochemistry would predict resistance to temozolomide in PCNSL. A retrospective study of newly-diagnosed and recurrent PCNSL patients treated at our institution was conducted to study the predictive value of MGMT immunohistochemistry for response to temozolomide. 20 patients who were treated with temozolomide as a single agent were identified during the study time period. 6/20 patients demonstrated a response, corresponding to an objective response rate of 30 % (95 % CI 8-52). Five patients with low MGMT level (<30 %) showed a response to temozolomide. Only one of 10 patients (10 %) with high MGMT level (≥30 %) exhibited a response to temozolomide. Small sample numbers precluded formal statistical comparisons. Two patients with complete response remain alive without progressive disease 6.7 and 7.2 years after temozolomide initiation. Immunohistochemistry can be performed on small biopsies to selectively assess MGMT status in tumor versus surrounding inflammation. MGMT analysis by immunohistochemistry may predict response to temozolomide in PCNSL and should be prospectively investigated.

Abstract: Transforming growth factor-beta (TGFbeta) superfamily of growth factors, which include bone morphogenetic proteins (BMPs), have multiple effects in osteoblasts. In this study, we examined the regulation of collagenase-3 promoter activity by BMP-2 in osteoblast-enriched (Ob) cells from fetal rat calvariae. BMP-2 suppressed the activity of a -2 kb collagenase-3 promoter/luciferase recombinant in a time- and dose-dependent manner. The BMP-2 effect on the collagenase-3 promoter was further tested in several collagenase-3 promoter deletion constructs and it was narrowed down to a -148 to -94 nucleotide segment of the promoter containing a runt domain factor 2 (Runx2) site at nucleotide -132 to -126. The effect of BMP-2 was obliterated in a collagenase-3 promoter/luciferase construct containing a mutated Runx2 (mRunx2) sequence indicating that the Runx2 site mediates the BMP-2 response. Electrophoretic mobility shift assays, using nuclear extracts from control and BMP-2-treated Ob cells, indicated that the Runx2 protein is a component of the specific DNA-protein complex formed on the Runx2 site and that the BMP-2 effect may be associated with minor protein modifications rather than major changes in the composition of specific proteins interacting with the Runx2 site. We confirmed that other members of the TGFbeta family can down-regulate the collagenase-3 promoter by showing that TGFbeta1 also suppresses the promoter activity in a time- and dose-dependent manner. In conclusion, this study demonstrates that BMP-2 and TGFbeta1 suppress collagenase-3 promoter activity in osteoblasts and establishes a link between BMP-2 action and collagenase-3 expression via Runx2, a major regulator of osteoblast formation and function.

Abstract: Bemisia tabaci MEAM1 (Gennadius), the sweetpotato whitefly, transmits Tomato yellow leaf curl virus (TYLCV), which causes significant yield losses annually in Florida and other tomato-producing regions. Field trials were carried out at the University of Florida's Gulf Coast Research and Education Center to evaluate integration of plastic mulch type, at-plant insecticide, and tomato variety for management of the sweetpotato whitefly and TYLCV. The tomato varieties Charger, Rally, and Tygress had significantly lower season-long densities of whitefly eggs and nymphs than Florida-47 in one or more trials. Aggressive chemical control measures failed to reduce virus incidence in two of the three trials and did not improve yield. In 2013, when virus pressure was extremely high, yield in the susceptible variety was decimated, but virus-tolerant varieties produced a crop. Egg and nymph densities tended to be lowest on TYLCV-tolerant varieties and on tomato grown on metalized mulch. Differences in yield were primarily due to tomato variety and TYLCV incidence. Intensive insecticide use common in Florida tomato production may reduce losses when viral pressure is moderate, but not when migration of viruliferous whiteflies into the field is constant. Without significant winter freezes or a coordinated host-free period to reduce whitefly populations, insecticidal control and repellent-metalized mulches will offer limited protection compared with genetic tolerance to the virus, which may also be overcome under high virus pressure.
