Abstract: The development of sensitive and versatile techniques to detect protein-protein interactions in vivo is important for understanding protein functions. The previously described techniques, fluorescence resonance energy transfer and bimolecular fluorescence complementation, which are used widely for protein-protein interaction studies in plants, require extensive instrumentation. To facilitate protein-protein interaction studies in plants, we adopted the luciferase complementation imaging assay. The amino-terminal and carboxyl-terminal halves of the firefly luciferase reconstitute active luciferase enzyme only when fused to two interacting proteins, and that can be visualized with a low-light imaging system. A series of plasmid constructs were made to enable the transient expression of fusion proteins or generation of stable transgenic plants. We tested nine pairs of proteins known to interact in plants, including Pseudomonas syringae bacterial effector proteins and their protein targets in the plant, proteins of the SKP1-Cullin-F-box protein E3 ligase complex, the HSP90 chaperone complex, components of disease resistance protein complex, and transcription factors. In each case, strong luciferase complementation was observed for positive interactions. Mutants that are known to compromise protein-protein interactions showed little or much reduced luciferase activity. Thus, the assay is simple, reliable, and quantitative in detection of protein-protein interactions in plants.

Abstract: Although universal quantum computers ideally solve problems such as factoring integers exponentially more efficiently than classical machines, the formidable challenges in building such devices motivate the demonstration of simpler, problem-specific algorithms that still promise a quantum speedup. We constructed a quantum boson-sampling machine (QBSM) to sample the output distribution resulting from the nonclassical interference of photons in an integrated photonic circuit, a problem thought to be exponentially hard to solve classically. Unlike universal quantum computation, boson sampling merely requires indistinguishable photons, linear state evolution, and detectors. We benchmarked our QBSM with three and four photons and analyzed sources of sampling inaccuracy. Scaling up to larger devices could offer the first definitive quantum-enhanced computation.

Abstract: Minimization with orthogonality constraints (e.g., $$X^\top X = I$$) and/or spherical constraints (e.g., $$\Vert x\Vert _2 = 1$$) has wide applications in polynomial optimization, combinatorial optimization, eigenvalue problems, sparse PCA, p-harmonic flows, 1-bit compressive sensing, matrix rank minimization, etc. These problems are difficult because the constraints are not only non-convex but numerically expensive to preserve during iterations. To deal with these difficulties, we apply the Cayley transform--a Crank-Nicolson-like update scheme--to preserve the constraints and based on it, develop curvilinear search algorithms with lower flops compared to those based on projections and geodesics. The efficiency of the proposed algorithms is demonstrated on a variety of test problems. In particular, for the maxcut problem, it exactly solves a decomposition formulation for the SDP relaxation. For polynomial optimization, nearest correlation matrix estimation and extreme eigenvalue problems, the proposed algorithms run very fast and return solutions no worse than those from their state-of-the-art algorithms. For the quadratic assignment problem, a gap 0.842 % to the best known solution on the largest problem "tai256c" in QAPLIB can be reached in 5 min on a typical laptop.

Abstract: Although β-CsPbI3 has a bandgap favorable for application in tandem solar cells, depositing and stabilizing β-CsPbI3 experimentally has remained a challenge. We obtained highly crystalline β-CsPbI3 films with an extended spectral response and enhanced phase stability. Synchrotron-based x-ray scattering revealed the presence of highly oriented β-CsPbI3 grains, and sensitive elemental analyses-including inductively coupled plasma mass spectrometry and time-of-flight secondary ion mass spectrometry-confirmed their all-inorganic composition. We further mitigated the effects of cracks and pinholes in the perovskite layer by surface treating with choline iodide, which increased the charge-carrier lifetime and improved the energy-level alignment between the β-CsPbI3 absorber layer and carrier-selective contacts. The perovskite solar cells made from the treated material have highly reproducible and stable efficiencies reaching 18.4% under 45 ± 5°C ambient conditions.
