Abstract: Some cases of Alzheimer's disease are inherited as an autosomal dominant trait. Genetic linkage studies have mapped a locus (AD3) associated with susceptibility to a very aggressive form of Alzheimer's disease to chromosome 14q24.3. We have defined a minimal cosegregating region containing the AD3 gene, and isolated at least 19 different transcripts encoded within this region. One of these transcripts (S182) corresponds to a novel gene whose product is predicted to contain multiple transmembrane domains and resembles an integral membrane protein. Five different missense mutations have been found that cosegregate with early-onset familial Alzheimer's disease. Because these changes occurred in conserved domains of this gene, and are not present in normal controls, they are likely to be causative of AD3.

Abstract: We report the cloning of a novel gene (E5-1) encoded on chromosome 1 which has substantial nucleotide and amino-acid sequence similarity to the S182 gene on chromosome 14q24.3. Mutations, including three new missense mutations in the S182 gene, are associated with the AD3 subtype of early-onset familial Alzheimer's disease (AD). Both the E5-1 and the S182 proteins are predicted to be integral membrane proteins with seven membrane-spanning domains, and a large exposed loop between the sixth and seventh transmembrane domains. Analysis of the nucleotide sequence of the open reading frame (ORF) of the E5-1 gene led to the discovery of two missense substitutions at conserved amino-acid residues in affected members of pedigrees with a form of familial AD that has a later age of onset than the AD3 subtype (50-70 years versus 30-60 years for AD3). These observations imply that the E5-1 gene on chromosome 1 and the S182 gene on chromosome 14q24.3 are members of a family of genes (presenilins) with related functions, and indicates that mutations in conserved residues of E5-1 could also play a role in the genesis of AD. Our results also indicate that still other AD susceptibility genes exist.

Abstract: In this paper, we present new conditions ensuring existence, uniqueness, and Global Asymptotic Stability (GAS) of the equilibrium point for a large class of neural networks. The results are applicable to both symmetric and nonsymmetric interconnection matrices and allow for the consideration of all continuous nondecreasing neuron activation functions. Such functions may be unbounded (but not necessarily surjective), may have infinite intervals with zero slope as in a piece-wise-linear model, or both. The conditions on GAS rely on the concept of Lyapunov Diagonally Stable (or Lyapunov Diagonally Semi-Stable) matrices and are proved by employing a class of Lyapunov functions of the generalized Lur'e-Postnikov type. Several classes of interconnection matrices of applicative interest are shown to satisfy our conditions for GAS. In particular, the results are applied to analyze GAS for the class of neural circuits introduced for solving linear and quadratic programming problems. In this application, the principal result here obtained is that these networks are GAS also when the constraint amplifiers are dynamical, as it happens in any practical implementation. >

Abstract: The effect of progesterone (P) on the cytokine production profile of Ag-specific human CD4+ T cell lines and clones was investigated. T cell lines specific for purified protein derivative or streptokinase (SK) derived in the presence of P exhibited significant increased ability to produce IL-5 in comparison with T cell lines derived in the absence of P. Moreover, IL-4 was significantly increased in SK-specific T cell lines derived in the presence of P in comparison with SK-specific T cell lines derived in the absence of this hormone. In addition, SK-specific T cell lines generated in the presence of P developed into T cell clones showing a Th0-, instead of Th1-like, cytokine profile. Furthermore, SK-specific T cell clones with an established Th1 profile of cytokine secretion did express mRNA for, and produced detectable amounts of, IL-4 when stimulated with P in combination with insoluble anti-CD3 mAb. Combined stimulation with P and insoluble anti-CD3 mAb also enabled Th1 clones to express CD30 on their surface membrane. These results indicate that P can favor the development of Th cells producing Th2-type cytokines and is an inducer of both transient IL-4 production and CD30 expression in established Th1 cells. Thus, P production at the placental level may be responsible, at least in part, for increased production of Th2-type cytokines which have been implied in fetal allograft survival and maintenance of successful pregnancy.

