Abstract: The aim of this review was to survey all fungal pathologists with an association with the journal Molecular Plant Pathology and ask them to nominate which fungal pathogens they would place in a 'Top 10' based on scientific/economic importance. The survey generated 495 votes from the international community, and resulted in the generation of a Top 10 fungal plant pathogen list for Molecular Plant Pathology. The Top 10 list includes, in rank order, (1) Magnaporthe oryzae; (2) Botrytis cinerea; (3) Puccinia spp.; (4) Fusarium graminearum; (5) Fusarium oxysporum; (6) Blumeria graminis; (7) Mycosphaerella graminicola; (8) Colletotrichum spp.; (9) Ustilago maydis; (10) Melampsora lini, with honourable mentions for fungi just missing out on the Top 10, including Phakopsora pachyrhizi and Rhizoctonia solani. This article presents a short resume of each fungus in the Top 10 list and its importance, with the intent of initiating discussion and debate amongst the plant mycology community, as well as laying down a bench-mark. It will be interesting to see in future years how perceptions change and what fungi will comprise any future Top 10.

Abstract: Procreation is essential for survival of species. Not surprisingly, complex neuronal networks have evolved to mediate the diverse internal and external environmental inputs that regulate reproduction in vertebrates. Ultimately, these regulatory factors impinge, directly or indirectly, on a final common pathway, the neurons producing the gonadotropin-releasing hormone (GnRH), which stimulates pituitary gonadotropin secretion and thereby gonadal function. Compelling evidence, accumulated in the last few years, has revealed that kisspeptins, a family of neuropeptides encoded by the Kiss1 gene and produced mainly by neuronal clusters at discrete hypothalamic nuclei, are pivotal upstream regulators of GnRH neurons. As such, kisspeptins have emerged as important gatekeepers of key aspects of reproductive maturation and function, from sexual differentiation of the brain and puberty onset to adult regulation of gonadotropin secretion and the metabolic control of fertility. This review aims to provide a comprehensive account of the state-of-the-art in the field of kisspeptin physiology by covering in-depth the consensus knowledge on the major molecular features, biological effects, and mechanisms of action of kisspeptins in mammals and, to a lesser extent, in nonmammalian vertebrates. This review will also address unsolved and contentious issues to set the scene for future research challenges in the area. By doing so, we aim to endow the reader with a critical and updated view of the physiological roles and potential translational relevance of kisspeptins in the integral control of reproductive function.

Abstract: Introduction: Cardiovascular disease remains the commonest health problem in developed countries, and residual risk after implementing all current therapies is still high. The use of marine omega-3 fatty acids (DHA and EPA) has been recommended to reduce cardiovascular risk by multiple mechanisms. Objectives: To update the current evidence on the influence of omega-3 on the rate of cardiovascular events. Review Methods: We used the MEDLINE and EMBASE databases to identify clinical trials and randomized controlled trials of omega-3 fatty acids (with quantified quantities) either in capsules or in dietary intake, compared to placebo or usual diet, equal to or longer than 6 months, and written in English. The primary outcome was a cardiovascular event of any kind and secondary outcomes were all-cause mortality, cardiac death and coronary events. We used RevMan 5·1 (Mantel-Haenszel method). Heterogeneity was assessed by the I 2 and Chi 2 tests. We included 21 of the 452 pre-selected studies. Results: We found an overall decrease of risk of suffering a cardiovascular event of any kind of 10 % (OR 0·90; [0·85–0·96], p = 0·001), a 9 % decrease of risk of cardiac death (OR 0·91; [0·83–0·99]; p = 0·03), a decrease of coronary events (fatal and non-fatal) of 18 % (OR 0·82; [0·75–0·90]; p − 4 ), and a trend to lower total mortality (5 % reduction of risk; OR 0·95; [0·89–1·02]; p = 0·15. Most of the studies analyzed included persons with high cardiovascular risk. Conclusions: marine omega-3 fatty acids are effective in preventing cardiovascular events, cardiac death and coronary events, especially in persons with high cardiovascular risk.

