TL;DR: Although PEGylation as a tool to improve the pharmacokinetics of nanomedicines is well established and is used clinically, other benefits of ‘PEGnology', including enhancement of physicochemical properties and/or biocompatibility of actives and or drug carriers, as well as mucosal delivery, have attracted less attention.
Abstract: Introduction: Recent biopharma deals related to nanocarrier drug delivery technologies highlight the emergence of nanomedicine. This is perhaps an expected culmination of many years of research demonstrating the potential of nanomedicine as the next generation of therapeutics with improved performance. PEGylated nanocarriers play a key role within this field. Areas covered: The drug delivery advantages of nanomedicines in general are discussed, focusing on nanocarriers and PEGylated nanomedicines, including products under current development/clinical evaluation. Well-established drug delivery benefits of PEGylation (e.g., prolonged circulation) are only briefly covered. Instead, attention is deliberately made to less commonly reported advantages of PEGylation, including mucosal delivery of nanomedicines. Finally, some of the issues related to the safety of PEGylated nanomedicines in clinical application are discussed. Abstract: Three- and four-year-old children (N = 131) were tested for their sensitivity to the accuracy and inaccuracy of informants. Children were presented with one of three conditions. In the Accurate-Inaccurate condition, one informant named objects accurately whereas the other named them inaccurately. In the Accurate-Neutral condition, one informant named objects accurately whereas the other merely drew attention to them. Finally in the Inaccurate-Neutral condition, one informant named objects inaccurately whereas the other merely drew attention to them. In subsequent test trials, 4-year-olds preferred to seek and accept information in a selective fashion across all three conditions, suggesting that they monitor informants for both accuracy and inaccuracy. By contrast, 3-year-olds were selective in the Accurate-Inaccurate and Inaccurate-Neutral conditions but not in the Accurate-Neutral condition, suggesting that they monitor informants only for inaccuracy and take accuracy for granted.

Abstract: This paper proposes an alternative approach to rural tourism that returns to a more traditional model of development: large, flagship attractions that act as a ‘growth pole’ for the local economy and community. It questions some of the accepted beliefs about sustainable rural tourism development current in recent years. It is based on a case study of Alnwick Garden in Northumberland, England. It suggests that, under certain circumstances, flagship or mega-attractions can not only increase substantially the number of visitors to rural areas but also, through appropriate policies and processes, can underpin the longer-term, sustainable development of those areas.

Abstract: Although rapid eye movement sleep (REM) is regularly implicated in emotional memory consolidation, the role of slow-wave sleep (SWS) in this process is largely uncharacterized. In the present study, we investigated the relative impacts of nocturnal SWS and REM upon the consolidation of emotional memories using functional magnetic resonance imaging (fMRI) and polysomnography (PSG). Participants encoded emotionally positive, negative, and neutral images (remote memories) before a night of PSG-monitored sleep. Twenty-four hours later, they encoded a second set of images (recent memories) immediately before a recognition test in an MRI scanner. SWS predicted superior memory for remote negative images and a reduction in right hippocampal responses during the recollection of these items. REM, however, predicted an overnight increase in hippocampal–neocortical connectivity associated with negative remote memory. These findings provide physiological support for sequential views of sleep-dependent memory processing, demonstrating that SWS and REM serve distinct but complementary functions in consolidation. Furthermore, these findings extend those ideas to emotional memory by showing that, once selectively reorganized away from the hippocampus during SWS, emotionally aversive representations undergo a comparably targeted process during subsequent REM.