Abstract: Evidence is accumulating to suggest the existence of polarized human T-cell responses, reminiscent of TH1 and TH2 subsets described for mouse T cells. Human TH1 cells preferentially develop during infections by intracellular bacteria and trigger phagocyte-mediated host defense, whereas TH2 cells, which predominate during helminthic infestations and in response to common environmental allergens, are responsible for phagocyte-independent host response. Human TH1 and TH2 cells exhibit not only different functional properties but probably also distinct surface markers; TH2, but not TH1, clones express membrane CD30 and release the soluble form of CD30, a member of the TNF receptor superfamily. The cytokine profile of "natural immunity" evoked by different offending agents in the context of different host genetic backgrounds appears to be the most critical factor in determining the phenotype of the subsequent specific response. IL-12 and IFN-alpha and gamma produced by macrophages and NK cells favor the development of TH1 cells, whereas the early production of IL-4 by a still-unidentified cell type favors the development of TH2 cells. Clearly, polarized human TH1 and TH2 responses not only play different roles in protection, they can also promote different immunopathological reactions. Strong and persistent TH1 responses seen to be involved in organ-specific autoimmunity, contact dermatitis, and some chronic inflammatory disorders of unknown etiology. In contrast, polarized TH2 responses favor a reduced protection against the majority of infectious agents (including HIV) and, in genetically predisposed hosts, are responsible for triggering of allergic atopic disorders.

Abstract: A large panel of human CD4+ T helper (Th) cell clones with established Th1, Th2, or Th0 profiles of cytokine secretion were examined for the expression of CD30, a member of the tumor necrosis factor receptor superfamily. Th1 clones expressed poor or no CD30 mRNA, and showed low or undetectable expression of both membrane and soluble CD30 (sCD30) protein, whereas Th2 clones showed both CD30 mRNA and membrane CD30 and released substantial amounts of sCD30. Th0 clones exhibited an intermediate pattern of CD30 expression and release. When T cells from the same donor were stimulated with three different antigens (purified protein derivative, PPD; Toxocara canis excretory/secretory antigen, TES; Lolium perenne group I, Lol p I), production of high concentrations of IFN-gamma, but not expression of CD30 or production of IL-4 and IL-5, were observed at any time after stimulation with PPD. Abstract: A new patch antenna is analysed which provides dual-frequency operation by means of two narrow slots close to the patch radiating edges. The two modes of operations show similar radiating properties. The ratio between the two frequencies can be well controlled within a range varying from 1.6 to 2, by using simple semi-empirical formulas derived from a physical model and tested by using a fullwave analysis. To obtain a more extended range of this frequency ratio, two tuning microstrip stubs are introduced on a back substrate. Satisfactory performances of simultaneous matching when using a single feed point is demonstrated. Several measurements are shown for both the input impedance and the radiation pattern.

Abstract: The performance of the ALEPH detector at the LEP e+e− collider is reviewed. The accuracy of the tracking detectors to measure the impact parameter and momentum of charged tracks is specified. Calorimeters are used to measure photons and neutral hadrons, and the accuracy obtained in energy and angle is given. An essential property of the detector is its ability to identify particles; the performance in identification of electrons, muons, neutrinos (from missing energy), charged hadrons, π0's and V0's is described.

Abstract: Objectives: To assess the evolution of cognitive dysfunction in early-onset multiple sclerosis, to identify clinical predictors of mental decline, and to determine its impact on a patient's everyday life. Design: The cognitive performance of 50 patients with multiple sclerosis on a neuropsychological battery was compared with that of 70 control subjects initially and again after a 4-year interval. Clinical predictors of cognitive impairment and its effect on daily life were analyzed by stepwise linear regression. Setting: The research clinic of a university department of neurology. Participants: A consecutive sample of 50 inpatients and outpatients with multiple sclerosis (mean disease duration, 1.58 years) and 70 demographically matched healthy control subjects selected from the patients' relatives and friends. Main Outcome Measures: Mean psychometric test scores of both groups at the initial and follow-up testing. Regression coefficients measuring the relationship between clinical parameters and cognitive capacity and between mental decline and performance of common tasks measured by the Environmental and the Incapacity Status scales. Results: Multiple sclerosis—related deficits in verbal memory and abstract reasoning on initial testing remained more or less stable on the retest, at which time linguistic disturbances on the Set and Token tests also emerged. A patient's initial disability level predicted decreased performance on only four of 13 cognitive variables, and disease duration did so on only two. Extent of intellectual decline on initial testing, initial disability level, and progressive course were independent determinants of handicap in a patient's work and social activities. Conclusions: Cognitive and neurological deficits appear not to develop in parallel. Yet cognitive dysfunction proves to be a predictor of handicap in everyday life, even in patients in the incipient phase of multiple sclerosis.

