Abstract: Summary An analysis of 25 microsatellite loci in 210 animals has been used to define the genetic structure of the Iberian pig, traditionally classified into several varieties. In addition, a sample of 20 Duroc pigs was used as an outgroup for topology trees. Inter-variety genetic variation was estimated by unbiased average heterozygosity and the number of alleles observed. Significant deviations from the Hardy‐ Weinberg equilibrium (HWE) were shown for 19 loci across the whole population. By contrast, equilibrium deviation within varieties was much lower. Genetic variation measures, genetic distance values and a neighbour-joining tree were used to estimate subdivision. In addition, an individual tree was constructed to contrast the assignation of animals into varieties. Despite the low bootstrap values obtained in the varieties neighbour-joining tree, the degree of genetic variation found was sufficient to support the division of the Iberian pig into varieties, although in some cases the traditional classification cannot be accepted. These results have shown the value of this marker panel in the study of intra-breed genetic structures.

Abstract: Data on genus and infectious by Rickettsia were retrospectively compiled from the critical review literature regarding all countries in Latin America, Caribbean islands, Portugal and Spain. We considered all Rickettsia records reported for human and/or animal hosts, and/or invertebrate hosts considered being the vector. In a few cases, when no direct detection of a given Rickettsia group or species was available for a given country, the serologic method was considered. A total of 13 Rickettsia species have been recorded in Latin America and the Caribbean. The species with the largest number of country confirmed records were Rickettsia felis (9 countries), R. prowazekii (7 countries), R. typhi (6 countries), R. rickettsii (6 countries), R. amblyommii (5 countries), and R. parkeri (4 countries). The rickettsial records for the Caribbean islands (West Indies) were grouped in only one geographical area. Both R. bellii, R. akari, and Candidatus �R. andeane� have been recorded in only 2 countries each, whereas R. massiliae, R. rhipicephali, R.monteiroi, and R. africae have each been recorded in a single country (in this case, R. africae has been recorded in nine Caribbean Islands). For El Salvador, Honduras, and Nicaragua, no specific Rickettsia has been reported so far, but there have been serological evidence of human or/and animal infection. The following countries remain without any rickettsial records: Belize, Venezuela, Guyana, Surinam, and Paraguay. In addition, except for a few islands, many Caribbean islands remain without records. A total of 12 Rickettsia species have been reported in Spain and Portugal: R. conorii, R. helvetica, R. monacensis, R. felis, R. slovaca, R. raoultii, R. sibirica, R. aeschlimannii, R. rioja, R. massiliae, R. typhi, and R. prowazekii. Amongst these Rickettsia species reported in Spain and Portugal, only R. prowazekii, R. typhi, R. felis, and R. massiliae have also been reported in Latin America. This study summarizes the current state of art on the rickettsial distribution in Latin America, Caribbean, Spain and Portugal. The data obtained allow a better understanding on rickettsial epidemiology and distribution of vector ecology.

Abstract: Regulated deficit irrigation (RDI) strategies, often applied in tree crops, require precise monitoring methods of water stress. Crop water stress index (CWSI), based on canopy temperature measurements, has shown to be a good indicator of water deficits in field crops but has seldom been used in trees. CWSI was measured on a continuous basis in a Central California mature pistachio orchard, under full and deficit irrigation. Two treatments—control, returning the full evapotranspiration (ETc) and RDI—irrigated with 40% ETc during stage 2 of fruit grow (shell hardening). During stage 2, the canopy temperature—measured continuously with infrared thermometers (IRT)—of the RDI treatment was consistently higher than the control during the hours of active transpiration; the difference decreasing after irrigation. The non-water-stressed baseline (NWSB), obtained from clear-sky days canopy–air temperature differential and vapour pressure deficit (VPD) in the control treatment, showed a marked diurnal variation in the intercept, mainly explained by the variation in solar radiation. In contrast, the NWSB slope remained practically constant along the day. Diurnal evolution of calculated CWSI was stable and near zero in the control, but showed a clear rising diurnal trend in the RDI treatment, increasing as water stress increased around midday. The seasonal evolution of the CWSI detected large treatment differences throughout the RDI stress period. While the CWSI in the well-irrigated treatment rarely exceeded 0.2 throughout the season, RDI reached values of 0.8–0.9 near the end of the stress period. The CWSI responded to irrigation events along the whole season, and clearly detected mild water stress, suggesting extreme sensitivity to variations in tree water status. It correlated well with midday leaf water potential (LWP), but was more sensitive than LWP at mild stress levels. We conclude that the CWSI, obtained from continuous nadir-view measurements with IRTs, is a good and very sensitive indicator of water stress in pistachio. We recommend the use of canopy temperature measurements taken from 1200 to 1500 h, together with the following equation for the NWSB: (T c − T a) = −1.33·VPD + 2.44. Measurements of canopy temperature with VPD < 2 kPa are likely to generate significant errors in the CWSI calculation and should be avoided.

Abstract: Kisspeptins, the products of the KiSS-1 gene acting via G protein-coupled receptor 54 (GPR54), have recently emerged as pivotal signals in the hypothalamic network triggering the preovulatory surge of gonadotropins and, hence, ovulation. Additional actions of kisspeptins at other levels of the hypothalamic-pituitary-ovarian axis have been suggested but remain to date scarcely studied. We report herein the pattern of expression of KiSS-1 and GPR54 in the human and nonhuman primate ovary and evaluate changes in ovarian KiSS-1 expression in a rat model of ovulatory dysfunction. KiSS-1 and GPR54 mRNAs were detected in human ovarian tissue and cultured granulosa-lutein cells. In good agreement, kisspeptin immunoreactivity was observed in cyclic human and marmoset ovaries, with prominent signals in the theca layer of growing follicles, corpora lutea, interstitial gland, and ovarian surface epithelium. GPR54 immunoreactivity was also found in human theca and luteal cells. Administration of indomethacin to cyclic female rats disturbed ovulation and resulted in a dramatic drop in ovarian KiSS-1, but not GPR54, cyclooxygenase-2 (COX-2), or progesterone receptor, mRNA levels at the time of ovulation; an effect mimicked by the selective COX-2 inhibitor NS398 and rescued by coadministration of PGE(2). Likewise, the stimulatory effect of human choriogonadotropin on ovarian KiSS-1 expression was partially blunted by indomethacin. In contrast, KiSS-1 mRNA levels remained unaltered in another model of ovulatory failure, i.e., the RU486-treated rat. In summary, we document for the first time the expression of KiSS-1/kisspeptin and GPR54 in the human and nonhuman primate ovary. In addition, we provide evidence for the ability of inhibitors of COX-2, known to disturb follicular rupture and ovulation, to selectively alter the expression of KiSS-1 gene in rat ovary. Altogether, our results are suggestive of a conserved role of local KiSS-1 in the direct control of ovarian functions in mammals.
