Abstract: Chemists of all fields currently publish about 50 000 crystal structures per year, the vast majority of which are X-ray structures. We determined two molecular structures by employing electron rather than X-ray diffraction. For this purpose, an EIGER hybrid pixel detector was fitted to a transmission electron microscope, yielding an electron diffractometer. The structure of a new methylene blue derivative was determined at 0.9 A resolution from a crystal smaller than 1×2 μm2 . Several thousand active pharmaceutical ingredients (APIs) are only available as submicrocrystalline powders. To illustrate the potential of electron crystallography for the pharmaceutical industry, we also determined the structure of an API from its pill. We demonstrate that electron crystallography complements X-ray crystallography and is the technique of choice for all unsolved cases in which submicrometer-sized crystals were the limiting factor.

Abstract: The e{sup +}e{sup -}{yields}pp cross section is determined over a range of pp masses, from threshold to 4.5 GeV/c{sup 2}, by studying the e{sup +}e{sup -}{yields}pp{gamma} process. The data set corresponds to an integrated luminosity of 232 fb{sup -1}, collected with the BABAR detector at the PEP-II storage ring, at an e{sup +}e{sup -} center-of-mass energy of 10.6 GeV. The mass dependence of the ratio of electric and magnetic form factors, G{sub E}/G{sub M}, is measured for pp masses below 3 GeV/c{sup 2}; its value is found to be significantly larger than 1 for masses up to 2.2 GeV/c{sup 2}. We also measure J/{psi}{yields}pp and {psi}(2S){yields}pp branching fractions and set an upper limit on Y(4260){yields}pp production and decay.

Abstract: Since the advent of the generation of human induced pluripotent stem cells (hiPSCs), numerous protocols have been developed to differentiate hiPSCs into cardiomyocytes and then subsequently assess their ability to recapitulate the properties of adult human cardiomyocytes. However, hiPSC-derived cardiomyocytes (hiPSC-CMs) are often assessed in single-cell assays. A shortcoming of these assays is the limited ability to characterize the physiological parameters of cardiomyocytes, such as contractile force, due to random orientations. This protocol describes the differentiation of cardiomyocytes from hiPSCs, which occurs within 14 d. After casting, cardiomyocytes undergo 3D assembly. This produces fibrin-based engineered heart tissues (EHTs)-in a strip format-that generate force under auxotonic stretch conditions. 10-15 d after casting, the EHTs can be used for contractility measurements. This protocol describes parallel expansion of hiPSCs; standardized generation of defined embryoid bodies, growth factor and small-molecule-based cardiac differentiation; and standardized generation of EHTs. To carry out the protocol, experience in advanced cell culture techniques is required.

Abstract: Quantum adiabatic passages can be greatly accelerated by a suitable control field, called a counter-diabatic field, which varies during the scan through resonance. Here, we implement this technique on the electron spin of a single nitrogen-vacancy center in diamond. We demonstrate two versions of this scheme. The first follows closely the procedure originally proposed by Demirplak and Rice [J. Phys. Chem. A 107, 9937 (2003)]. In the second scheme, we use a control field whose amplitude is constant but whose phase varies with time. This version, which we call the rapid-scan approach, allows an even faster passage through resonance and therefore makes it applicable also for systems with shorter decoherence times.

Abstract: We present measurements and calculations of optical Rabi oscillations in the excitonic ground-state transition of an InGaAs quantum dot ensemble at low temperature. Rabi oscillations which are damped versus pulse area and change period when changing pulse duration are observed. Comparisons with calculations show that the observed damping is not intrinsic to a single dot. Dephasing processes and the biexciton resonance change the amplitude and the period of the oscillations, respectively, while the damping versus pulse area is due to a distribution of transition dipole moments in the ensemble.
