Trace analysis of protein amino acids as n(o, s)-isobutyloxycarbonyl tert.-butyldimethylsilyl derivatives in aqueous samples

Abstract: N(O, S)-isoButyloxycarbonyl(-isoEOC) derivatization with subsequent solid-phase extraction and silylation was investigated for the trace analysis of protein amino acids in complex aqueous samples. The amino acids in basic aqueous medium were allowed to react with isobutyl chloroformate. After acidification the resulting N(O, S)-isoBOC amino acids were solid-phase extracted using Chromosorb P as solid sorbent and then converted to tert.-butyldimethylsilyl derivatives, followed by direct gas chromatographic analysis. The application of the method to the amino acid profiling of various complex samples is demonstrated.