Abstract: Sirtuins 1-7 (SIRT1-7) belong to the third class of deacetylase enzymes, which are dependent on NAD(+) for activity. Sirtuins activity is linked to gene repression, metabolic control, apoptosis and cell survival, DNA repair, development, inflammation, neuroprotection, and healthy aging. Because sirtuins modulation could have beneficial effects on human diseases there is a growing interest in the discovery of small molecules modifying their activities. We review here those compounds known to activate or inhibit sirtuins, discussing the data that support the use of sirtuin-based therapies. Almost all sirtuin activators have been described only for SIRT1. Resveratrol is a natural compound which activates SIRT1, and may help in the treatment or prevention of obesity, and in preventing tumorigenesis and the aging-related decline in heart function and neuronal loss. Due to its poor bioavailability, reformulated versions of resveratrol with improved bioavailability have been developed (resVida, Longevinex(®) , SRT501). Molecules that are structurally unrelated to resveratrol (SRT1720, SRT2104, SRT2379, among others) have been also developed to stimulate sirtuin activities more potently than resveratrol. Sirtuin inhibitors with a wide range of core structures have been identified for SIRT1, SIRT2, SIRT3 and SIRT5 (splitomicin, sirtinol, AGK2, cambinol, suramin, tenovin, salermide, among others). SIRT1 inhibition has been proposed in the treatment of cancer, immunodeficiency virus infections, Fragile X mental retardation syndrome and for preventing or treating parasitic diseases, whereas SIRT2 inhibitors might be useful for the treatment of cancer and neurodegenerative diseases.

Abstract: :::::::::::::::: :::::::::::::::: ::::::::: Past research has demonstrated the effects of bullying can be severe and long term for the individuals involved. The main aim of this study is to analyze the emotional impact on victims of traditional bullying, both direct and indirect forms, and of cyberbullying through mobile phones and the Internet. A sample of 5,862 adolescents from three different countries, Italy (N = 1,964), Spain (N = 1,671), and England (N = 2,227), responded to a questionnaire that asked if they had experience of various forms of bullying, and the consequent emotional impact. The results show that both traditional bullying and cyberbullying have a significant prevalence in the samples. Emotional responses are linked to types of bullying. Analysis of answers identified specific emotional profiles for the different types of bullying and cyberbullying. Direct bullying and cyberbullying via mobile phone showed similar profiles, and also indirect bullying and cyberbullying using the Internet. Similarities and differences between profiles are discussed and some hypotheses are presented to explain the results. Abstract: A progressive global increase in the burden of allergic diseases has affected the industrialized world over the last half century and has been reported in the literature. The clinical evidence reveals a general increase in both incidence and prevalence of respiratory diseases, such as allergic rhinitis (common hay fever) and asthma. Such phenomena may be related not only to air pollution and changes in lifestyle, but also to an actual increase in airborne quantities of allergenic pollen. Experimental enhancements of carbon dioxide (CO2) have demonstrated changes in pollen amount and allergenicity, but this has rarely been shown in the wider environment. The present analysis of a continental-scale pollen data set reveals an increasing trend in the yearly amount of airborne pollen for many taxa in Europe, which is more pronounced in urban than semi-rural/rural areas. Climate change may contribute to these changes, however increased temperatures do not appear to be a major influencing factor. Instead, we suggest the anthropogenic rise of atmospheric CO2 levels may be influential.

