Abstract: Conflicts between biodiversity conservation and human activities are becoming increasingly apparent in all European landscapes. The intensification of agricultural and silvicultural practices, land abandonment and other land uses such as recreation and hunting are all potential threats to biodiversity that can lead to conflicts between stakeholder livelihoods and biodiversity conservation. To address the global decline in biodiversity there is, therefore, a need to identify the drivers responsible for conflicts between human activities and the conservation of European biodiversity and to promote the management of these conflicts. Here, the drivers of biodiversity conflicts are analysed in a European context for five habitat types: agricultural landscapes, forests, grasslands, uplands and freshwater habitats. A multi- disciplinary approach to conflict management is described, with active stakeholder involvement at every stage of conflict identification and management as well as a range of other approaches including stakeholder dialogue and education, consumer education, improvement of political and legislative frameworks, financial incentives, and planning infrastructure.

Abstract: We evaluated the antagonistic effects of newly identified lactic acid bacteria (LAB) in the genera Lactobacillus and Bifidobacterium, originating from the honey stomach, on the honey bee pathogen, Paenibacillus larvae. We used inhibition assays on agar plates and honey bee larval bioassays to investigate the effects of honey bee LAB on P. larvae growth in vitro and on AFB infection in vivo. The individual LAB phylotypes showed different inhibition properties against P. larvae growth on agar plates, whereas a combination of all eleven LAB phylotypes resulted in a total inhibition (no visible growth) of P. larvae. Adding the LAB mixture to the larval food significantly reduced the number of AFB infected larvae in exposure bioassays. The results demonstrate that honey bee specific LAB possess beneficial properties for honey bee health. Possible benefits to honey bee health by enhancing growth of LAB or by applying LAB to honey bee colonies should be further investigated.

Abstract: A new pathway for triacylglycerol biosynthesis involving a phospholipid:diacylglycerol acyltransferase (PDAT) was recently described (Dahlqvist A, Stahl U, Lenman M, Banas A, Lee M, Sandager L, Ronne H, Stymne S, [2000] Proc Natl Acad Sci USA 97: 6487-6492). The LRO1 gene that encodes the PDAT was identified in yeast (Saccharomyces cerevisiae) and shown to have homology with animal lecithin:cholesterol acyltransferase. A search of the Arabidopsis genome database identified the protein encoded by the At5g13640 gene as the closest homolog to the yeast PDAT (28% amino acid identity). The cDNA of At5g13640 (AtPDAT gene) was overexpressed in Arabidopsis behind the cauliflower mosaic virus promoter. Microsomal preparations of roots and leaves from overexpressers had PDAT activities that correlated with expression levels of the gene, thus demonstrating that this gene encoded PDAT (AtPDAT). The AtPDAT utilized different phospholipids as acyl donor and accepted acyl groups ranging from C10 to C22. The rate of activity was highly dependent on acyl composition with highest activities for acyl groups containing several double bonds, epoxy, or hydroxy groups. The enzyme utilized both sn-positions of phosphatidylcholine but had a 3-fold preference for the sn-2 position. The fatty acid and lipid composition as well as the amounts of lipids per fresh weight in Arabidopsis plants overexpressing AtPDAT were not significantly different from the wild type. Microsomal preparations of roots from a T-DNA insertion mutant in the AtPDAT gene had barely detectable capacity to transfer acyl groups from phospholipids to added diacylglycerols. However, these microsomes were still able to carry out triacylglycerol synthesis by a diacylglycerol:diacylglycerol acyltransferase reaction at the same rate as microsomal preparations from wild type.

Abstract: When pathways for groundwater flow are defined in grid based elevation models, the grid structure creates some obstacles as it only allows for eight flow directions. Earlier algorithms have used arbitrarily designed flow distributions, which have resulted in either too diverging or too converging flow patterns. This study defines a computationally simple distribution function, where the converging nature (x) can be altered. A material of reference flow distributions was produced through simulation in four elevation models. The distribution function was tested for different values on x, against the reference. The results show a stable correlation pattern between the tested function and the reference. Optimum values on x were found, which suggest that the flow distribution from a grid cell should be weighted with the slope gradient raised to the power of 4–6.
