Abstract: Insomnia and the inability to sleep affect people’s health and well-being. However, its systematic estimates of prevalence and distribution in the general population in China are still lacking. A population-based cluster sampling survey was conducted in the rural and urban areas of Hunan, China. Subjects (n = 26,851) were sampled from the general population, with a follow-up using the Pittsburgh Sleep Quality Index (PSQI) for interview to assess quality of sleep and Insomnia (PSQI score >5). While the overall prevalence of insomnia was 26.6%, and little difference was found between males (26.3%) and females (27.0%); the mean PSQI score was 4.26 (±2.67), and significant higher in females (4.32 ± 2.70) than males (4.21 ± 2.64, p = 0.003). Individuals in the rural areas tended to report a higher PSQI score (4.45 ± 2.81) than urban residents did (4.18 ± 2.60) (p < 0.001) and the estimates of prevalence of insomnia was 29.4% in the rural areas, significant higher than 25.5% in the urban areas (p < 0.001). Multiple logistic regression analysis showed that female gender, older age, higher level of education, being unmarried, living in the rural area, cigarette smoking and alcohol drinking were associated with insomnia. Our study may provide important information for general and mental health research.

Abstract: Background: The fatigue that users suffer when using steady-state visual evoked potential (SSVEP)-based brain-computer interfaces (BCIs) can cause a number of serious problems such as signal quality degradation and system performance deterioration, users’ discomfort and even risk of photosensitive epileptic seizures, posing heavy restrictions on the applications of SSVEP-based BCIs. Towards alleviating the fatigue, a fundamental step is to measure and evaluate it but most existing works adopt self-reported questionnaire methods which are subjective, offline and memory dependent. This paper proposes an objective and real-time approach based on electroencephalography (EEG) spectral analysis to evaluate the fatigue in SSVEP-based BCIs. Methods: How the EEG indices (amplitudes in δ, θ, α and β frequency bands), the selected ratio indices (θ/α and (θ + α)/β), and SSVEP properties (amplitude and signal-to-noise ratio (SNR)) changes with the increasing fatigue level are investigated through two elaborate SSVEP-based BCI experiments, one validates mainly the effectiveness and another considers more practical situations. Meanwhile, a self-reported fatigue questionnaire is used to provide a subjective reference. ANOVA is employed to test the significance of the difference between the alert state and the fatigue state for each index. Results: Consistent results are obtained in two experiments: the significant increases in α and (θ+ α)/β, as well as the decrease in θ/α are found associated with the increasing fatigue level, indicating that EEG spectral analysis can provide robust objective evaluation of the fatigue in SSVEP-based BCIs. Moreover, the results show that the amplitude and SNR of the elicited SSVEP are significantly affected by users’ fatigue. Conclusions: The experiment results demonstrate the feasibility and effectiveness of the proposed method as an objective and real-time evaluation of the fatigue in SSVEP-based BCIs. This method would be helpful in understanding the fatigue problem and optimizing the system design to alleviate the fatigue in SSVEP-based BCIs.

Abstract: Berberine (BBR) is a renowned natural compound that exhibits potent neuroprotective activities. However, the cellular and molecular mechanisms are still unclear. Hormesis is an adaptive mechanism generally activated by mild oxidative stress to protect the cells from further damage. Many phytochemicals have been shown to induce hormesis. This study aims to investigate whether the neuroprotective activity of BBR is mediated by hormesis and the related signaling pathways in 6-OHDA-induced PC12 cells and zebrafish neurotoxic models. Our results demonstrated that BBR induced a typical hormetic response in PC12 cells, i.e. low dose BBR significantly increased the cell viability, while high dose BBR inhibited the cell viability. Moreover, low dose BBR protected the PC12 cells from 6-OHDA-induced cytotoxicity and apoptosis, whereas relatively high dose BBR did not show neuroprotective activity. The hormetic and neuroprotective effects of BBR were confirmed to be mediated by up-regulated PI3K/AKT/Bcl-2 cell survival and Nrf2/HO-1 antioxidative signaling pathways. In addition, low dose BBR markedly mitigated the 6-OHDA-induced dopaminergic neuron loss and behavior movement deficiency in zebrafish, while high dose BBR only slightly exhibited neuroprotective activities. These results strongly suggested that the neuroprotection of BBR were attributable to the hormetic mechanisms via activating cell survival and antioxidative signaling pathways.
