Abstract: Electrophysiological mapping was used to study frequency representation in the inferior colliculus (IC) of the mouse. In the lateral nucleus (LN) only part of the frequency range of hearing was represented and tonotopicity was separate from that in the rest of the IC. Highest frequencies occupied the medial part (M) of the central nucleus (CN). A single complete representation of the hearing range was present only if representations in the dorsal cortex (plus dorsomedial nucleus) and CN (including M) were combined. Continuous isofrequency planes making up these nuclei (without the lateral part of the CN) were reconstructed. They tilted from medial to lateral and from caudal to rostral. The steepness of the slopes increased from caudal to rostral and from dorsal to ventral (i.e., with increasing frequency). Isofrequency planes had similar angles of deviation from the horizontal plane as described for dendritic laminae in the CN. Differences of mapping in the lateral part of the CN from that in the rest of the CN could be explained by the different organization of laminae in this part. The relative amounts of IC depth and volume occupied by parts of the mouse audible frequency range were quantified. Frequency representation along IC depth was not proportional to that along cochlear length. Compared with the relative density of afferent nerve fiber supply within given frequency ranges represented along the basilar membrane, there is a relative under-representation in the IC up to 15-20 kHz and an over-representation of higher frequencies. Highest absolute tone sensitivity (lowest threshold) was found in neurons forming a column (running perpendicular to isofrequency planes) in the center of the IC. Results are discussed with regard to frequency representation, intrinsic neuronal organization, and functional segregation in the IC of mammals.

Abstract: Interphase chromatin is arranged into topologically separated domains comprising gene expression and replication units through genomic sequence elements, so-called MAR or SAR regions (for matrix- or scaffold-associating regions). S/MAR regions are located near the boundaries of actively transcribed genes and were shown to influence their activity. We show that scaffold attachment factor B (SAF-B), which specifically binds to S/MAR regions, interacts with RNA polymerase II (RNA pol II) and a subset of serine-/arginine-rich RNA processing factors (SR proteins). SAF-B localized to the nucleus in a speckled pattern that coincided with the distribution of the SR protein SC35. Furthermore, we show that overexpressed SAF-B induced an increase of the 10S splice product using an E1A reporter gene and repressed the activity of an S/MAR flanked CAT reporter gene construct in vivo . This indicates an association of SAF-B with SR proteins and components of the transcription machinery. Our results describe the coupling of a chromatin organizing S/MAR element with transcription and pre-mRNA processing components and we propose that SAF-B serves as a molecular base to assemble a 'transcriptosome complex' in the vicinity of actively transcribed genes.

Abstract: Methane emission rates from an Italian rice paddy field showed diel and seasonal variations. The seasonal variations were not closely related to soil temperatures. However, the dieL changes of CH4 fluxes were significantly correlated with the diel changes of the temperature in a particular soil depth. The soil depths with the best correlations between CH4 flux and temperature were shallow (1–5cm) in May and June, deep (10–15cm) in June and July, and again shallow (1–5 cm) in August. Apparent activation energies (Ea) calculated from these correlations using the Arrhenius model were relatively low (50–150 kJ mol−1) in May and June, but increased to higher values (80–450 kJ mol−1) in August. In the laboratory, CH4 emission from two rice cultures incubated at temperatures between 20 and 38°C showed Eα. values of 41 and 53 kJ mol−1) Methane production in anoxic paddy soil suspensions incubated between 7 and 43°C showed Eα values between 53 and 132 kJ mol−1 with an average value of 85 kJ mol−1) and in pure cultures of hydrogenotrophic methanogenic bacteria Ea values between 77 and 173 (average 126) kJ mol−1. It is suggested that diel changes of soil properties other than temperature affect CH4 emission rates, e.g. diel changes in root exudation or in efficiency of CH4 oxidation in the rhizosphere.
