Abstract: Although bacterial polyketides are of considerable biomedical interest, the molecular biology of polyketide biosynthesis in Bacillus spp., one of the richest bacterial sources of bioactive natural products, remains largely unexplored. Here we assign for the first time complete polyketide synthase (PKS) gene clusters to Bacillus antibiotics. Three giant modular PKS systems of the trans-acyltransferase type were identified in Bacillus amyloliquefaciens FZB 42. One of them, pks1, is an ortholog of the pksX operon with a previously unknown function in the sequenced model strain Bacillus subtilis 168, while the pks2 and pks3 clusters are novel gene clusters. Cassette mutagenesis combined with advanced mass spectrometric techniques such as matrix-assisted laser desorption ionization-time of flight mass spectrometry and liquid chromatography-electrospray ionization mass spectrometry revealed that the pks1 (bae) and pks3 (dif) gene clusters encode the biosynthesis of the polyene antibiotics bacillaene and difficidin or oxydifficidin, respectively. In addition, B. subtilis OKB105 (pheA sfp0), a transformant of the B. subtilis 168 derivative JH642, was shown to produce bacillaene, demonstrating that the pksX gene cluster directs the synthesis of that polyketide.

Abstract: We reformulate a class of non-linear stochastic optimal control problems introduced by Todorov (in Advances in Neural Information Processing Systems, vol. 19, pp. 1369---1376, 2007) as a Kullback-Leibler (KL) minimization problem. As a result, the optimal control computation reduces to an inference computation and approximate inference methods can be applied to efficiently compute approximate optimal controls. We show how this KL control theory contains the path integral control method as a special case. We provide an example of a block stacking task and a multi-agent cooperative game where we demonstrate how approximate inference can be successfully applied to instances that are too complex for exact computation. We discuss the relation of the KL control approach to other inference approaches to control.

Abstract: Natural hazard prediction and efficient crust exploration require dense seismic observations both in time and space. Seismological techniques provide ground-motion data, whose accuracy depends on sensor characteristics and spatial distribution. Here we demonstrate that dynamic strain determination is possible with conventional fibre-optic cables deployed for telecommunication. Extending recently distributed acoustic sensing (DAS) studies, we present high resolution spatially un-aliased broadband strain data. We recorded seismic signals from natural and man-made sources with 4-m spacing along a 15-km-long fibre-optic cable layout on Reykjanes Peninsula, SW-Iceland. We identify with unprecedented resolution structural features such as normal faults and volcanic dykes in the Reykjanes Oblique Rift, allowing us to infer new dynamic fault processes. Conventional seismometer recordings, acquired simultaneously, validate the spectral amplitude DAS response between 0.1 and 100 Hz bandwidth. We suggest that the networks of fibre-optic telecommunication lines worldwide could be used as seismometers opening a new window for Earth hazard assessment and exploration.

Abstract: There are surprisingly many people out there that obviously think that carbon footprinting is a new thing. They obviously are not aware of the fact that it has been around for decades—just being called differently, i.e. the result of the life cycle impact category indicator global warming potential (GWP). However, carbon footprinting (CFP) is really fashionable these days. Like with all fashion, not all that glitters is gold. Taking carbon footprinting as the one and only yardstick, one has to face life-threatening trade-offs. If carbon footprint is the way to go, we need to shut down each waste-water treatment plant in the world, because it leads to an increased carbon footprint. You should also tear out the catalytic converter and diesel particulate filters from cars, because they lead to a higher CFP. Nuclear power would be obviously a most preferable energy generation option, because it has a lower carbon footprint than even most renewable energy sources—at least based on information provided by pertinent EPDs (Vattenfall 2005; 2007a, b). Recycling paper should be stopped, because compared to virgin paper with a carbon footprint close to ‘zero’, it comes with a higher burden—unless renewable energy is used for the processes necessary (Carbon Trust 2006). But, on the other hand, we have the market demand. Whether it is real or just perceived or just desired seems not so important. There is enough momentum for numerous international, national and sectoral initiatives underway to deal with CFP:
