Abstract: Many classes of silver nanoparticles (Ag-NPs) have been synthesized and widely applied, but the genotoxicity of Ag-NPs and the factors leading to genotoxicity remain unknown. Therefore, the purpose of this study is to elucidate the genotoxic effects of Ag-NPs in lung and the role of oxidative stress on the genotoxic effects of Ag-NPs. For this, Ag-NPs were completely dispersed in medium by sonication and filtration. The Ag-NPs dispersed in medium were 43-260nm in size. We observed distinct uptake of Ag-NPs into BEAS-2B cells. The Ag-NPs aggregates were wrapped with an endocytic vesicle within the cytoplasm and nucleus of BEAS-2B cells. In the comet assay and micronucleus (MN) assay for BEAS-2B cells, Ag-NPs stimulated DNA breakage and MN formation in a dose-dependent manner. The genotoxic effect of Ag-NPs was partially blocked by scavengers. In particular, of the scavengers tested, superoxide dismutase most significantly blocked the genotoxic effects in both the cytokinesis-block MN assay and the comet assay. In the modified comet assay, Ag-NPs induced a significant increase in oxidative DNA damage. Furthermore, in the oxidative stress assay, Ag-NPs significantly increased the reactive oxygen radicals. These results suggest that Ag-NPs have genotoxic effects in BEAS-2B cells and that oxidative stress stimulated by Ag-NPs may be an important factor in their genotoxic effects.

Abstract: Numerous cell types have shown a remarkable ability to detect and move along gradients in stiffness of an underlying substrate—a process known as durotaxis The mechanisms underlying durotaxis are still unresolved, but generally believed to involve active sensing and locomotion Here, we show that simple liquid droplets also undergo durotaxis By modulating substrate stiffness, we obtain fine control of droplet position on soft, flat substrates Unlike other control mechanisms, droplet durotaxis works without imposing chemical, thermal, electrical, or topographical gradients We show that droplet durotaxis can be used to create large-scale droplet patterns and is potentially useful for many applications, such as microfluidics, thermal control, and microfabrication

Abstract: Ambient backscatter communication technology has been introduced recently, and is quickly becoming a promising choice for self-sustainable communication systems, as an external power supply or a dedicated carrier emitter is not required. By leveraging existing RF signal resources, ambient backscatter technology can support sustainable and independent communications and consequently open up a whole new set of applications that facilitate Internet of things (IoT). In this article, we study an integration of ambient backscatter with wireless powered communication networks (WPCNs). We first present an overview of backscatter communication systems with an emphasis on the emerging ambient backscatter technology. Then we propose a novel hybrid transmitter design by combining the advantages of both ambient backscatter and wireless powered communications. Furthermore, in the cognitive radio environment, we introduce a multiple access scheme to coordinate hybrid data transmissions. The performance evaluation shows that the hybrid transmitter outperforms traditional designs. In addition, we discuss open issues related to ambient backscatter networking.

Abstract: Here, we demonstrate that electroporation-enhanced immunization with a rationally designed HPV DNA vaccine (GX-188E), preferentially targeting HPV antigens to dendritic cells, elicits a significant E6/E7-specific IFN-γ-producing T-cell response in all nine cervical intraepithelial neoplasia 3 (CIN3) patients. Importantly, eight out of nine patients exhibit an enhanced polyfunctional HPV-specific CD8 T-cell response as shown by an increase in cytolytic activity, proliferative capacity and secretion of effector molecules. Notably, seven out of nine patients display complete regression of their lesions and viral clearance within 36 weeks of follow up. GX-188E administration does not elicit serious vaccine-associated adverse events at all administered doses. These findings indicate that the magnitude of systemic polyfunctional CD8 T-cell response is the main contributing factor for histological, cytological and virological responses, providing valuable insights into the design of therapeutic vaccines for effectively treating persistent infections and cancers in humans.