Abstract: We tested the hypothesis that the leaf area/sapwood area ratio in Scots pine (Pinus sylvestris L.) is influenced by site differences in water vapor pressure deficit of the air (D). Two stands of the same provenance were selected, one in western Scotland and one in eastern England, so that effects resulting from age, genetic variability, density and fertility were minimized. Compared with the Scots pine trees at the cooler and wetter site in Scotland, the trees at the warmer and drier site in England produced less leaf area per unit of conducting sapwood area both at a stem height of 1.3 m and at the base of the live crown, whereas stem permeability was similar at both sites. Also, trees at the drier site had less leaf area per unit branch cross-sectional area at the branch base than trees at the wetter site. For each site, the average values for leaf area, sapwood area and permeability were used, together with values of transpiration rates at different D, to calculate average stem water potential gradients. Changes in the leaf area/sapwood area ratio acted to maintain a similar water potential gradient in the stems of trees at both sites despite climatic differences between the sites.

Abstract: The SOHO Ultraviolet Coronagraph Spectrometer (UVCS/SOHO) is composed of three reflecting telescopes with external and internal occultation and a spectrometer assembly consisting of two toric grating spectrometers and a visible light polarimeter. The purpose of the UVCS instrument is to provide a body of data that can be used to address a broad range of scientific questions regarding the nature of the solar corona and the generation of the solar wind. The primary scientific goals are the following: to locate and characterize the coronal source regions of the solar wind, to identify and understand the dominant physical processes that accelerate the solar wind, to understand how the coronal plasma is heated in solar wind acceleration regions, and to increase the knowledge of coronal phenomena that control the physical properties of the solar wind as determined by in situ measurements. To progress toward these goals, the UVCS will perform ultraviolet spectroscopy and visible polarimetry to be combined with plasma diagnostic analysis techniques to provide detailed empirical descriptions of the extended solar corona from the coronal base to a heliocentric height of 12 solar radii.

Abstract: In this study, changes in plasma levels of calcitonin gene-related peptide (CGRP) and substance P (SP) during a spontaneous-like cluster headache attack provoked by nitroglycerin were evaluated. Peptide variations after spontaneous or sumatriptan-induced remission were also assessed. Blood was collected from the external jugular vein homolateral to the pain side of 30 male cluster headache patients; 18 men were in an active and 12 in a remission one. Plasma levels of CGRP and SP were determined using sensitive radioimmunoassays for each peptide. CGRP-like immunoreactivity (CGRP-LI) was found to be augmented in patients in an active period and became elevated further at the peak of the provoked attack. A complete reversal occurred both after spontaneous and sumatriptan-induced remission. On the contrary, nitroglycerin neither provoked a cluster headache attack nor altered CGRP-LI in the patients in a remission period. The augmented levels of CGRP-LI measured before and after nitroglycerin administration, when the provoked attack reached the maximum intensity, suggest an activation of the trigeminovascular system during the active period of cluster headache. Moreover, the clinical and biochemical actions showed by sumatriptan stress the involvement of serotonin in cluster headache mechanisms.

