Abstract: Plant exposure to abiotic stresses leads to an accumulation of reactive oxygen species with the concomitant increase in antioxidant defense mechanisms. Previous studies showed that exogenous application of proline mitigate the deleterious effects caused by oxidative stress due to its ability to increase the activity of antioxidant enzymes. However, there are no reports of the effects of high endogenous accumulation of proline in the transcriptional pattern of antioxidant enzymes genes under normal conditions of water supply or in response to water deficit. Here, we show that isoforms of four antioxidant enzymes genes (Ascorbate peroxidase-APX, Catalase-CAT, Superoxide dismutase-SOD and Glutathione reductase-GR) were differentially regulated in leaves of Swingle citrumelo transgenic plants with high endogenous proline accumulation submitted to water deficits and also under normal water supply condition. Proline per se caused a two-fold change in the transcription activity of APX1, APXcl, CAT2 and Cu/ZnSOD2, while during water deficit proline influenced mRNAs levels in APXs and Cu/ZnSODs isoforms, MnSODmit and GRcl. This study adds new information on the role of proline during drought conditions and, more important, without the potential confounding effects imposed by water deficiency. We showed that, in addition to its known effects on diverse plant physiological and biochemical processes, high endogenous proline can also acts as a regulatory/signalling molecule capable of altering the transcript levels of stress-related genes.

Abstract: Aspergillus fumigatus is a primary and opportunistic pathogen, as well as a major allergen, of mammals. The Ca+2-calcineurin pathway affects virulence, morphogenesis and antifungal drug action in A. fumigatus. Here, we investigated three components of the A. fumigatus Ca+2-calcineurin pathway, pmcA,-B, and -C, which encode calcium transporters. We demonstrated that CrzA can directly control the mRNA accumulation of the pmcA-C genes by binding to their promoter regions. CrzA-binding experiments suggested that the 5′-CACAGCCAC-3′ and 5′-CCCTGCCCC-3′ sequences upstream of pmcA and pmcC genes, respectively, are possible calcineurin-dependent response elements (CDREs)-like consensus motifs. Null mutants were constructed for pmcA and -B and a conditional mutant for pmcC demonstrating pmcC is an essential gene. The ΔpmcA and ΔpmcB mutants were more sensitive to calcium and resistant to manganese and cyclosporin was able to modulate the sensitivity or resistance of these mutants to these salts, supporting the interaction between calcineurin and the function of these transporters. The pmcA-C genes have decreased mRNA abundance into the alveoli in the ΔcalA and ΔcrzA mutant strains. However, only the A. fumigatus ΔpmcA was avirulent in the murine model of invasive pulmonary aspergillosis.

Abstract: The potential use of corn bran as a component for manufacturing snacks through thermoplastic extrusion was investigated. Corn bran (180, 250 and 320 g/kg) was blended with corn meal according to incomplete three level factorial design. Moisture (160, 190 and 220 g/kg), extrusion temperature (150, 170 and 190 °C) and glycerol monostearate content (0, 4 and 8 g/kg) were also studied. Dependent variables assessed were radial expansion ratio, longitudinal expansion, specific volume, hardness and fracturability. The snacks were also evaluated by an acceptance test of individual characteristics using a nontrained panel. The attributes evaluated were appearance, palatability and general acceptability. Analysis of variance showed that the best combination of general acceptability, expansion and higher level of fibre was reached on snacks prepared with 250 g/kg corn bran, higher temperature, lower moisture content and medium level of glycerol monostearate. The corn bran content significantly affected the radial expansion ratio, appearance and general acceptability of the product. All of them showed decreased values with increasing corn bran content.
