Abstract: Abnormal epigenetic regulation has been implicated in oncogenesis. We report here the identification of somatic mutations by exome sequencing in acute monocytic leukemia, the M5 subtype of acute myeloid leukemia (AML-M5). We discovered mutations in DNMT3A (encoding DNA methyltransferase 3A) in 23 of 112 (20.5%) cases. The DNMT3A mutants showed reduced enzymatic activity or aberrant affinity to histone H3 in vitro. Notably, there were alterations of DNA methylation patterns and/or gene expression profiles (such as HOXB genes) in samples with DNMT3A mutations as compared with those without such changes. Leukemias with DNMT3A mutations constituted a group of poor prognosis with elderly disease onset and of promonocytic as well as monocytic predominance among AML-M5 individuals. Screening other leukemia subtypes showed Arg882 alterations in 13.6% of acute myelomonocytic leukemia (AML-M4) cases. Our work suggests a contribution of aberrant DNA methyltransferase activity to the pathogenesis of acute monocytic leukemia and provides a useful new biomarker for relevant cases.

Abstract: The distributed energy resources (DER) comprise several technologies, such as diesel engines, micro turbines, fuel cells, photovoltaic, small wind turbines, etc. The coordinated operation and control of DER together with controllable loads and storage devices, such as flywheels, energy capacitors and batteries are central to the concept of MicroGrid (MG). MG can operate interconnected to the main distribution grid, or in an islanded mode. This paper reviews the researches and studies on MG technology. The operation of MG and the MG in the market environment are also described in the paper.

Abstract: The synthesis of water-soluble nitrogen-doped carbon dots has received great attention, due to their wide applications in oxygen reduction reaction, cell imaging, sensors, and drug delivery. Herein, nitrogen-doped, carbon-rich, highly photoluminescent carbon dots have been synthesized for the first time from ammonium citrate under hydrothermal conditions. The obtained nitrogen-doped carbon dots possess bright blue luminescence, short fluorescence lifetime, pH-sensitivity and excellent stability at a high salt concentration. They have potential to be used for pH sensors, cell imaging, solar cells, and photocatalysis.

Abstract: Sirtuins such as SIRT1 are conserved protein NAD(+)-dependent deacylases and thus their function is intrinsically linked to cellular metabolism. Over the past two decades, accumulating evidence has indicated that sirtuins are not only important energy status sensors but also protect cells against metabolic stresses. Sirtuins regulate the aging process and are themselves regulated by diet and environmental stress. The versatile functions of sirtuins including, more specifically, SIRT1 are supported by their diverse cellular location allowing cells to sense changes in energy levels in the nucleus, cytoplasm, and mitochondrion. SIRT1 plays a critical role in metabolic health by deacetylating many target proteins in numerous tissues, including liver, muscle, adipose tissue, heart, and endothelium. This sirtuin also exerts important systemic effects via the hypothalamus. This review will cover these topics and suggest that strategies to maintain sirtuin activity may be on the horizon to forestall diseases of aging.
