Abstract: Minor salivary gland biopsy specimens from patients with Sjogren's syndrome (primary and secondary) and from normal controls were examined with the four step biotin-avidin-immunoperoxidase assay. The composition of the infiltrating cells was similar in patients with both primary and secondary Sjogren's syndrome, consisting primarily of T lymphocytes with predominance of T helper/inducer cells. B lymphocytes (Leu-14) were approximately 20-35% of the infiltrating lymphocytes, while only a few OKM1 (monocytes/macrophages) cells and Leu-7+ (natural killer; NK) cells were observed. The majority of infiltrating lymphocytes expressed HLA-DR antigens. In the biopsy specimens of the controls there were no infiltrates; the scattered lymphocytes, however, were also predominantly T lymphocytes. Finally, the glandular epithelial cells (ducts and acini) were inappropriately expressing HLA-DR antigens, in contrast with controls where minimal HLA-DR expression was found.

Abstract: The incidence and nature of cryoglobulins, as well as their correlation with the clinical and serologic picture of Sjogren's syndrome (SS), were studied in the sera of 30 consecutive primary SS patients. Sera from 29 patients with systemic lupus erythematosus, 58 patients with rheumatoid arthritis, and 125 healthy blood donors were also studied. It was shown that one-third of the patients with SS had cryoglobulinemia. These cryoglobulins were mixed monoclonal IgM immunoglobulins, whereas those observed in systemic lupus erythematosus and rheumatoid arthritis patients were mixed polyclonal. The nature of the cryoglobulins was demonstrated using high-resolution electrophoresis combined with immunofixation. The presence of cryoglobulins in the sera of SS patients correlated with extraglandular disease and with antibodies to Ro (SS-A) and IgM rheumatoid factor. SS patients with cryoglobulins had lower serum C4 levels than did patients without cryoglobulins. These findings suggest that SS expresses, in addition to polyclonal B cell hyperreactivity, a monoclonal process in the absence of lymphoid neoplasia. Further, they show that the extraglandular manifestations of the syndrome may be due to an immune complex-mediated pathology.

Abstract: Kainic acid injections into the nucleus accumbens in rats induced severe loss of neuronal perikarya and the presence of gliosis in its vicinity, without affecting more distant areas. Spontaneous locomotor activity was elevated in the lesioned rats. After a low dose of d-amphetamine (1.5 mg/kg) no significant differences in locomotor activity were found between lesioned and sham-operated rats, while the increase in locomotor activity normally induced by a moderate dose of apomorphine (1 mg/kg) was blocked in lesioned rats. These results indicate that although dopamine receptors on the nucleus accumbens neurons are involved in the mechanisms mediating locomotor behaviour, the locomotor stimulant effect of d-amphetamine is not exclusively dependent on intra-accumbens dopaminergic activity.

Abstract: Aldehyde dehydrogenase was measured in primary cultures of hepatocytes obtained with a two-step collagenase perfusion either from human hepatic tissue or from livers of Fisher rats. Basal enzyme activity declines gradually as a function of time in culture, but remains at all times higher when measured with propionaldehyde and NAD (P/NAD) than with benzaldehyde and NADP (B/NADP). Treatment of the cultures with 2 microM of 3-methylcholanthrene for four days significantly increased the B-NADP activity of human and rat hepatocytes (tenfold and eightfold respectively). In human hepatocytes 3-methylcholanthrene increases also the P/NAD activity, but to a lesser extent (twofold), compared to the B/NADP activity. Due to the significant enhancement of B/NADP activity in cultures of human and rat hepatocytes after application of 3-methylcholanthrene, the initial difference in the basal activity levels between the P/NAD and B/NADP forms diminishes or, in the case of human hepatocytes, is even inverted. These results show for the first time that aldehyde dehydrogenase activity is increased in cultured human hepatocytes. This biochemical property is preserved in human and rat hepatocyte cultures, despite the rather quick loss of the basal aldehyde dehydrogenase activity.

Abstract: The Neurospora crassa am gene was used as a heterologous probe to identify clones from two independently constructed Aspergillus nidulans gene libraries. These clones have a common HindIII 1.85 kb fragment. This A. nidulans nucleotide stretch hybridises to a N. crassa 2.7 kb BamHI fragment of wild type DNA but not to a co-migrating fragment from the DNA of the N. crassa am132 deletion mutant. One A. nidulans clone was shown to complement the N. crasse am132 deletion strain. The N. crassa transformants show low levels (∼5%) of heterologous glutamate dehydrogenase activity. The A. nidulans gdhA gene was found to locate in N. crassa at both the “homologous” (i.e. am) site as well as non-nomologous sites. Partial nucleotide analysis of the fragment has revealed the 5′ end of the locus and considerable homology with the N. crassa am gene. We concluded that we have cloned the A. nidulans gdhA gene.

Abstract: The diamino pyrimidine ligand 2,4-diamino-5(3',4',5'-trimethovybenvyl)pyrimidine trimethoprim (trim) reacts with K2PtCl4, RhCl3 and AuCl3 to give the solid compleves (trimH)2[PtCl4], Pt(trim)2Cl2, Rh(trim)-(trimH)Cl4, Au(trim)Cl3 and Au(trim)3Cl3. Properties, conductance measurements and spectral (IR, electronic, nuclear magnetic resonance) data are discussed in terms of the nature of the bonding and the possible structural types. Trimethoprim acts as a bridging bidentate ligand, bonding through the amino groups.

Abstract: The sequential polypeptides (L-Arg-X-Gly)n, where X represents amino acid residues Ala, Val, and Leu, were prepared as models of arginine-rich histones to be used in studying their structure and their interactions with DNA. The polymerization was carried out on the pentachlorophenyl active esters of the appropriate tripeptides, while the toluene-4-sulfonyl group was used for protecting the arginine guanido group. CD was employed to investigate the conformation of (L-Arg-X-Gly)n polymers in aqueous solutions, at different pH, as well as in trifluoroenthanol and hexafluoroisopropyl alcohol solutions. In aqueous solutions (at pH 7 and 12) the prepared sequential polymers behaved as a random coil. The CD spectra in various trifluoroethanol–water or hexafluoroisopropyl alcohol–water mixtures indicated that the degree of helical conformation of the studied polytripeptides increased in the order of Ala Val Leu. The opposite was true for the β-structure. Characteristics of β-turn are excluded from the poly(L-Arg-L-Leu-Gly), which assumed the most pronounced helical conformation. The poly(L-Arg-L-Val-Gly) exerts a significant preference to the β-turn structure compared to that of poly(L-Arg-L-Ala-Gly). Thus the probability for helical, β-structure or β-turn conformations of the polymers was analyzed in relation to the bulkiness and length, and to the special features of the X-residue side chain (β-branching). We concluded that the prepared sequential arginine-containing polypeptides are plausible models for histone fractions, f3 and f2α1.
