Abstract: 1. Steady lengthenings at different velocities (0.025-1.2 microns/s per half-sarcomere; temperature 2-5.5 degrees C) were imposed on isolated frog muscle fibres at the isometric tetanus plateau by means of a loudspeaker motor. The lengthening at the sarcomere level was measured by means of a striation follower either in fixed-end or in length-clamp mode. The force response was measured by a capacitance gauge transducer (resonance frequency 50 kHz). Preparations showing gross non-homogeneity during lengthening were excluded. 2. A steady tension was in all cases reached after about 20 nm per half-sarcomere of lengthening. Tension during this steady phase rose with speed of elongation up to 0.25-0.4 micron/s per half-sarcomere, when tension was 1.9-2 times isometric tetanic force (T0). Further increase in speed produced only very little increase in the steady tension. 3. During the transitory phase, before steady tension was reached, the tension rose monotonically if speed of lengthening was less than 0.25-0.3 micron/s per half-sarcomere; at higher speed the tension rose above the steady level, reaching a peak when extension was 10-14 nm per half-sarcomere, and then fell to the steady level. Tension at the peak continued to rise with speed of lengthening above 0.3 micron/s per half-sarcomere. 4. During the tension rise within the transitory phase of force response the segment elongated at a speed 15-20% lower than that imposed on the whole fibre, as a consequence of tendon compliance. 5. During the steady phase, non-homogeneity of lengthening speed began above a speed of lengthening which varied from fibre to fibre. At speeds below this value, segments elongated at the same speed as that imposed on the fibre. 6. Tension responses to large step stretches (up to 12 nm per half-sarcomere), applied at the plateau of isometric tetanus, showed that the instantaneous elasticity of contractile machinery is not responsible for the limit in force attained with high-speed lengthening. 7. Instantaneous stiffness was determined during the steady state of force response by superposing small steps (less than 1.5 nm per half-sarcomere) on steady lengthening at different velocities. Stiffness was 10-20% larger during lengthening than at the plateau of isometric tetanus and remained practically constant, independent of lengthening velocity, in the range of velocities used. 8. The results indicate that steady lengthening of a tetanized fibre induces a cross-bridge cycle characterized by fast detachment of the cross-bridge extended beyond a critical level.(ABSTRACT TRUNCATED AT 400 WORDS)

Abstract: Mrk 231 is a nearby ultra-luminous IR galaxy exhibiting a kpc-scale, multi-phase AGN-driven outflow. This galaxy represents the best target to investigate in detail the morphology and energetics of powerful outflows, as well as their still poorly-understood expansion mechanism and impact on the host galaxy. In this work, we present the best sensitivity and angular resolution maps of the molecular disk and outflow of Mrk 231, as traced by CO(2−1) and (3−2) observations obtained with the IRAM/PdBI. In addition, we analyze archival deep Chandra and NuSTAR X-ray observations. We use this unprecedented combination of multi-wavelength data sets to constrain the physical properties of both the molecular disk and outflow, the presence of a highly-ionized ultra-fast nuclear wind, and their connection. The molecular CO(2−1) outflow has a size of ~1 kpc, and extends in all directions around the nucleus, being more prominent along the south-west to north-east direction, suggesting a wide-angle biconical geometry. The maximum projected velocity of the outflow is nearly constant out to ~1 kpc, thus implying that the density of the outflowing material must decrease from the nucleus outwards as ~r-2. This suggests that either a large part of the gas leaves the flow during its expansion or that the bulk of the outflow has not yet reached out to ~1 kpc, thus implying a limit on its age of ~1 Myr. Mapping the mass and energy rates of the molecular outflow yields OF = [500−1000] M⊙ yr-1 and Ėkin,OF = [7−10] × 10^(43) erg s^(-1). The total kinetic energy of the outflow is Ekin,OF is of the same order of the total energy of the molecular disk, Edisk. Remarkably, our analysis of the X-ray data reveals a nuclear ultra-fast outflow (UFO) with velocity −20 000 km s^(-1), UFO = [0.3−2.1] M_⊙ yr^(-1), and momentum load UFO/ rad = [0.2−1.6]. We find Ėkin,UFO ~ Ėkin,OF as predicted for outflows undergoing an energy conserving expansion. This suggests that most of the UFO kinetic energy is transferred to mechanical energy of the kpc-scale outflow, strongly supporting that the energy released during accretion of matter onto super-massive black holes is the ultimate driver of giant massive outflows. The momentum flux OF derived for the large scale outflows in Mrk 231 enables us to estimate a momentum boost OF/ UFO ≈ [30−60]. The ratios Ėkin,UFO/Lbol,AGN = [1−5] % and Ėkin,OF/Lbol,AGN = [1−3] % agree with the requirements of the most popular models of AGN feedback.

Abstract: Summary Objective To assess the effect of rivastigmine in patients with mild cognitive impairment (MCI) on the time to clinical diagnosis of Alzheimer's disease (AD) and the rate of cognitive decline. Methods The study was a double-blind, randomised, placebo-controlled trial of up to 48 months. All patients had MCI operationally defined by having cognitive symptoms, a global clinical dementia rating stage of 0·5, a score of less than 9 on the New York University delayed paragraph recall test, and by not meeting the diagnostic criteria for AD. Primary efficacy variables were time to clinical diagnosis of AD, and change in performance on a cognitive test battery. This study is registered with the US National Institutes of Health clinical trials database (ClinicalTrials.gov), number NCT00000174. Findings Of 1018 study patients enrolled, 508 were randomly assigned to rivastigmine and 510 to placebo; 17·3% of patients on rivastigmine and 21·4% on placebo progressed to AD (hazard ratio 0·85 [95% CI 0·64–1·12]; p=0·225). There was no significant difference between the rivastigmine and placebo groups on the standardised Z score for the cognitive test battery measured as mean change from baseline to endpoint (−0·10 [95% CI −0·63 to 0·44], p=0·726). Serious adverse events were reported by 141 (27·9%) rivastigmine-treated patients and 155 (30·5%) patients on placebo; adverse events of all types were reported by 483 (95·6%) rivastigmine-treated patients and 472 (92·7%) placebo-treated patients. The predominant adverse events were cholinergic: the frequencies of nausea, vomiting, diarrhoea, and dizziness were two to four times higher in the rivastigmine group than in the placebo group. Interpretation There was no significant benefit of rivastigmine on the progression rate to AD or on cognitive function over 4 years. The overall rate of progression from MCI to AD in this randomised clinical trial was much lower than predicted. Rivastigmine treatment was not associated with any significant safety concerns.

Abstract: The energetic convenience of electrolytic water splitting is limited by thermodynamics. Consequently, significant levels of hydrogen production can only be obtained with an electrical energy consumption exceeding 45 kWh kg(-1)H2. Electrochemical reforming allows the overcoming of such thermodynamic limitations by replacing oxygen evolution with the oxidation of biomass-derived alcohols. Here we show that the use of an original anode material consisting of palladium nanoparticles deposited on to a three-dimensional architecture of titania nanotubes allows electrical energy savings up to 26.5 kWh kg(-1)H2 as compared with proton electrolyte membrane water electrolysis. A net energy analysis shows that for bio-ethanol with energy return of the invested energy larger than 5.1 (for example, cellulose), the electrochemical reforming energy balance is advantageous over proton electrolyte membrane water electrolysis.
