Abstract: Interleukin-1 (IL-1)alpha is an autocrine/paracrine agent of the skeletal tissue and it regulates bone remodeling. Collagenase-3 or matrix metalloproteinase (MMP)-13 is expressed in osteoblasts and its expression is modulated by several cytokines including IL-1alpha. Because the molecular mechanism of increased synthesis of collagenase-3 in bone cells by IL-1alpha is not known, we investigated if collagenase-3 expression by IL-1alpha in osteoblasts is mediated by transcriptional or post-transcriptional mechanisms. Exposure of rat osteoblastic cultures (Ob cells) to IL-1alpha at concentrations higher than 0.5 nM increased the synthesis of collagenase-3 mRNA up to eightfold and the secretion of immunoreactive protein up to 21-fold. The effects of IL-1alpha on collagenase-3 were time- and dose-dependent. Although prostaglandins stimulate collagenase-3 expression, stimulation of collagenase-3 in Ob cells by IL-1alpha was not mediated through increased biosynthesis of prostaglandins. The half-life of collagenase-3 mRNA from control and IL-1alpha-treated Ob cells was similar suggesting that the stabilization of collagenase-3 mRNA did not contribute to the increase in collagenase-3. However, IL-1alpha stimulated the rate of transcription of the collagenase-3 gene by twofold to fourfold indicating regulation of collagenase-3 expression in Ob cells at the transcriptional level. Stimulation of collagenase-3 by IL-1alpha in osteoblasts may in part mediate the effects of IL-1alpha in bone metabolism.

Abstract: A method of fabricating individualized electron shields with openings corresponding to desired field shapes and sizes is discussed. The electron shields are made from an alloy called Lipowitz' metal. These shields are designed to be mounted on any of six cone sizes provided with the Varian's Clinac 18 linear accelerator. The effect of these shields on electron dosimetry is shown.

Abstract: BACKGROUND: Studies have consistently found that parental and peer drinking behaviors significantly influence adolescent drinking behavior and that adolescent drinking has a significant effect on their drinking-and-driving behavior. Building upon these studies, the present article assesses whether parental and peer drinking behaviors have direct and indirect effects on adolescent drinking and driving as well as whether they moderate the effect of adolescent drinking on their drinking-and-driving behavior. METHODS: The assessment is conducted using data collected from the Buffalo Longitudinal Survey of Young Men (BLSYM) with Ordinary Least Squares (OLS) regression analyses. RESULTS: The data reveal that peer drinking has direct and indirect effects on adolescent drinking-and-driving behavior when adolescent drinking behavior is controlled. It also moderates the effect of adolescent drinking behavior on their drinking and driving. However, parental drinking does not have these direct and interactive effects, although it may have an indirect effect on adolescent drinking and driving via adolescent drinking behavior. CONCLUSIONS: These findings imply that peer drinking behavior should be considered seriously in prevention and intervention for reducing the risk of adolescent drinking-and-driving behavior. Abstract: Background: Technology is increasingly used to enhance pharmacy education. We sought to evaluate student learning and preparedness for community introductory pharmacy practice experiences (IPPEs) after implementation of “MyDispense” into experiential education. Methods: Both first-year pharmacy students and assigned community IPPE preceptors were eligible. Students were stratified based on previous community pharmacy experience (< or ≥ 50 h), then randomized to complete MyDispense exercises before IPPE (group A) or after 24–32 h of IPPE (group B). We evaluated preceptors’ assessment of student readiness using a 6-item Likert scale survey and students’ readiness and opinion of MyDispense using an anonymous 9-item survey. Descriptive statistics were used to characterize data. The Mann–Whitney U test was used to compare groups and a p-value < 0.05 was considered statistically significant. Results: Of 177 eligible students, 155 were randomized and 56 completed study. Group A included 32 students; 56.3% had prior community practice experience. Group B included 24 students; 50% had prior community practice experience. Forty-eight preceptors were enrolled. Students who completed exercises before rotation received higher preceptor scores for patient counseling of self-care and of medications (p < 0.05 for both). Students self-assessed their counseling skills lower than all other skills; 30.4% and 42.9% of students felt mostly or always prepared to counsel for self-care and medications, respectively. Students found MyDispense straightforward, realistic, and appreciated the ability to practice in a safe, electronic, community pharmacy, patient-care environment. Conclusion: Simulation-based software, such as MyDispense, can enhance learner understanding of the prescription fill and counseling process in a community pharmacy practice setting.
