Abstract: Protected areas are the single most important conservation tool. The global protected-area network has grown substantially in recent decades, now occupying 11.5% of Earth's land surface, but such growth has not been strategically aimed at maximizing the coverage of global biodiversity. In a previous study, we demonstrated that the global network is far from complete, even for the representation of terrestrial vertebrate species. Here we present a first attempt to provide a global framework for the next step of strategically expanding the network to cover mammals, amphibians, freshwater turtles and tortoises, and globally threatened birds. We identify unprotected areas of the world that have remarkably high conservation value (irreplaceability) and are under serious threat. These areas concentrate overwhelmingly in tropical and subtropical moist forests, particularly on tropical mountains and islands. The expansion of the global protected-area network in these regions is urgently needed to prevent the loss of unique biodiversity.

Abstract: Climate change alone is unlikely to drive severe tropical forest degradation in the next few decades, but an alternative process associated with severe weather and forest fires is already operating in southeastern Amazonia. Recent droughts caused greatly elevated fire-induced tree mortality in a fire experiment and widespread regional forest fires that burned 5–12% of southeastern Amazon forests. These results suggest that feedbacks between fires and extreme climatic conditions could increase the likelihood of an Amazon forest “dieback” in the near-term. To secure the integrity of seasonally dry Amazon forests, efforts to end deforestation must be accompanied by initiatives that reduce the accidental spread of land management fires into neighboring forest reserves and effectively suppress forest fires when they start.

Abstract: Glycosylphosphatidylinositol (GPI) anchors and glycoinositolphospholipids (GIPLs) from parasitic protozoa have been shown to exert a wide variety of effects on cells of the host innate immune system. However, the receptor(s) that are triggered by these protozoan glycolipids has not been identified. Here we present evidence that Trypanosoma cruzi-derived GPI anchors and GIPLs trigger CD25 expression on Chinese hamster ovary-K1 cells transfected with CD14 and Toll-like receptor-2 (TLR-2), but not wild-type (TLR-2-deficient) Chinese hamster ovary cells. The protozoan-derived GPI anchors and GIPLs containing alkylacylglycerol and saturated fatty acid chains or ceramide were found to be active in a concentration range of 100 nM to 1 microM. More importantly, the GPI anchors purified from T. cruzi trypomastigotes, which contain a longer glycan core and unsaturated fatty acids in the sn-2 position of the alkylacylglycerolipid component, triggered TLR-2 at subnanomolar concentrations. We performed experiments with macrophages from TLR-2 knockout and TLR-4 knockout mice, and found that TLR-2 expression appears to be essential for induction of IL-12, TNF-alpha, and NO by GPI anchors derived from T. cruzi trypomastigotes. Thus, highly purified GPI anchors from T. cruzi parasites are potent activators of TLR-2 from both mouse and human origin. The activation of TLR-2 may initiate host innate defense mechanisms and inflammatory response during protozoan infection, and may provide new strategies for immune intervention during protozoan infections.

Abstract: In this article we review Raman studies of defects and dopants in graphene as well as the importance of both for device applications. First a brief overview of Raman spectroscopy of graphene is presented. In the following section we discuss the Raman characterization of three defect types: point defects, edges, and grain boundaries. The next section reviews the dependence of the Raman spectrum on dopants and highlights several common doping techniques. In the final section, several device applications are discussed which exploit doping and defects in graphene. Generally defects degrade the figures of merit for devices, such as carrier mobility and conductivity, whereas doping provides a means to tune the carrier concentration in graphene thereby enabling the engineering of novel material systems. Accurately measuring both the defect density and doping is critical and Raman spectroscopy provides a powerful tool to accomplish this task.
