Abstract: Bone marrow (BM) metastasis of breast cancer (BC) can recur even decades after initial diagnosis and treatment, implying the long-term survival of disseminated cancer cells in a dormant state. Here we investigated the role of microRNAs (miRNA) transmitted from BM stroma to BC cells via gap junctions and exosomes in tumor cell quiescence. MDA-MB-231 and T47D BC cells arrest in G(0) phase of the cell cycle when cocultured with BM stroma. Analyses of miRNA expression profiles identified numerous miRNAs implicated in cell proliferation including miR-127, -197, -222, and -223 targeting CXCL12. Subsequently, we showed that these CXCL12-specific miRNAs are transported from BM stroma to BC cells via gap junctions, leading to reduced CXCL12 levels and decreased proliferation. Stroma-derived exosomes containing miRNAs also contributed to BC cell quiescence, although to a lesser degree than miRNAs transmitted via gap junctions. This study shows that the transfer of miRNAs from BM stroma to BC cells might play a role in the dormancy of BM metastases.

Abstract: DNA transport through the cell membrane is an essential requirement for gene therapy, which utilizes oligonucleotides and plasmid DNA. However, membrane transport of DNA is an inefficient process, and the mechanism(s) by which this process occurs is not clear. Although viral vectors are effective in gene therapy, the immune response elicited by viral proteins poses a major problem. Therefore, several laboratories are involved in the development of nonviral DNA delivery vehicles. These vehicles include polyamines, polycationic lipids, and neutral polymers, capable of condensing DNA to nanoparticles with radii of 20−100 nm. Although the structural and energetic forces involved in DNA condensation have been studied by physical biochemists for the past 25 years, this area has experienced a resurgence of interest in recent years because of the influx of biotechnologists involved in developing gene therapy protocols to combat a variety of human diseases. Abstract: Helminths induce potent T helper 2 (TH2)-type immune responses that can mediate worm expulsion, but the role of this response in controlling the acute tissue damage caused by migrating multicellular parasites through vital tissues remains uncertain. We used a helminth infection model in which parasitic nematode larvae migrate transiently through the lung, resulting in hemorrhage and inflammation. We found that IL-17 initially contributed to inflammation and lung damage, whereas subsequent IL-4 receptor (IL-4R) signaling reduced elevations in IL-17 mRNA levels, enhanced the expression of insulin-like growth factor 1 (IGF-1) and IL-10 and stimulated the development of M2 macrophages, all of which contributed to the rapid resolution of tissue damage. These studies indicate an essential role for TH2-type immune responses in mediating acute wound healing during helminth infection.

Abstract: 113 children were seen at 1 and 6 years of age in order to examine the relationship between the quality of the early attachment relationship and later psychopathology. On the basis of scores from the Achenbach and Edelbrock Child Behavior Profile, an outcome measure of psychopathology at 6 years, the results indicated different outcomes for male and female children. For males, attachment classification at 1 year was significantly related to later psychopathology; insecurely attached males showed more psychopathology than securely attached males. No relationship between attachment and later psychopathology was observed for females. Even for males, the attachment classification only partly predicted later behavioral problems. Several other factors, including life-stress events and family demographic variables, appeared to influence the development of psychopathology. The findings suggest that although the child's attachment relationship plays an important role in the development of psychopathology, the child is neither made invulnerable by an early secure attachment nor doomed to psychopathology by an insecure attachment.