Abstract: We present a technique for estimating the spatial layout of humans in still images--the position of the head, torso and arms. The theme we explore is that once a person is localized using an upper body detector, the search for their body parts can be considerably simplified using weak constraints on position and appearance arising from that detection. Our approach is capable of estimating upper body pose in highly challenging uncontrolled images, without prior knowledge of background, clothing, lighting, or the location and scale of the person in the image. People are only required to be upright and seen from the front or the back (not side). We evaluate the stages of our approach experimentally using ground truth layout annotation on a variety of challenging material, such as images from the PASCAL VOC 2008 challenge and video frames from TV shows and feature films. We also propose and evaluate techniques for searching a video dataset for people in a specific pose. To this end, we develop three new pose descriptors and compare their classification and retrieval performance to two baselines built on state-of-the-art object detection models.

Abstract: In the current climate of several interrelated impending global crises, namely, climate change, chemicals, energy, and oil, the impact of green chemistry with respect to chemicals and biofuels generated from within a holistic concept of a biorefinery is discussed. Green chemistry provides unique opportunities for innovation via product substitution, new feedstock generation, catalysis in aqueous media, utilization of microwaves, and scope for alternative or natural solvents. The potential of utilizing waste as a new resource and the development of integrated facilities producing multiple products from biomass is discussed under the guise of biorefineries. Biofuels are discussed in depth, as they not only provide fuel (energy) but are also a source of feedstock chemicals. In the future, the commercial success of biofuels commensurate with consumer demand will depend on the availability of new green (bio)chemical technologies capable of converting waste biomass to fuel in a context of a biorefinery.

Abstract: Forest restoration projects have become increasingly common around the world and planting trees is almost always a key component. Low seedling survival and growth may result in restoration failures and various mechanical site preparation techniques for treatment of soils and vegetation are important tools used to help counteract this. In this article, we synthesize the current state-of-knowledge concerning mechanical site preparation for improved tree establishment when carried out in different forest restoration situations, point out critical research gaps and provide some recommendations for future directions. Mechanical site preparation often results in improved seedling survival and growth. However, if not intensive methods with much soil disturbance are used, it is a rather ineffective tool for controlling competing vegetation. Methods such as scarification, mounding and subsoiling also lead to multiple interactions among soil physical and chemical properties that affect plant survival and growth, and it may be difficult to determine the actual cause–effect relationship of any positive seedling responses. Most research to date on mechanical site preparation and plantation performance has been conducted using a few conifer tree species. Seedling responses differ among tree species and alternative species are often used during restoration compared to production forestry indicating a need for additional research for improved understanding. Several management objectives such as soil protection and increased biodiversity are many times relevant during forest restoration, and mechanical site preparation methods should be implemented carefully because they can have large impacts on the environment.

Abstract: Unbleached (UN), oxygen-delignified and fully-bleached (FB) birch fibers with a residual lignin content of ca. 3, 2 and <1 %, respectively, were used to produce nanofibrillated cellulose (NFC) and nanopaper by using an overpressure device. The tensile index, elongation and elastic modulus of nanopaper were compared and the effect of residual cell wall components accessed. Under similar manufacturing conditions, UN NFC produced nanopaper with a density of 0.99 g/cm3, higher than that from FB NFC (0.7 g/cm3). This translated in much lower air permeability in the case of UN nanopaper (1 and 11 mL/min for UN and FB samples, respectively). Fundamentally, these observations are ascribed to the finer fibrils produced during microfluidization of UN fibers compared to those from lower yield counterparts (AFM roughness of 8 and 17 nm and surface areas of 124 and 98 m2/g for NFC from UN and FB fibers, respectively). As a result, values of stress at break and energy absorption of nanopaper from high yield fibers are distinctively higher than those from fully bleached NFC. Interactions of water with the surface and bulk material were affected by the chemical composition and structure of the nanofibrils. While UN nanopaper presented higher water contact angles their sorption capacity (and rate of water absorption) was much higher than those measured for nanopaper from FB NFC. These and other observations provided in this contribution are proposed to be related to the mechanoradical scavenging capacity of lignin in high shear microfluidization and the presence of residual heteropolysaccharides.

