Abstract: The substantial antimicrobial efficacy of nanoparticles against phytopathogens has been extensively investigated for advanced agricultural applications. However, few reports have focused on soilborne pathogenic bacteria. The aim of this study was to obtain sustainably synthesized copper oxide nanoparticles (CuONPs) using papaya leaf extracts and investigate the bactericidal activity of these CuONPs against Ralstonia solanacearum, the cause of bacterial wilt, under laboratory and greenhouse conditions. The results showed that CuONPs possessed strong antibacterial activity and that all R. solanacearum were killed after exposure to 250 μg mL−1 CuONPs. CuONPs could interact with bacterial cells to prevent biofilm formation, reduce swarming motility and disturb ATP production. Ultrastructural observations by transmission electron microscopy (TEM) revealed that after interactions with CuONPs, bacterial cells suffered significantly from nanomechanical damage to the cytomembrane, accompanied by the absorption of multiple nanoparticles. In addition, molecular studies identified the downregulation mechanism of a series of genes involving pathogenesis and motility. The control efficiency of CuONPs in tobacco bacterial wilt disease management under greenhouse conditions was verified by root irrigation application, demonstrating that as-prepared CuONPs significantly reduced the disease occurrence and disease index. Our studies focused on developing biosynthesized nanoparticles as a biocompatible alternative for soilborne disease management.

Abstract: We investigated flexible liposomes as a potential oral drug delivery system. However, enhanced membrane fluidity and structural deformability may necessitate liposomal surface modification when facing the harsh environment of the gastrointestinal tract. In the present study, silica-coated flexible liposomes loaded with curcumin (CUR-SLs) having poor water solubility as a model drug were prepared by a thin-film method with homogenization, followed by the formation of a silica shell by the sol-gel process. We systematically investigated the physical properties, drug release behavior, pharmacodynamics, and bioavailability of CUR-SLs. CUR-SLs had a mean diameter of 157 nm and a polydispersity index of 0.14, while the apparent entrapment efficiency was 90.62%. Compared with curcumin-loaded flexible liposomes (CUR-FLs) without silica-coatings, CUR-SLs had significantly higher stability against artificial gastric fluid and showed more sustained drug release in artificial intestinal fluid as determined by in vitro release assays. The bioavailability of CUR-SLs and CUR-FLs was 7.76- and 2.35-fold higher, respectively, than that of curcumin suspensions. Silica coating markedly improved the stability of flexible liposomes, and CUR-SLs exhibited a 3.31-fold increase in bioavailability compared with CUR-FLs, indicating that silica-coated flexible liposomes may be employed as a potential carrier to deliver drugs with poor water solubility via the oral route with improved bioavailability.

Abstract: Carbon dots doped with germanium (GeCDs) were firstly prepared by a new simple 15 min carbonation synthesis route, exhibiting excitation-independent photoluminescence (PL), which could avoid autofluorescence in bioimaging applications. The as-prepared GeCDs have low cell toxicity, good biocompatibility, high intracellular delivery efficiency, stability and could be applied for detection of mercury(II) ions with excellent selectivity in complicated medium. It is to be noted that the as-prepared GeCDs used as a new type of probe for visualization of dynamic invasions of mercury(II) ions into Hep-2 cells display greatly different properties from most of the previously reported CDs which are regularly responsive to iron ions. All the results suggest that the GeCDs can be employed for visualization and monitoring of the significant physiological changes of living cells induced by Hg2+.
