Abstract: ▪ Abstract Among the heavy metal-binding ligands in plant cells the phytochelatins (PCs) and metallothioneins (MTs) are the best characterized. PCs and MTs are different classes of cysteine-rich, heavy metal-binding protein molecules. PCs are enzymatically synthesized peptides, whereas MTs are gene-encoded polypeptides. Recently, genes encoding the enzyme PC synthase have been identified in plants and other species while the completion of the Arabidopsis genome sequence has allowed the identification of the entire suite of MT genes in a higher plant. Recent advances in understanding the regulation of PC biosynthesis and MT gene expression and the possible roles of PCs and MTs in heavy metal detoxification and homeostasis are reviewed.

Abstract: Qualitative research aims to address questions concerned with developing an understanding of the meaning and experience dimensions of humans' lives and social worlds. Central to good qualitative research is whether the research participants' subjective meanings, actions and social contexts, as understood by them, are illuminated. This paper aims to provide beginning researchers, and those unfamiliar with qualitative research, with an orientation to the principles that inform the evaluation of the design, conduct, findings and interpretation of qualitative research. It orients the reader to two philosophical perspectives, the interpretive and critical research paradigms, which underpin both the qualitative research methodologies most often used in mental health research, and how qualitative research is evaluated. Criteria for evaluating quality are interconnected with standards for ethics in qualitative research. They include principles for good practice in the conduct of qualitative research, and for trustworthiness in the interpretation of qualitative data. The paper reviews these criteria, and discusses how they may be used to evaluate qualitative research presented in research reports. These principles also offer some guidance about the conduct of sound qualitative research for the beginner qualitative researcher.

Abstract: SUMMARY Clusters, Grids, and peer-to-peer (P2P) networks have emerged as popular paradigms for next generation parallel and distributed computing. They enable aggregation of distributed resources for solving largescale problems in science, engineering, and commerce. In Grid and P2P computing environments, the resources are usually geographically distributed in multiple administrative domains, managed and owned by different organizations with different policies, and interconnected by wide-area networks or the Internet. This introduces a number of resource management and application scheduling challenges in the domain of security, resource and policy heterogeneity, fault tolerance, continuously changing resource conditions, and politics. The resource management and scheduling systems for Grid computing need to manage resources and application execution depending on either resource consumers’ or owners’ requirements, and continuously adapt to changes in resource availability. The management of resources and scheduling of applications in such large-scale distributed systems is a complex undertaking. In order to prove the effectiveness of resource brokers and associated scheduling algorithms, their performance needs to be evaluated under different scenarios such as varying number of resources and users with different requirements. In a Grid environment, it is hard and even impossible to perform scheduler performance evaluation in a repeatable and controllable manner as resources and users are distributed across multiple organizations with their own policies. To overcome this limitation, we have developed a Java-based discrete-event Grid simulation toolkit called GridSim. The toolkit supports modeling and simulation of heterogeneous Grid resources (both time- and space-shared), users and application models. It provides primitives for creation of application tasks, mapping of tasks to resources, and their management. To demonstrate suitability of the GridSim toolkit, we have simulated a Nimrod-G

Abstract: We demonstrate simultaneous phase and amplitude extraction from a single defocused image of a homogeneous object. Subject to the assumptions explicitly stated in the derivation, the algorithm solves the twin-image problem of in-line holography and is capable of analysing data obtained using X-ray microscopy, electron microscopy, neutron microscopy or visible-light microscopy, especially as they relate to defocus and point projection methods. Our simple, robust, non-iterative and computationally efficient method is applied to data obtained using an X-ray phase contrast ultramicroscope.

Abstract: The dephasing of particle plasmons is investigated using light-scattering spectroscopy on individual gold nanoparticles. We find a drastic reduction of the plasmon dephasing rate in nanorods as compared to small nanospheres due to a suppression of interband damping. The rods studied here also show very little radiation damping, due to their small volumes. These findings imply large local-field enhancement factors and relatively high light-scattering efficiencies, making metal nanorods extremely interesting for optical applications. Comparison with theory shows that pure dephasing and interface damping give negligible contributions to the total plasmon dephasing rate.

Abstract: Perforin/granzyme-induced apoptosis is the main pathway used by cytotoxic lymphocytes to eliminate virus-infected or transformed cells. Studies in gene-disrupted mice indicate that perforin is vital for cytotoxic effector function; it has an indispensable, but undefined, role in granzyme-mediated apoptosis. Despite its vital importance, the molecular and cellular functions of perforin and the basis of perforin and granzyme synergy remain poorly understood. The purpose of this review is to evaluate critically recent findings on cytotoxic granule-mediated cell death and to assess the functional significance of postulated cell-death pathways in appropriate pathophysiological contexts, including virus infection and susceptibility to experimental or spontaneous tumorigenesis.

Abstract: Optimal filtering problems are ubiquitous in signal processing and related fields. Except for a restricted class of models, the optimal filter does not admit a closed-form expression. Particle filtering methods are a set of flexible and powerful sequential Monte Carlo methods designed to. solve the optimal filtering problem numerically. The posterior distribution of the state is approximated by a large set of Dirac-delta masses (samples/particles) that evolve randomly in time according to the dynamics of the model and the observations. The particles are interacting; thus, classical limit theorems relying on statistically independent samples do not apply. In this paper, our aim is to present a survey of convergence results on this class of methods to make them accessible to practitioners.