Abstract: The aim of this paper is to explore the characteristics and motivations of culinary tourists whose destination is the city of Cordoba (Spain) while attempting to determine whether gastronomy is an important aspect of the trip or if it is perceived simply as a secondary activity. Fieldwork was carried out among a series of restaurant establishments in the city of Cordoba. A demand survey was conducted in 10 particular establishments that were selected among those offering local dishes and which are regularly visited by tourists and the questionnaire was distributed among clients obtaining 206 valid answers. Our results reveal the strengths and weaknesses of Cordoba's restaurant sector. While 10% state that the cuisine is one of the main reasons for visiting the city, 68% believe that the local cuisine is an important but not an essential aspect of their trip and the rest view it as being secondary. Abstract: Molecular markers (SSR, SNP and DArT) and agronomical traits have been used in the world’s largest olive (Olea europaea L.) germplasm collection (IFAPA, Centre Alameda del Obispo, Cordoba, Spain) to study the patterns of genetic diversity and underlying genetic structure among 361 olive accessions. In addition the marker data were used to construct a set of core collections by means of two different algorithms (MSTRAT and PowerCore) based on M (maximization) strategy. Our results confirm that the germplasm collection is a useful source of genetically diverse material. We also found that geographical origin is an important factor structuring genetic diversity in olive. Subsets of 18, 27, 36, 45 and 68 olive accessions, representing respectively 5%, 7.5%, 10%, 12.5% and 19% of the whole germplasm collection, were selected based on the information obtained by all the data set as well as each marker type considered individually. According to our results, the core collections that represent between 19% and 10% of the total collection size could be considered as optimal to retain the bulk of the genetic diversity found in this collection. Due to its high efficiency at capturing all the alleles/traits states found in the whole collection, the core size of 68 accessions could be of special interest for genetic conservation applications in olive. The high average genetic distance and diversity and the almost equal representation of accessions from different geographical regions indicate that the core size of 36 accessions, could be the working collection for olive breeders.

Abstract: This study tested the hypothesis that the expression of CD112 and CD155 (DNAM-1 ligands) on leukemic blasts induces a decreased expression of the activating receptor DNAM-1 on natural killer (NK) cells from acute myeloid leukemia (AML) patients. DNAM-1 is a co-receptor involved in the activation of NK cell cytotoxicity after its interaction with its ligands CD112 and CD155 on target cells. Here we study the expression of DNAM-1 on NK cells and DNAM-1 ligands on blasts from AML patients stratified by age. The results demonstrate that NK cells from AML patients younger than 65 years have a reduced expression of DNAM-1 compared with age-matched controls. The analysis of DNAM-1 ligands showed a high expression of CD112 and CD155 on leukemic blasts. An inverse correlation between CD112 expression on leukemic blasts and DNAM-1 expression on NK cells was found. Furthermore, downregulation of DNAM-1 was induced on healthy donors' NK cells after in vitro culture with leukemic blasts expressing DNAM-1 ligands. In conclusion, these results support the hypothesis that receptor-ligand crosslinking downregulates DNAM-1 expression on NK cells from patients <65 years of age. Considering the relevance of DNAM-1 in NK recognition and killing of leukemic cells, the reduced expression of this receptor on NK cells from AML patients can represent an additional mechanism of tumor escape.

