Abstract: Modelling studies have predicted that coffee crop will be endangered by future global warming, but recent reports highlighted that high [CO2] can mitigate heat impacts on coffee. This work aimed at identifying heat protective mechanisms promoted by CO2 in Coffea arabica (cv. Icatu and IPR108) and C. canephora cv. Conilon CL153. Plants were grown at 25/20 oC (day/night), under 380 or 700 μL CO2 L-1, and then gradually submitted to 31/25, 37/30 and 42/34 oC. Relevant heat tolerance up to 37/30 oC for both [CO2] and all coffee genotypes was observed, likely supported by the maintenance or increase of the pools of several protective molecules (neoxanthin, lutein, carotenes, α-tocopherol, HSP70, raffinose), activities of antioxidant enzymes, such as superoxide dismutase (SOD), ascorbate peroxidase (APX), glutathione reductase (GR), catalase (CAT), and the upregulated expression of some genes (ELIP, Chaperonin 20). However, at 42/34 oC a tolerance threshold was reached, mostly in the 380-plants and Icatu. Adjustments in raffinose, lutein, β-carotene, α-tocopherol and HSP70 pools, and the upregulated expression of genes related to protective (ELIPS, HSP70, Chape 20 and 60) and antioxidant (CAT, CuSOD2, APX Cyt, APX Chl) proteins were largely driven by temperature. However, enhanced [CO2] maintained higher activities of GR (Icatu) and CAT (Icatu and IPR108), kept (or even increased) the Cu,Zn-SOD, APX and CAT activities, and promoted a greater upregulation of those enzyme genes, as well as those related to HSP70, ELIPs, Chaperonins in CL153 and Icatu.. These changes likely favoured the maintenance of reactive oxygen species at controlled levels and contributed to mitigate of photosystem II photoinhibition at the highest temperature. Overall, our results highlighted the important role of enhanced [CO2] on the coffee crop acclimation and sustainability under predicted future global warming scenarios.

Abstract: The severity of plant diseases, traditionally the proportion of the plant tissue exhibiting symptoms, is a key quantitative variable to know for many diseases and is prone to error. Good quality disease severity data should be accurate (close to the true value). Earliest quantification of disease severity was by visual estimates. Sensor-based image analysis including visible spectrum and hyperspectral and multispectral sensors are established technologies that promise to substitute, or complement visual ratings. Indeed, these technologies have measured disease severity accurately under controlled conditions but are yet to demonstrate their full potential for accurate measurement under field conditions. Sensor technology is advancing rapidly, and artificial intelligence may help overcome issues for automating severity measurement under hyper-variable field conditions. The adoption of appropriate scales, training, instruction and aids (standard area diagrams) has contributed to improved accuracy of visual estimates. The apogee of accuracy for visual estimation is likely being approached, and any remaining increases in accuracy are likely to be small. Due to automation and rapidity, sensor-based measurement offers potential advantages compared with visual estimates, but the latter will remain important for years to come. Mobile, automated sensor-based systems will become increasingly common in controlled conditions and, eventually, in the field for measuring plant disease severity for the purpose of research and decision making.

Abstract: An improved method for the Agrobacterium infiltration of epicotyl segments of ‘Pineapple’ sweet orange [Citrus sinensis (L.) Osbeck] and ‘Swingle’ citrumelo [Citrus paradisi Macf. X Poncirus trifoliata (L.) Raf.] was developed in order to increase transformation frequency. Sonication-assisted Agrobacterium-mediated transformation (SAAT), vacuum infiltration, and a combination of the two procedures were compared with conventional Agrobacterium-mediated inoculation method (‘dipping’ method). It was observed that the morphogenic potential of the epicotyl segments decreased as the duration of SAAT and vacuum treatments increased. Transient GUS expression was not affected by the different SAAT treatments, but it was significantly enhanced by the vacuum infiltration treatments. The highest transformation efficiencies were obtained when the explants were subjected to a combination of SAAT for 2 s followed by 10 min of vacuum infiltration. PCR and Southern blot analysis of the uidA gene were used to confirm the integration of the transgenes. The transformation frequencies achieved in this study (8.4% for ‘Pineapple’ sweet orange and 11.2% for ‘Swingle’ citrumelo) are the highest ones reported for both cultivars.

Abstract: BACKGROUND: Peanut contamination by fungi is a concern of processors and consumers owing to the association of these micro-organisms with quality deterioration and aflatoxin production. In this study the fungicidal and detoxifying effects of ozone on aflatoxins in peanuts was investigated. Peanut kernels were ozonated at concentrations of 13 and 21 mg L −1 for periods of 0, 24, 48, 72 and 96 h. RESULTS: Ozone was effective in controlling total fungi and potentially aflatoxigenic species in peanuts, with a reduction in colony-forming units per gram greater than 3 log cycles at the concentration of 21 mg L −1 after 96 h of exposure. A reduction in the percentage of peanuts with internal fungal populations was also observed, particularly after exposure to ozone at 21 mg L −1 . A reduction inthe concentrationsof total aflatoxinsand aflatoxinB1 of approximately30 and25% respectively was observed for kernels exposed to ozone at 21 mg L −1 for 96 h. CONCLUSION: It was concluded that ozone is an important alternative for peanut detoxification because it is effective in controlling potentially aflatoxigenic fungi and also acts in the reduction of aflatoxin levels in kernels. c � 2011 Society of Chemical Industry
