Abstract: Cancer immunotherapy based on genetically redirecting T cells has been used successfully to treat B cell malignancies1-3. In this strategy, the T cell genome is modified by integration of viral vectors or transposons encoding chimaeric antigen receptors (CARs) that direct tumour cell killing. However, this approach is often limited by the extent of expansion and persistence of CAR T cells4,5. Here we report mechanistic insights from studies of a patient with chronic lymphocytic leukaemia treated with CAR T cells targeting the CD19 protein. Following infusion of CAR T cells, anti-tumour activity was evident in the peripheral blood, lymph nodes and bone marrow; this activity was accompanied by complete remission. Unexpectedly, at the peak of the response, 94% of CAR T cells originated from a single clone in which lentiviral vector-mediated insertion of the CAR transgene disrupted the methylcytosine dioxygenase TET2 gene. Further analysis revealed a hypomorphic mutation in this patient's second TET2 allele. TET2-disrupted CAR T cells exhibited an epigenetic profile consistent with altered T cell differentiation and, at the peak of expansion, displayed a central memory phenotype. Experimental knockdown of TET2 recapitulated the potency-enhancing effect of TET2 dysfunction in this patient's CAR T cells. These findings suggest that the progeny of a single CAR T cell induced leukaemia remission and that TET2 modification may be useful for improving immunotherapies.

Abstract: Pandemic influenza A viruses that emerge from animal reservoirs are inevitable. Therefore, rapid genomic analysis and creation of vaccines are vital. We developed a multisegment reverse transcription-PCR (M-RTPCR) approach that simultaneously amplifies eight genomic RNA segments, irrespective of virus subtype. M-RTPCR amplicons can be used for high-throughput sequencing and/or cloned into modified reverse-genetics plasmids via regions of sequence identity. We used these procedures to rescue a contemporary H3N2 virus and a swine origin H1N1 virus directly from human swab specimens. Together, M-RTPCR and the modified reverse-genetics plasmids that we designed streamline the creation of vaccine seed stocks (9 to 12 days).

Abstract: Demand-side management, together with the integration of distributed energy generation and storage, are considered increasingly essential elements for implementing the smart grid concept and balancing massive energy production from renewable sources. We focus on a smart grid in which the demand-side comprises traditional users as well as users owning some kind of distributed energy sources and/or energy storage devices. By means of a day-ahead optimization process regulated by an independent central unit, the latter users intend to reduce their monetary energy expense by producing or storing energy rather than just purchasing their energy needs from the grid. In this paper, we formulate the resulting grid optimization problem as a noncooperative game and analyze the existence of optimal strategies. Furthermore, we present a distributed algorithm to be run on the users' smart meters, which provides the optimal production and/or storage strategies, while preserving the privacy of the users and minimizing the required signaling with the central unit. Finally, the proposed day-ahead optimization is tested in a realistic situation.

Abstract: We report the findings of a meta-analytic review of experimental studies concerned with the biasing effect of physical attractiveness on a variety of job-related outcomes. In support of implicit personality theory, attractive individuals were found to fare better than unattractive individuals in terms of a number of such outcomes. The weighted mean effect size, d, was .37 for all studies. In addition, tests for moderating effects showed that (a) the attractiveness bias did not differ between studies that provided low versus high amounts of job-relevant information about the targets, (b) the same bias was greater for within-subjects research designs than for between-subjects designs, (c) professionals were as susceptible to the bias as were college students, (d) attractiveness was as important for men as for women, and (e) the biasing effect of attractiveness has decreased in recent years. Implications of these findings are considered.