Abstract: Nitric oxide (NO) produces rapid osteoclast detachment and contraction in vitro, and this effect is accompanied by a profound inhibition of bone resorption. Work by others has confirmed these findings in vivo: inhibition of NO synthase [NOS; L-arginine, NADPH: oxygen oxidoreductase (NO-forming), EC 1.14.13.39] in normal rats is followed by increased bone resorption reflected by a marked loss in bone mineral density. In our present study, immunocytochemistry and Northern blotting show the presence of the constitutive calcium-sensitive NOS isoform (cNOS) in normal rat osteoclasts and in the human preosteoclast cell line (FLG 29.1). The inducible NOS isoform (iNOS) was also clearly demonstrable in the rat cells especially after treatment with gamma interferon (IFN-gamma) and bacterial wall products [lipopolysaccharide (LPS)], while a basal level of transcript was detected in the untreated human preosteoclast line. However NADPH-diaphorase activity was intense only in neonatal rat osteoclasts attached to bone, perhaps reflecting either enhancement of cNOS activity by calcium or increased amounts of the inducible isoform in activated osteoclasts in situ compared with isolated neonatal rat osteoclasts. These actively resorb devitalized bone but the untreated cells contain relatively low levels of NOS; they are extremely sensitive to inhibition by NO. The iNOS inhibitor aminoguanidine markedly enhances in vitro resorption by activated NOS-rich chick osteoclasts and by normal rat osteoclasts treated with LPS or IFN-gamma. In contrast, the nonselective NOS inhibitor NG-monomethyl-L-arginine inhibits resorption by untreated neonatal rat osteoclasts. Thus, osteoclast function may require intermittent calcium-stimulated increases in NO production by cNOS against a basal inhibitory background activity of the iNOS isoform. However, bone resorption depends on precursor replication and on the activity of the mature cells, and we found that the NO donor 3-morpholinosydnonimine (SIN-1) (50 microM) profoundly depressed replication in the human preosteoclast line. Taken together, these results strongly suggest that NO maintains a central control of bone resorption in both avian and mammalian species by exerting a powerful tonic restraint of osteoclast numbers and activity. The presence of NOS in human cells implies a similar function in man and that conventional views of calcium homoeostasis and skeletal metabolism will need substantial revision. Since NO also influences behavior of the osteoblast, the bone-forming cell, in vitro, a similar effect in vivo might imply a general influence on bone remodeling.

Abstract: This paper deals with an efficient dynamic channel allocation (DCA) technique applicable to terrestrial mobile cellular networks. A channel (or resource) is a fixed frequency bandwidth (FDMA), a specific time-slot within a frame (TDMA), or a particular code (CDMA), depending on the multiple access technique used. A cost function has been defined by which the optimum channel to be assigned on demand can be selected. In addition, a suitable mobility model has been derived to determine the effects of handovers on network performance. The performance of the proposed DCA technique has been derived by computer simulations in terms of call blocking and handover failure probabilities. Abstract: The content of total and carcinogenic polycyclic aromatic hydrocarbons (PAHs) in Italian foods was measured. The highest levels of PAHs were found in pizza baked in wood‐burning ovens and barbecued beef and pork. Relatively high levels were also found in beet greens and squash, apples and bread, fried beef, pork and rabbit, cured meats and chocolate. Conversely low levels were detected in potatoes and cooked fish, beverages and eggs. The daily intake of total and carcinogenic PAHs also was calculated by multiplying the average consumption of each food by its mean concentration of PAHs. Cereal and milk products, meat, vegetables and fruits were the highest contributors to total PAH intake, since these products are the most important dietary components in Italy. The calculated total dietary PAH intake was 3 μg/day per person. The calculated intake of carcinogenic PAHs was 1.4 μg/day per person. Abstract: We have recently shown that CD30, a member of the tumor necrosis factor/nerve growth factor receptor superfamily, is preferentially expressed by human T cell clones producing T helper (Th) type 2 cytokines. We report here that costimulation with an agonistic anti-CD30 monoclonal antibody enhanced antigen (Ag)-induced proliferation and cytokine secretion by established human Th2 and Th0 clones. Moreover, costimulation of peripheral blood mononuclear cells with the same anti-CD30 monoclonal antibody resulted in the preferential development of Ag-specific T cell lines and clones showing a Th2-like profile of cytokine secretion. In contrast, early blockade in bulk culture of CD30 ligand-CD30 interaction shifted the development of Ag-specific T cells towards the opposite (Th1-like) phenotype. Taken together, these data suggest that CD30 triggering of activated Th cells by CD30 ligand-expressing Ag-presenting cells may represent an important costimulatory signaling for the development of Th2-type responses.