Abstract: Kisspeptins (Kp), products of the Kiss1 gene that act via Gpr54 to potently stimulate GnRH secretion, operate as mediators of other regulatory signals of the gonadotropic axis. Mouse models of Gpr54 and/or Kiss1 inactivation have been used to address the contribution of Kp in the central control of gonadotropin secretion; yet, phenotypic and hormonal differences have been detected among the transgenic lines available. We report here a series of neuroendocrine analyses in male mice of a novel Gpr54 knockout (KO) model, generated by heterozygous crossing of a loxP-Gpr54/Protamine-Cre double mutant line. Gpr54-null males showed severe hypogonadotropic hypogonadism but retained robust responsiveness to GnRH. Gonadotropic responses to the agonist of ionotropic glutamate receptors, N-methyl-d-aspartate, were attenuated, but persisted, in Gpr54-null mice. In contrast, LH secretion after activation of metabotropic glutamate receptors was totally preserved in the absence of Gpr54 signaling. Detectable, albeit reduced, LH responses were also observed in Gpr54 KO mice after intracerebroventricular administration of galanin-like peptide or RF9, putative antagonist of neuropeptide FF receptors for the mammalian ortholog of gonadotropin-inhibiting hormone. In contrast, the stimulatory effect of senktide, agonist of neurokinin B (NKB; cotransmitter of Kiss1 neurons), was totally abrogated in Gpr54 KO males. Lack of Kp signaling also eliminated feedback LH responses to testosterone withdrawal. However, residual but sustained increases of FSH were detected in gonadectomized Gpr54 KO males, in which testosterone replacement failed to fully suppress circulating FSH levels. In sum, our study provides novel evidence for the relative importance of Kp-dependent vs. -independent actions of several key regulators of GnRH secretion, such as glutamate, galanin-like peptide, and testosterone. In addition, our data document for the first time the indispensable role of Kp signaling in mediating the stimulatory effects of NKB on LH secretion, thus supporting the hypothesis that NKB actions on GnRH neurons are indirectly mediated via its ability to regulate Kiss1 neuronal output.

Abstract: In seasonal mammals, a distinct photoneuroendocrine circuit that involves the pineal hormone melatonin tightly synchronizes reproduction with seasons. In the Syrian hamster, a seasonal model in which sexual activity is inhibited by short days, we have previously shown that the potent GnRH stimulator, kisspeptin, is crucial to convey melatonin's message; however, the precise mechanisms through which melatonin affects kisspeptin remain unclear. Interestingly, rfrp gene expression in the neurons of the dorsomedial hypothalamic nucleus, a brain region in which melatonin receptors are present in the Syrian hamster, is strongly down-regulated by melatonin in short days. Because a large body of evidence now indicates that RFamide-related peptide (RFRP)-3, the product of the rfrp gene, is an inhibitor of gonadotropin secretion in various mammalian species, we sought to investigate its effect on the gonadotrophic axis in the Syrian hamster. We show that acute central injection of RFRP-3 induces c-Fos expression in GnRH neurons and increases LH, FSH, and testosterone secretion. Moreover, chronic central administration of RFRP-3 restores testicular activity and Kiss1 levels in the arcuate nucleus of hamsters despite persisting photoinhibitory conditions. By contrast RFRP-3 does not have a hypophysiotrophic effect. Overall, these findings demonstrate that, in the male Syrian hamster, RFRP-3 exerts a stimulatory effect on the reproductive axis, most likely via hypothalamic targets. This places RFRP-3 in a decisive position between the melatonergic message and Kiss1 seasonal regulation. Additionally, our data suggest for the first time that the function of this peptide depends on the species and the physiological status of the animal model.

Abstract: The negative impacts of severe drought on the growth and vigor of tree species and their relationship with forest decline have not been properly evaluated taking into account the differential responses to such stress of trees, sites and species. We evaluated these responses by quantifying the changes in radial growth of plantations of four pine species (Pinus sylvestris, Pinus nigra, Pinus pinaster, Pinus halepensis) which showed distinct decline and defoliation levels in southeastern Spain. We used dendrochronological methods, defoliation records, linear mixed models of basal area increment and dynamic factor analysis to quantify the responses of trees at the species and individual scales to site conditions and drought stress. In the region a temperature rise and a decrease in spring precipitation have led to drier conditions during the late twentieth century characterized by severe droughts in the 1990s and 2000s. As expected, the defoliation levels and the reductions in basal area increment were higher in those species more vulnerable to drought-induced xylem embolism (P. sylvestris) than in those more resistant (P. halepensis). Species adapted to xeric conditions but with high growth rates, such as P. pinaster, were also vulnerable to drought-induced decline. The reduction in basal area increment and the defoliation events occurred after consecutive severe droughts. A decrease in spring precipitation, which is the main driver of radial growth, is the most plausible cause of recent forest decline. The sharp growth reduction and widespread defoliation of the most affected pine plantations of Scots pine make their future persistence in drought-prone sites unlikely under the forecasted warmer and drier conditions.

