Abstract: Summary Background Drug patch tests (PTs) can reproduce delayed hypersensitivity to drugs and entail a moderate re-exposure of patients to offending drugs. Objectives To determine the value of PTs for identifying the responsible drug in severe cutaneous adverse drug reactions (SCARs) such as acute generalized exanthematous pustulosis (AGEP), drug reaction with eosinophilia and systemic symptoms (DRESS) and Stevens–Johnson syndrome/toxic epidermal necrolysis (SJS/TEN). Methods In a multicentre study, PTs were conducted on patients referred for DRESS, AGEP or SJS/TEN within 1 year of their SCAR. All drugs administered in the 2 months prior to and the week following the onset of the SCAR were tested. Results Among the 134 patients included (48 male, 86 female; mean age 51·7 years), positive drug PTs were obtained for 24 different drugs. These included positive tests for 64% (46/72) of patients with DRESS, 58% (26/45) of those with AGEP and 24% (4/17) of those with SJS/TEN, with only one relapse of AGEP. The value of PTs depended on the type of drug and the type of SCAR (e.g. carbamazepine was positive in 11/13 DRESS cases but none of the five SJS/TEN cases). PTs were frequently positive for beta lactams (22 cases), pristinamycin (11 cases) and in DRESS with pump proton inhibitors (five cases), but were usually negative for allopurinol and salazopyrin. Of 18 patients with DRESS, eight had virus reactivation and positive PTs. In DRESS, multiple drug reactivity was frequent (18% of cases), with patients remaining sensitized many years later. Conclusions PTs are useful and safe for identifying agents inducing SCAR.

Abstract: Recent years have faced stimulating developments in the functionalization of electrode surfaces with biological materials, notably due to the significant input of nanosciences and nanotechnology. In this review (over 450 references), we are discussing the interest of both nano-objects (metal nanoparticles and quantum dots, carbon nanotubes and graphene) and nano-engineered and/or nanostructured materials (template-based materials, advanced organic polymers) for the rational design of bio-functionalized electrodes and related (bio)sensing systems. The attractiveness of such nanomaterials relies not only on their ability to act as effective immobilization matrices, which are, e.g., likely to enhance the long-term stability of bioelectrochemical devices, but also on their intrinsic and unique features (large surface areas, electrocatalytic properties, controlled morphology and structure, possible use as labels) that can be advantageously combined with the functioning of biomolecules, thus contributing to improved bioelectrode performance in terms of sensitivity and selectivity (enzymatic biosensors, DNA sensors, immunosensors and cell sensors) or power (biofuel cells).

Abstract: We report here a simple method for controlling the crystal size and morphology of zeolitic imidazolate framework-8 (ZIF-8) nanocrystals in methanol solution. ZIF-8 crystals were prepared by mixing 2-methylimidazole (Hmim) with various zinc salts for 1 h and using a Hmim/Zn(+2) salt molar ratio of 8/1. All products prepared were assigned to a sodalite-type structure and both particle size and morphology were found to be dependent on the reactivity of the Zn(+2) salt. Small ZIF-8 crystals with diameters varying between ca. 50 and 200 nm were obtained with reactive zinc salts like Zn(acac)2, Zn(NO3)2, ZnSO4 or Zn(ClO4)2 as demonstrated by SEM, TEM and DLS analyses. The use of ZnCl2, Zn(OAc)2 or ZnI2 afforded crystals with sizes varying between ca. 350 and 650 nm. Finally, the low reactive ZnBr2 salt was found to generate microsized crystals. Taking ZIF-8 crystals prepared from Zn(NO3)2, Zn(OAc)2 and ZnBr2 as representatives and through thermogravimetric analysis and BET measurements, we also demonstrated that changes in particle size induced changes in stability and adsorption properties. The small sized ZIF-8 crystals produced from Zn(NO3)2 were found to exhibit the highest surface area (1700 m2 g−1) and the best catalytic activity in Knoevenagel and Friedlander reactions.

Abstract: Objective We developed a validated index for assessing histological disease activity in UC and established its responsiveness. Methods Two hundred biopsies were scored. The outcome was the Global Visual Evaluation (GVE). Eight histological features were tested. The Nancy index was developed by multiple linear regression and bootstrap process to create an index that best matched the GVE. Goodness of fit was assessed by the adjusted R squared (adjusted R 2 ). The second step was the validation of the index: 100 biopsies were scored for the Nancy index by three pathologists from different centres. Inter-reader reliability was evaluated for each reader. The relationship between the change of the Nancy index and the Geboes index was assessed to assess the responsiveness. Results After backward selection with bootstrap validation, 3/8 items were selected: ulceration (adjusted R 2 =0.55), acute inflammatory infiltrate (adjusted R 2 =0.88) and chronic inflammatory infiltrate (adjusted R 2 =0.79). The Nancy index is defined by a 5-level classification ranging from grade 0 (absence of significant histological disease activity) to grade 4 (severely active disease). The intraclass correlation coefficient (ICC) for the intrareader reliability was 0.88 (95% CI 0.82 to 0.92) and the index had good inter-reader reliability (ICC=0.86 (0.81 to 0.99)). The correlation between the Nancy index and the Geboes score or the GVE was very good. The index had a good responsiveness with a high correlation between changes in the Geboes score and changes in the Nancy index (0.910 (0.813 to 0.955)). Conclusions A three descriptor histological index has been validated for use in clinical practice and clinical trials.