Abstract: Efficient dynamic channel allocation techniques with handover queuing suitable for applications in mobile satellite cellular networks, are discussed. The channel assignment on demand is performed on the basis of the evaluation of a suitable cost function. Geostationary and low Earth orbit (LEO) satellites have been considered. In order to highlight the better performance of the dynamic techniques proposed, a performance comparison with a classical fixed channel allocation (FCA) has been carried out, as regards the probability that a newly arriving call is not completely served. It has also been shown that a higher traffic density, with respect to GEO systems, is manageable by means of LEO satellites. >

Abstract: 1. Relaxin (RLX) is a multifunctional hormone which, besides its role in pregnancy and parturition, has also been shown to influence the cardiovascular system. In this study, we investigated the effect of RLX on coronary flow of rat and guinea-pig hearts, isolated and perfused in a Langendorff apparatus. RLX was either added to the perfusion fluid at a concentration of 5 x 10(-9) M for a 20-min perfusion, or given as a bolus into the aortic cannula at concentrations of 10(-9) M, 5 x 10(-8) M dissolved in 1 ml of perfusion fluid. 2. RLX, given either for a 20-min perfusion or as a bolus in the aortic cannula to guinea-pig and rat isolated hearts, increased the coronary flow and the amount of nitrite, a stable end-product of nitric oxide (NO) metabolism, that appeared in the perfusates in a concentration-dependent fashion. 3. The increase in coronary flow and in nitrite in the perfusates induced by RLX was significantly reduced by pretreatment with the nitric oxide synthase (NOS) inhibitor, NG-monomethyl-L-arginine (L-NMMA, 10(-4) M). 4. The effects of RLX on coronary flow and nitrite amounts in the perfusates were compared with those induced by the endothelium-dependent vasodilator agent, acetylcholine (ACh, 10(-8)-10(-7) M), and by the endothelium-independent vasodilator agent, sodium nitroprusside (SNP, 10(-7)-10(-6) M). The results obtained show that RLX is more effective than ACh and SNP in increasing coronary flow. 5 The results of this study show that RLX increases coronary flow through stimulation of NO production; hence this hormone should be regarded as a novel agent capable of improving myocardial perfusion.

Abstract: The authors investigated the theoretical question of whether different kinds of encoding can affect judgments of learning (JOLs) beyond any indirect effects arising from the differences those kinds of encoding produce on the likelihood of recall. They found that JOLs were more accurate after encoding by means of intentional learning than after encoding by means of incidental learning, even when the likelihood of recall did not differ for those kinds of encoding (Experiment 1), and were more accurate when intentional encoding occurred by generating the responses than by reading the responses (Experiment 2). An aggregation effect for JOLs was also discovered: Making JOLs about the likelihood of recall for an aggregate of items yielded less overconfidence (and even underconfidence) in contrast to the typical overconfidence of item-by-item JOLs. The overall pattern of findings suggests that JOLs are theoretically rich and are based on more than whatever underlies the likelihood of recall.

Abstract: Ultramafic xenoliths from alkali basalts in the Per§ani Mountains in the Eastern Transylvanian Basin (ETB) of Romania are mainly spinel Iherzolites, although spinel harzburgites, websterites, clinopyroxenites and amphibole pyroxenites are also present. Amphibole veins cut some spinel peridotite samples. All are derived from the shallow lithospheric upper mantle. In general, textural variations are restricted to protogranular and porphyroclastic types, compared with the more varied textures found in mantle xenoliths from the alkali basalts of the neighbouring Pannonian Basin. Also, ETB peridotites are richer in amphibole. Thus, the mantle beneath the edge of the ETB is less deformed but more strongly metasomatized than the mantle closer to the centre of the Pannonian Basin. Mineralogical and bulk-rock geochemical variations resemble those of spinel Iherzolites from other sub-continental shallow mantle xenolith suites. There is no apparent correlation between deformation and geochemistry, and much of the major and trace element variation is due to variable extraction of picritic melts. The REE patterns of separated clinopyroxenes from the peridotite xenoliths are mostly LREE depleted, although clinopyroxenes from regions adjacent to amphibole veins have experienced an enrichment in La and Ce and a change in their Sr and Nd isotopic values towards those of the vein, while still retaining an overall LREE depletion. Clinopyroxenes from the websterites and clinopyroxenites are more variable.