Abstract: Human genetic studies have revealed that neurokinin B (NKB) and its receptor, neurokinin-3 receptor (NK3R), are essential elements for normal reproduction; however, the precise role of NKB–NK3R signaling in the initiation of puberty remains unknown We investigated here the regulation of Tac2 and Tacr3 mRNAs (encoding NKB and NK3R, respectively) in female rats and demonstrated that their hypothalamic expression is increased along postnatal maturation At puberty, both genes were widely expressed throughout the brain, including the lateral hypothalamic area and the arcuate nucleus (ARC)/medial basal hypothalamus, where the expression of Tacr3 increased across pubertal transition We showed that central administration of senktide (NK3R agonist) induced luteinizing hormone (LH) secretion in prepubertal and peripubertal females Conversely, chronic infusion of an NK3R antagonist during puberty moderately delayed the timing of vaginal opening (VO) and tended to decrease LH levels The expression of NKB and its receptor was sensitive to changes in metabolic status during puberty, as reflected by a reduction in Tacr3 (and, to a lesser extent, Tac2) expression in the ARC after a 48 h fast Yet, acute LH responses to senktide in pubertal females were preserved, if not augmented, under fasting conditions, suggesting sensitization of the NKB–NK3R–gonadotropin-releasing hormone signaling pathway under metabolic distress Moreover, repeated administration of senktide to female rats with pubertal arrest due to chronic undernutrition rescued VO (in ∼50% of animals) and potently elicited LH release Altogether, our observations suggest that NKB–NK3R signaling plays a role in pubertal maturation and that its alterations may contribute to pubertal disorders linked to metabolic stress and negative energy balance

Abstract: Conservation agriculture (CA) helps to mitigate climate change. Firstly, the modifications introduced by CA on the carbon dynamics in the soil directly result in an increase of the carbon (C) in the soil fraction. Secondly, CA drastically reduces C oxidation processes by diminishing the mechanical manipulation of the soil. Spain's position in relation to the Kyoto Protocol must be improved, as is one of the European countries in a non-compliance situation. With the aim of providing knowledge about the potential of CA as C sink in Spain, 29 articles on this subject were reviewed. According to 2010 CA uptake, the results demonstrated that conservation practices have the potential to promote the fixation in soil of about 2 Gg year−1 more C than traditional tillage (TT) systems. As indicated by Tebrugge (2001) , 3.7 Mg of CO2 are generated from 1 Mg of C through microbial oxidation processes taking place in the ground, meaning that through CA almost 7.5 Gg of CO2 could be sequestered from the atmosphere every year until the equilibrium is reached. C fixation was found to be irregular over time. C fixation rates were high in newly implemented systems during the first 10 years, reaching top values of 0.85 Mg ha−1 year−1 for no-tillage (NT) and 1.54 Mg ha−1 year−1 for cover crops (CC) implemented in-between perennial tree rows. After those first 10 years, it followed a period of lower but steady growth until equilibrium was reached. Nevertheless, C decreases of 0.16 Mg ha−1 year−1 in the first 10 years may be expected when practicing minimum tillage (MT). C sequestration rate resulted higher in case farmers do crop rotations in NT and MT rather than monoculture. In woody crops, studies reported higher C fixation values for native species when compared to sowed CC. Also, climate conditions seem to affect C sequestration rate in Spain. Although in NT differences observed between maritime and continental climates are not pronounced, as approximately 25% of the values recorded in both climates are equal, in the case of MT about 75% of maritime climate values result higher than the continental situation.
