Abstract: Although single base-pair substitutions in splice junctions constitute at least 10% of all mutations causing human inherited disease, the factors that determine their phenotypic consequences at the RNA level remain to be fully elucidated. Employing a neural network for splice-site recognition, we performed a meta-analysis of 478 disease-associated splicing mutations, in 38 different genes, for which detailed laboratory-based mRNA phenotype assessment had been performed. Inspection of the ±50-bp DNA sequence context of the mutations revealed that exon skipping was the preferred phenotype when the immediate vicinity of the affected exon–intron junctions was devoid of alternative splice-sites. By contrast, in the presence of at least one such motif, cryptic splice-site utilization, became more prevalent. This association was, however, confined to donor splice-sites. Outside the obligate dinucleotide, the spatial distribution of pathological mutations was found to differ significantly from that of SNPs. Whereas disease-associated lesions clustered at positions –1 and +3 to +6 for donor sites and –3 for acceptor sites, SNPs were found to be almost evenly distributed over all sequence positions considered. When all putative missense mutations in the vicinity of splice-sites were extracted from the Human Gene Mutation Database for the 38 studied genes, a significantly higher proportion of changes at donor sites (37/152; 24.3%) than at acceptor splice-sites (1/142; 0.7%) was found to reduce the neural network signal emitted by the respective splice-site. Based upon these findings, we estimate that some 1.6% of disease-causing missense substitutions in human genes are likely to affect the mRNA splicing phenotype. Abstract: Nitrate and ammonium have different effects on many biochemical and physiological processes in plants, and at high concentrations this can lead to markedly different growth responses. Most plant species show reduced growth, smaller leaves and a stunted root system when exposed to high ammonium concentrations, and in severe cases this leads to chlorosis. Although well known, ammonium toxicity is poorly understood and is generally considered to be the result of one or more of the following effects; (i) ammonium-induced mineral nutrient deficiency, arising from the impaired uptake of metal ions; (ii) secondary growth inhibition arising from the acidification of the rooting medium; (iii) alterations in intracellular pH and osmotic balance; (iv) uncoupling of photophosphorylation from electron transport, following the accumulation of ammonium in leaves; and (v) altered polyamine and phytohormone metabolism.
These hypotheses are reviewed in the light of the available literature and experimental evidence from own experiments. It is concluded that no mechanism on its own provides an adequate explanation of the available data.

Abstract: Fermenthistoehemische Untersuchungen yon Go~o~I (1953), Bv~aSTO~E (1957) sowie MOLO~EY, McPi~E~so~ and F L I ~ E L ~ (1960) ergaben, dab menschliehe Gewebsmastzellen und die ZelIen der normalen oder leuk~mischen neutrophilen Myelopoese mit Ausnahme der Myeloblasten Chloracylester mit einer Intensit~t spalten, die yon anderen Gewebskomponenten bci weitem nicht erreicht wird. Diese VerhMten gestattet die genannten Zellen fast elektiv darzustellen. Als fiir diesen Zweek besonders gut geeignet erwies sieh das Substrat I~aphthol-ASD-Ch]oraeetat. Bei methodischen Studien zum Nachweis der NaphtholASD-Chloracetat-Esterase zeigte sich, dub das Ferment auBerordentlich stabil und gegeniiber den Einfliissen histologiseher Teehnik weitgehend unempfindlieh ist. So konnten wir in mehr Ms zwei Jahre lang unfixiert und lufttroeken aufbewahrten Blutausstriehen erstaunlicherweise kein Naehlassen der NeutrophilenaktivitEt Ieststellen. Sogar troekenes, d. h. wasserfreies Erhitzen bis 100°C und auch die bei der routinem~Bigen Paraffineinbettang verwendeten organischen Verbindungen (/~thylalkohol, Methylbenzoat, Benzol, Paraffin) verursachten keine naehweisbare Aktivit~tsdepression. Nur bei Erhitzen der Pr~p~rate im Wasserbad trat oberhalb 60°C eine Aktivit~tsminderung ein, die mit steigender Temperatur und verI~ngerter Einwirkungszeit in vSlligen Aktivit~tsverlust fiberging. Dieses ungewShnliche VerhMtcn