Abstract: Filamins (FLN) are large dimeric proteins that cross-link actin and work as important scaffolds in human cells. FLNs consist of an N-terminal actin-binding domain followed by 24 immunoglobulin-like domains (FLN1–24). FLN domains are divided into four subgroups based on their amino acid sequences. One of these subgroups, including domains 4, 9, 12, 17, 19, 21, and 23, shares a similar ligand-binding site between the β strands C and D. Several proteins, such as integrins β2 and β7, glycoprotein Ibα (GPIbα), and migfilin, have been shown to bind to this site. Here, we computationally estimated the binding free energies of filamin A (FLNa) subunits with bound peptides using the molecular mechanics-generalized Born surface area (MMGBSA) method. The obtained computational results correlated well with the experimental data, and they ranked efficiently both the binding of one ligand to all used FLNa-domains and the binding of all used ligands to FLNa21. Abstract: More than 1 year ago, the subject area on Life Cycle Sustainability Assessment has been launched (Zamagni 2012) in order to provide a virtual place in which scientists from different disciplinary fields could discuss the main challenges in addressing sustainability with a life cycle perspective. The challenge has been taken on also by the editors of three subject areas that deal with sustainability from complementary angles—Life Cycle Sustainability Assessment (LCSA), Life Cycle Costing (LCC), and Social Life Cycle Assessment (SLCA). The result was the launch of a call for papers for a special issue on LCSA, whose results we are glad to present. Our initiative took its point of departure from two facts: (1) sustainability has become quite a keyword in any decisioncontext situation and the scientific community has the duty to provide its contribution in demonstrating what might be sustainable and how to measure it with a scientific approach; (2) the life cycle approach is considered to provide a valuable support in integrating sustainability into the design, innovation, and evaluation of products and services, as can be seen in several environmental policies at the European and international level in which life cycle thinking represents the backbone. As far as the timely and challenging aspects of sustainability are concerned, the EU has mainstreamed sustainable development into a broad range of its policies, such as the initiatives against climate change and the promotion of a low-carbon economy. The European Council (2009) confirmed that sustainable development remains a fundamental objective of the European Union under the Lisbon Treaty. Moreover, as emphasized in the Presidency’s report on the review of the Union’s Sustainable Development Strategy, the strategy will continue to provide a long-term vision and constitute the overarching policy framework for all Union policies and strategies (Council of the European Union 2009). Sustainability is a concept that needs to be addressed not only at the policy level but also in the business context: many companies have included sustainability in their mission, also driven by an increasing demand for sustainable products by more aware consumers. Although the policy and the business context deal with sustainability in different ways, given the diversity of needs, a common element for both is the complexity entailed by the assessment. Abstract: Theoretical models are presented for the enhancement of the electron density at a positron in a semiconductor or insulator host. The model better suited for typical semiconductors is based on the many-body theory for the screening of a positron in electron gas. The starting point of the model for insulators is the atomic polarizability. The common parameter in both models is the high-frequency dielectric constant. Moreover, the enhancement depends on the ambient electron density in the semiconductor model and on the unit-cell volume in the insulator model. With use of the models developed, positron lifetimes in perfect semiconductor and insulator crystals have been calculated. In the calculations, three-dimensional electron densities and electrostatic potentials are obtained by atomic superposition and the fully three-dimensional positron wave functions are solved by a relaxation method. The calculated positron lifetimes agree with the experimental ones within a few picoseconds. Moreover, we have used the model to predict lifetimes of positrons trapped by lattice defects such as vacancies and vacancy clusters.

Abstract: In this study adaptations in body composition, physical fitness and metabolic health were examined during 21 weeks of endurance and/or strength training in 39- to 64-year-old healthy women. Subjects (n = 62) were randomized into endurance training (E), strength training (S), combined strength and endurance training (SE), or control groups (C). S and E trained 2 and SE 2 + 2 times in a week. Muscle strength and maximal oxygen uptake (VO2max) were measured. Leg extension strength increased 9 ± 8% in S (P < 0.001), 12 ± 8% in SE (P < 0.001) and 3 ± 4% in E (P = 0.036), and isometric bench press 20% only in both S and SE (P < 0.001). VO2max increased 23 ± 18% in E and 16 ± 12% in SE (both P < 0.001). The changes in the total body fat (dual X-ray absorptiometry) did not differ between groups, but significant decreases were observed in E (−5.9%, P = 0.022) and SE (−4.8%, P = 0.005). Lean mass of the legs increased 2.2–2.9% (P = 0.004–0.010) in S, SE and E. There were no differences between the groups in the changes in blood lipids, blood pressure or serum glucose and insulin. Total cholesterol and low-density lipoprotein cholesterol decreased and high-density lipoprotein cholesterol increased in E. Both S and SE showed small decreases in serum fasting insulin. Both endurance and strength training and their combination led to expected training-specific improvements in physical fitness, without interference in fitness or muscle mass development. All training methods led to increases in lean body mass, but decreases in body fat and modest improvements in metabolic risk factors were more evident with aerobic training than strength training.
