Abstract: Osteoarthritis, the clinical syndrome of joint pain and dysfunction caused by joint degeneration, affects more people than any other joint disease. There are no consistently effective methods for preventing osteoarthritis or slowing its progression, and symptomatic treatments provide limited benefit for many patients. Osteoarthritis disables about 10% of people who are older than 60 years, compromises the quality of life of more than 20 million Americans, and costs the United States economy more than $60 billion per year. The incidence of osteoarthritis rises precipitously with age; as a result, the prevalence and burden of this disorder is increasing rapidly. Study of the patterns osteoarthritis incidence and prevalence shows that it occurs frequently in the hand, foot, knee, spine and hip, but rarely in the ankle, wrist, elbow, and shoulder, and the most important universal risk factors are age, excessive joint loading, and joint injury. Analysis of the impact of osteoarthritis raises questions that include: Why does the incidence increase progressively with age? Why are some joints rarely affected? How do mechanical forces cause joint degeneration? What biologic and mechanical factors slow or accelerate the rate of joint degeneration? Answering these questions could lead to effective methods of preventing osteoarthritis and slowing its progression.

Abstract: Tumors require a blood supply for growth and hematogenous dissemination. Much attention has been focused on the role of angiogenesis—the recruitment of new vessels into a tumor from pre-existing vessels. However, angiogenesis may not be the only mechanism by which tumors acquire a microcirculation. Highly aggressive and metastatic melanoma cells are capable of forming highly patterned vascular channels in vitro that are composed of a basement membrane that stains positive with the periodic acid-Schiff (PAS) reagent in the absence of endothelial cells and fibroblasts. These channels formed in vitro are identical morphologically to PAS-positive channels in histological preparations from highly aggressive primary uveal melanomas, in the vertical growth phase of cutaneous melanomas, and in metastatic uveal and cutaneous melanoma. The generation of microvascular channels by genetically deregulated, aggressive tumor cells was termed “vasculogenic mimicry” to emphasize their de novo generation without participation by endothelial cells and independent of angiogenesis. Techniques designed to identify the tumor microcirculation by the staining of endothelial cells may not be applicable to tumors that express vasculogenic mimicry. Although it is not known if therapeutic strategies targeting endothelial cells will be effective in tumors whose blood supply is formed by tumor cells in the absence of angiogenesis, the biomechanical and molecular events that regulate vasculogenic mimicry provide opportunities for the development of novel forms of tumor-targeted treatments. The unique patterning characteristic of vasculogenic mimicry provides an opportunity to design noninvasive imaging techniques to detect highly aggressive neoplasms and their metastases.

Abstract: Meinshausen and Buhlmann [Ann. Statist. 34 (2006) 1436–1462] showed that, for neighborhood selection in Gaussian graphical models, under a neighborhood stability condition, the LASSO is consistent, even when the number of variables is of greater order than the sample size. Zhao and Yu [(2006) J. Machine Learning Research 7 2541–2567] formalized the neighborhood stability condition in the context of linear regression as a strong irrepresentable condition. That paper showed that under this condition, the LASSO selects exactly the set of nonzero regression coefficients, provided that these coefficients are bounded away from zero at a certain rate. In this paper, the regression coefficients outside an ideal model are assumed to be small, but not necessarily zero. Under a sparse Riesz condition on the correlation of design variables, we prove that the LASSO selects a model of the correct order of dimensionality, controls the bias of the selected model at a level determined by the contributions of small regression coefficients and threshold bias, and selects all coefficients of greater order than the bias of the selected model. Moreover, as a consequence of this rate consistency of the LASSO in model selection, it is proved that the sum of error squares for the mean response and the lα-loss for the regression coefficients converge at the best possible rates under the given conditions. An interesting aspect of our results is that the logarithm of the number of variables can be of the same order as the sample size for certain random dependent designs.
