Abstract: Exploring advanced porous materials is of critical importance in the development of science and technology. Porous polymers, being famous for their all-organic components, tailored pore structures, and adjustable chemical components, have attracted an increasing level of research interest in a large number of applications, including gas adsorption/storage, separation, catalysis, environmental remediation, energy, optoelectronics, and health. Recent years have witnessed tremendous research breakthroughs in these fields thanks to the unique pore structures and versatile skeletons of porous polymers. Here, recent milestones in the diverse applications of porous polymers are presented, with an emphasis on the structural requirements or parameters that dominate their properties and functionalities. The Review covers the following applications: i) gas adsorption, ii) water treatment, iii) separation, iv) heterogeneous catalysis, v) electrochemical energy storage, vi) precursors for porous carbons, and vii) other applications (e.g., intelligent temperature control textiles, sensing, proton conduction, biomedicine, optoelectronics, and actuators). The key requirements for each application are discussed and an in-depth understanding of the structure-property relationships of these advanced materials is provided. Finally, a perspective on the future research directions and challenges in this field is presented for further studies.

Abstract: Although 5-methylcytosine (m5C) is a widespread modification in RNAs, its regulation and biological role in pathological conditions (such as cancer) remain unknown. Here, we provide the single-nucleotide resolution landscape of messenger RNA m5C modifications in human urothelial carcinoma of the bladder (UCB). We identify numerous oncogene RNAs with hypermethylated m5C sites causally linked to their upregulation in UCBs and further demonstrate YBX1 as an m5C 'reader' recognizing m5C-modified mRNAs through the indole ring of W65 in its cold-shock domain. YBX1 maintains the stability of its target mRNA by recruiting ELAVL1. Moreover, NSUN2 and YBX1 are demonstrated to drive UCB pathogenesis by targeting the m5C methylation site in the HDGF 3' untranslated region. Clinically, a high coexpression of NUSN2, YBX1 and HDGF predicts the poorest survival. Our findings reveal an unprecedented mechanism of RNA m5C-regulated oncogene activation, providing a potential therapeutic strategy for UCB.

Abstract: We present CodeBERT, a bimodal pre-trained model for programming language (PL) and natural language (NL). CodeBERT learns general-purpose representations that support downstream NL-PL applications such as natural language code search, code documentation generation, etc. We develop CodeBERT with Transformer-based neural architecture, and train it with a hybrid objective function that incorporates the pre-training task of replaced token detection, which is to detect plausible alternatives sampled from generators. This enables us to utilize both “bimodal” data of NL-PL pairs and “unimodal data, where the former provides input tokens for model training while the latter helps to learn better generators. We evaluate CodeBERT on two NL-PL applications by fine-tuning model parameters. Results show that CodeBERT achieves state-of-the-art performance on both natural language code search and code documentation generation. Furthermore, to investigate what type of knowledge is learned in CodeBERT, we construct a dataset for NL-PL probing, and evaluate in a zero-shot setting where parameters of pre-trained models are fixed. Results show that CodeBERT performs better than previous pre-trained models on NLPL probing.

Abstract: Circular RNAs are a class of regulatory RNA transcripts, which are ubiquitously expressed in eukaryotes. In the current study, we evaluate the function of a novel circRNA derived from the β-catenin gene locus, circβ-catenin. Circβ-catenin is predominantly localized in the cytoplasm and displays resistance to RNase-R treatment. We find that circβ-catenin is highly expressed in liver cancer tissues when compared to adjacent normal tissues. Silencing of circβ-catenin significantly suppresses malignant phenotypes in vitro and in vivo, and knockdown of this circRNA reduces the protein level of β-catenin without affecting its mRNA level. We show that circβ-catenin affects a wide spectrum of Wnt pathway-related genes, and furthermore, circβ-catenin produces a novel 370-amino acid β-catenin isoform that uses the start codon as the linear β-catenin mRNA transcript and translation is terminated at a new stop codon created by circularization. We find that this novel isoform can stabilize full-length β-catenin by antagonizing GSK3β-induced β-catenin phosphorylation and degradation, leading to activation of the Wnt pathway. Our findings illustrate a non-canonical function of circRNA in modulating liver cancer cell growth through the Wnt pathway, which can provide novel mechanistic insights into the underlying mechanisms of hepatocellular carcinoma.

Abstract: A new fabrication strategy of the graphene-coated solid-phase microextraction (SPME) fiber is developed. Graphite oxide was first used as starting coating material that covalently bonded to the fused-silica substrate using 3-aminopropyltriethoxysilane (APTES) as cross-linking agent and subsequently deoxidized by hydrazine to give the graphene coating in situ. The chemical bonding between graphene and the silica fiber improve its chemical stability, and the obtained fiber was stable enough for more than 150 replicate extraction cycles. The graphene coating was wrinkled and folded, like the morphology of the rough tree bark. Its performance is tested by headspace (HS) SPME of polycyclic aromatic hydrocarbons (PAHs) followed by GC/MS analysis. The results showed that the graphene-coated fiber exhibited higher enrichment factors (EFs) from 2-fold for naphthalene to 17-fold for B(b)FL as compared to the commercial polydimethylsioxane (PDMS) fiber, and the EFs increased with the number of condensed rings of PAHs. The strong adsorption affinity was believed to be mostly due to the dominant role of π-π stacking interaction and hydrophobic effect, according to the results of selectivity study for a variety of organic compounds including PAHs, the aromatic compounds with different substituent groups, and some aliphatic hydrocarbons. For PAHs analysis, the graphene-coated fiber showed good precision (<11%), low detection limits (1.52-2.72 ng/L), and wide linearity (5-500 ng/L) under the optimized conditions. The repeatability of fiber-to-fiber was 4.0-10.8%. The method was applied to simultaneous analysis of eight PAHs with satisfactory recoveries, which were 84-102% for water samples and 72-95% for soil samples, respectively.
