Abstract: Increasing cuticular wax accumulation in plants has been associated with improving drought tolerance in plants. In this study, a cDNA clone encoding the SlSHN1 transcription factor, the closest ortholog to WIN/SHN1 gene in Arabidopsis, was isolated from tomato plant. Expression analysis of SlSHN1 indicated that it is induced in response to drought conditions. The over-expression of SlSHN1 in tomato under the control of the constitutive CaMV 35S promoter produced plants that showed mild growth retardation phenotype with shiny and dark green leaves. Scanning electron microscopy showed that the over-expression of SlSHN1 in tomato resulted in higher cuticular wax deposition on leaf epidermial tissue when compared to non-transformed plants. Expression analysis in transgenic lines over-expressing SlSHN1 indicated that several wax-related synthesis genes were induced. Transgenic tomato plants over-expressing SlSHN1 showed higher drought tolerance when compared with wild type plants; this was reflected in delayed wilting of transgenic lines, improved water status and reduced water loss rate when compared with wild type plants. In conclusion, the SlSHN1 gene can modulate wax accumulation and could be utilized to enhance drought tolerance in tomato plant.

Abstract: There is increasing interest in the pharmaceutical and food industries to substitute synthetic chemicals with naturally occurring compounds possessing bioactive properties. Plants are valuable sources of bioactive compounds. The present study investigates the chemical composition and antioxidant, antimicrobial, and anticancer activities of ethanolic extracts (EEs) and essential oils (EOs) from two species in the Lamiaceae family, Ocimum basilicum L. and Thymus algeriensis Boiss. & Reut., cultivated in the Algerian Saharan Atlas. The total flavonoid contents of the plants’ ethanolic extracts were determined by the aluminium chloride method, while the total phenols were determined using the Folin-Ciocalteu method. Essential oils were obtained by hydrodistillation of the aerial parts of the plants and were analysed by GC-MS. The free radical-scavenging ability and antioxidant potential of the plants’ EEs and EOs were probed using the 2, 2-diphenyl-picrylhydrazyl radical-scavenging, ABTS radical-scavenging, ferric-reducing power and phosphomolybdenum assays. The antimicrobial activities were evaluated against several pathogens characteristic of gram-negative bacteria (three species), gram-positive bacteria (three species) and fungi (two species). The microdilution method was used to estimate the minimum inhibitory concentrations (MICs). The oils’ anticancer potential against several cancer types was also studied using the MTT assay and reported as the toxic doses that resulted in a 50% reduction in cancer cell growth (LD50). Phenolic compounds in the EEs from both plants were analysed by HPLC and demonstrated a rich flavonoid content. Chemical analysis of the essential oil from Ocimum basilicum revealed 26 unique compounds, with linalool (52.1%) and linalyl acetate (19.1%) as the major compounds. A total of 29 compounds were identified in the essential oil from Thymus algeriensis, with α-terpinyl acetate (47.4%), neryl acetate (9.6%), and α-pinene (6.8%) as the major compounds. The ethanolic extracts and essential oils from both plants exhibited moderate antioxidant activities and moderate to weak antimicrobial activities. Furthermore, anticancer activities against the examined human cancer cell lines were associated with only the EOs from both plants, with LD50 values ranging between 300 and 1000 μg/mL. The results suggest that the bioactive compounds found in the ethanolic extracts and essential oils from Ocimum basilicum and Thymus algeriensis, with diverse antioxidant, antimicrobial and anticancer activities, may have beneficial applications in nutraceutical and pharmaceutical technologies.

Abstract: The main objective of this study was to assess the spatial and temporal variability of groundwater level fluctuations in the Amman–Zarqa basin, during the period 2001–2005. In the year 2003, as a consequence of war, there was a sudden increase in the population in this basin. Knowing that the basin is already heavily populated and witnesses most of the human and industrial activities in Jordan, this study was prompted to help make wise water resources management decisions to cope with the new situation. Data from 31 fairly distributed wells in the upper aquifer of the basin were subjected to geostatistical treatment. Kriging interpolation techniques have indicated that the groundwater flow directions remained almost constant over the years. The two main directions are SW–NE and E–W. Kriging mapped fluctuations have also showed that drop and rise events are localized in the basin. Forecasting possibilities for management purposes were tackled using autocorrelation analysis. The constructed autocorrelograms indicated, in general, the temporal dependence of seasonal water level fluctuations, and that forecasting can be carried out within a period of 3–21 months. Several suggestions were made to mitigate the drop and rise hazards in the detected sites.