ermSglicht den Naehweis des Fermentes an gew6hnliehen Para/]inschnitten. Jedoch gestattet die yon MoLo~¥ u. Mitarb. angegebene Methodik keine befriedigende cytologische Auswertung yon Schnittpr/~paraten. Deshalb arbeite~en wir eine eigene Modifikation aus. Als Kupplungssalz benutzten wir das yon D~vis and OmCSTEZ~ angegebene hexazotierte Pararosanilin. Dies verbinder sieh mit dem Spaltprodukt des NaphthoLASD-CMoracetates zu einem wasserunl6slichen, leuehtend roten l~arbstoff und gewii~hrleistet eine sehr exakte Lokalisation der Fermentaktivit/~t. Praktiseh gehen wir folgendermaBen vor. Ein Tropfen 4%iges Pararosanilin in 2nHCI und ein Tropfen 4%ige w~Brige NatriumnitritlSsung werden vermiseht und nach eingetretener Hexazotierung (nuch 30--60 sec) mit 30 ml Miehae]ispuffer (0,1 M; pH 6.8--7,6) verdfinnt. Der pH-Wert wird unter pH-Meter-Kontrolle mit HC1 auf 6,3 eingesteltt. Dann werden 10 mg NaphthoLASD-Chlor-acetat in 1 ml Dimethytformamid aufgelOst. Beide LSsungen werden unter kr~ftigem l~fihren zusammengegeben, wobei ein milchiger, hellroter, flockiger Niederschlag auf~ritt. Das Filtrat ist jedoch klar und leicht rStlieh gef~rbt. In dieses Medium werden bei Zimmertemperatur mOgliehst diinne, dem Objekt.tri~ger gut anhaftende, in fiblicher Weise entparaffiniel%e Sehnitte eingestellt. Die .Inkubationszeit betr/igt 30rain. Naeh der Inkubation werden die Schnitte gut unter Leitungswasser abgespfilt, 5 rain in saurem H~malaun naeh M~.~E~ gegenge~rbt, gebl~ut und in Gelatinol eingedeckt. Auf diese Weise erzielten wir selbst in Schnitten yon 12 Ja.hre a lan formalinfixierten ParaffinbtSeken eine tadellose Reaktion. Aueh in Maximowscher LSsung fixiertes Untersuchungsgut erwies sich Ms geeignet. SchlieBhch liefert das Verfahren bei Blurund Knochenmarkausstrichen sowie Tupfpriiparaten (in Methanol-Formol nach I~rLow fixiert) und bei nativen Kryostatschnitten ausgezeichnete Resultate. l~ach Entkalkung mit Salzs~ure-Ameisensi~ure-Gemisch geht die Aktivit/~t im Knoehengewebe verloren, nicht dagegen bei Anwendung yon EDTA. Die angegebene Methodik hat mehrere Vorzfige. Dureh Verwendung yon Paraffinschnitten ist gegenfiber Gefrierschnitten aus nativem oder formolfixiertem Material eine se:hr viel bessere Erhaltung morphologischer Details erreicht. Hexazotiertes Pararosanilin als Kupplungssalz bewirkt eine exakte Lokalisation des Reaktionsproduktes bei fast vollkommen feh]ender Untergrundfi~rbung. Die leuehtend rote, sehr kr/iftige Anfiirbung hebt die positiven Zellen gegeniiber den negativen sehr deutlich hervor und kontrastiert stark zu den blau gefi~rbten Zellkemen. SchlieBlich ist die beim Verfahren yon MOLO~V.Y u. Mitarb. h/iufig ungleichm~Bige Anf~rbung yon sehr zelh'eichen Pr~paraten weitgehend verringert. Besonders vorteilhaft ist jedoch, dab die Reaktion an routinem~Big hergestellten Paraffinschnitten durchgefiihrt werden kann. Aui diese Weise ist es mSgtich, schneU und ohne zcitraubende spezielle Fixiernngsund Einbettungsverfahren Untersuchungsgut in beliebiger Menge zu erhalten. Unsere Beobaehtung ermSglicht eine rasche und sichere Orientierung fiber Menge und Verteitungsmuster der mit der Reaktion erfaBbaren Zetlen im Paraffinschnitt. Insbesondere abet gelingt es, die selbst bei Giemsa-Farbung im Schnittpr/iparat oft schwer yon anderen Zel]en abgrenzbaren nicht

Abstract: Essential tremor is a common movement disorder. Tremor severity and handicap vary widely, but most patients with essential tremor do not receive a diagnosis and hence are never treated. Furthermore, many patients abandon treatment because of side-effects or poor efficacy. A newly developed algorithm, based on the logarithmic relation between tremor amplitude and clinical tremor ratings, can be used to compare the magnitude of effect of available treatments. Drugs with established efficacy (propranolol and primidone) produce a mean tremor reduction of about 50%. Deep brain stimulation (DBS) in the thalamic nucleus ventrointermedius or neighbouring subthalamic structures reduces tremor by about 90%. However, no controlled trials of DBS have been done, and the best target is still uncertain. Better drugs are needed, and controlled trials are required to determine the safety and efficacy of DBS in the nucleus ventrointermedius and neighbouring subthalamic structures.
