Abstract: More than 20 monoclonal antibodies have been approved as therapeutic drugs by the US Food and Drug Administration, and it is quite likely that the number of approved antibodies will double in the next 7-10 years. Antibody drugs show several desirable characteristics, including good solubility and stability, long persistence in the body, high selectivity and specificity, and low risk for bioconversion to toxic metabolites. However, many antibody drugs demonstrate attributes that complicate drug development, including very poor oral bioavailability, incomplete absorption following intramuscular or subcutaneous administration, nonlinear distribution, and nonlinear elimination. In addition, antibody administration often leads to an endogenous antibody response, which may alter the pharmacokinetics and efficacy of the therapeutic antibody. Antibodies have been developed for a wide range of disease conditions, with effects produced through a complex array of mechanisms. This article attempts to provide a brief overview of the main determinants of antibody pharmacokinetics and pharmacodynamics. Abstract: This article reviews the literature on the relationship among parenting practices, parenting styles, and adolescent school achievement. The review of the empirical research indicates that parental involvement and monitoring are robust predictors of adolescent achievement. Several studies, however, indicate that parental involvement declines in adolescence, prompting the call for future research on the reasons for and associated consequences of this decline. Furthermore, the review indicates that authoritative parenting styles are often associated with higher levels of student achievement, although these findings are not consistent across culture, ethnicity, and socioeconomic status. Darling and Steinberg’s contextual model of parenting provides a promising model to help resolve these discrepancies, however, further research is needed to examine the major linkages of the model. It is also argued that the contextual model should expand its notion of context towards the larger cultural and economic context in which families reside.

Abstract: Hypoxic-ischemic injury to the periventricular cerebral white matter [periventricular leukomalacia (PVL)] results in cerebral palsy and is the leading cause of brain injury in premature infants. The principal feature of PVL is a chronic disturbance of myelination and suggests that oligodendrocyte (OL) lineage progression is disrupted by ischemic injury. We determined the OL lineage stages at risk for injury during the developmental window of vulnerability for PVL (23–32 weeks, postconceptional age). In 26 normal control autopsy human brains, OL lineage progression was defined in parietal white matter, a region of predilection for PVL. Three successive OL stages, the late OL progenitor, the immature OL, and the mature OL, were characterized between 18 and 41 weeks with anti-NG2 proteoglycan, O4, O1, and anti-myelin basic protein (anti-MBP) antibodies. NG2+O4+ late OL progenitors were the predominant stage throughout the latter half of gestation. Between 18 and 27 weeks, O4+O1+ immature OLs were a minor population (9.9 ± 2.1% of total OLs; n = 9). Between 28 and 41 weeks, an increase in immature OLs to 30.9 ± 2.1% of total OLs (n = 9) was accompanied by a progressive increase in MBP+ myelin sheaths that were restricted to the periventricular white matter. The developmental window of high risk for PVL thus precedes the onset of myelination and identifies the late OL progenitor as the major potential target. Moreover, the decline in incidence of PVL at ∼32 weeks coincides with the onset of myelination in the periventricular white matter and suggests that the risk for PVL is related to the presence of late OL progenitors in the periventricular white matter.

Abstract: We present photometric observations of an apparent Type Ia supernova (SN Ia) at a redshift of ~1.7, the farthest SN observed to date. The supernova, SN 1997ff, was discovered in a repeat observation by the Hubble Space Telescope (HST) of the Hubble Deep Field-North (HDF-N) and serendipitously monitored with NICMOS on HST throughout the Thompson et al. Guaranteed-Time Observer (GTO) campaign. The SN type can be determined from the host galaxy type: an evolved, red elliptical lacking enough recent star formation to provide a significant population of core-collapse supernovae. The classification is further supported by diagnostics available from the observed colors and temporal behavior of the SN, both of which match a typical SN Ia. The photometric record of the SN includes a dozen flux measurements in the I, J, and H bands spanning 35 days in the observed frame. The redshift derived from the SN photometry, z = 1.7 ± 0.1, is in excellent agreement with the redshift estimate of z = 1.65 ± 0.15 derived from the U300B450V606I814J110J125H160H165Ks photometry of the galaxy. Optical and near-infrared spectra of the host provide a very tentative spectroscopic redshift of 1.755. Fits to observations of the SN provide constraints for the redshift-distance relation of SNe Ia and a powerful test of the current accelerating universe hypothesis. The apparent SN brightness is consistent with that expected in the decelerating phase of the preferred cosmological model, ΩM ≈ 1/3,ΩΛ ≈ . It is inconsistent with gray dust or simple luminosity evolution, candidate astrophysical effects that could mimic previous evidence for an accelerating universe from SNe Ia at z ≈ 0.5. We consider several sources of potential systematic error, including gravitational lensing, supernova misclassification, sample selection bias, and luminosity calibration errors. Currently, none of these effects alone appears likely to challenge our conclusions. Additional SNe Ia at z > 1 will be required to test more exotic alternatives to the accelerating universe hypothesis and to probe the nature of dark energy.