Abstract: The equations of motion governing the two-dimensional flow of pedestrians are derived for flows of both single and multiple pedestrian types. Two regimes of flow, a high-density (subcritical) and a low-density (supercritical) flow regimes, are possible, rather than two flow regimes for each type of pedestrian. A subcritical flow always fills the space available. However, a supercritical flow may either fill the space available or be self-confining for each type of pedestrian, depending on the boundary location. Although, the equations governing these flows are simultaneous, time-dependent, non-linear, partial differential equations, remarkably they may be made conformally mappable. The solution of these equations becomes trivial in many situations. Free streamline calculations, utilizing this property, reveal both upstream and downstream separation of the flow of pedestrians around an obstacle. Such analysis tells much about the nature of the assumptions used in various models for the flow of pedestrians. The present theory is designed for the development of general techniques to understand the motion of large crowds. However, it is also useful as a predictive tool. The behavior predicted by these equations of motion is compared with aerial observations for the Jamarat Bridge near Mecca, Saudi Arabia. It is shown that, for this important case, pedestrians, that is pilgrims, aim at achieving each immediate goal in minimum time rather than achieving all goals in overall minimum time. Typical of many examples, this case illustrated the strong dependence of path on the psychological state of the pedestrians involved. It is proposed that the flow of pedestrians over the Jamarat Bridge be improved by appropriate barrier placement, that force an effective global view of the goals.

Abstract: The accelerated development in peer-to-peer and Grid computing has positioned them as promising next-generation computing platforms. They enable the creation of virtual enterprises for sharing resources distributed across the world. However, resource management, application development and usage models in these environments is a complex undertaking. This is due to the geographic distribution of resources that are owned by different organizations or peers. The resource owners of each of these resources have different usage or access policies and cost models, and varying loads and availability. In order to address complex resource management issues, we have proposed a computational economy framework for resource allocation and for regulating supply and demand in Grid computing environments. This framework provides mechanisms for optimizing resource provider and consumer objective functions through trading and brokering services. In a real world market, there exist various economic models for setting the price of services based on supply-and-demand and their value to the user. They include commodity market, posted price, tender and auction models. In this paper, we discuss the use of these models for interaction between Grid components to decide resource service value, and the necessary infrastructure to realize each model. In addition to usual services offered by Grid computing systems, we need an infrastructure to support interaction protocols, allocation mechanisms, currency, secure banking and enforcement services. We briefly discuss existing technologies that provide some of these services and show their usage in developing the Nimrod-G grid resource broker. Furthermore, we demonstrate the effectiveness of some of the economic models in resource trading and scheduling using the Nimrod/G resource broker, with deadline and cost constrained scheduling for two different optimization strategies, on the World-Wide Grid testbed that has resources distributed across five continents.

Abstract: Alcohol and tobacco consumption are closely correlated and published results on their association with breast cancer have not always allowed adequately for confounding between these exposures. Over 80% of the relevant information worldwide on alcohol and tobacco consumption and breast cancer were collated, checked and analysed centrally. Analyses included 58515 women with invasive breast cancer and 95067 controls from 53 studies. Relative risks of breast cancer were estimated, after stratifying by study, age, parity and, where appropriate, women's age when their first child was born and consumption of alcohol and tobacco. The average consumption of alcohol reported by controls from developed countries was 6.0 g per day, i.e. about half a unit/drink of alcohol per day, and was greater in ever-smokers than never-smokers, (8.4 g per day and 5.0 g per day, respectively). Compared with women who reported drinking no alcohol, the relative risk of breast cancer was 1.32 (1.19 - 1.45, P < 0.00001) for an intake of 35 - 44 g per day alcohol, and 1.46 (1.33 - 1.61, P < 0.00001) for greater than or equal to 45 g per day alcohol. The relative risk of breast cancer increased by 7.1% (95% CI 5.5-8.7%; P<0.00001) for each additional 10 g per day intake of alcohol, i.e. for each extra unit or drink of alcohol consumed on a daily basis. This increase was the same in ever-smokers and never-smokers (7.1 % per 10 g per day, P < 0.00001, in each group). By contrast, the relationship between smoking and breast cancer was substantially confounded by the effect of alcohol. When analyses were restricted to 22 255 women with breast cancer and 40 832 controls who reported drinking no alcohol, smoking was not associated with breast cancer (compared to never-smokers, relative risk for ever-smokers= 1.03, 95% CI 0.98 - 1.07, and for current smokers=0.99, 0.92 - 1.05). The results for alcohol and for tobacco did not vary substantially across studies, study designs, or according to 15 personal characteristics of the women; nor were the findings materially confounded by any of these factors. If the observed relationship for alcohol is causal, these results suggest that about 4% of the breast cancers in developed countries are attributable to alcohol. In developing countries, where alcohol consumption among controls averaged only 0.4 g per day, alcohol would have a negligible effect on the incidence of breast cancer. In conclusion, smoking has little or no independent effect on the risk of developing breast cancer; the effect of alcohol on breast cancer needs to be interpreted in the context of its beneficial effects, in moderation, on cardiovascular disease and its harmful effects on cirrhosis and cancers of the mouth, larynx, oesophagus and liver. Abstract: G protein-coupled receptors (GPCRs) represent the largest family of cell-surface receptors. These receptors are natural allosteric proteins because agonist-mediated signaling by GPCRs requires a conformational change in the receptor protein transmitted between two topographically distinct binding sites, one for the agonist and another for the G protein. It is now becoming increasingly recognized, however, that the agonist-bound GPCR can also form ternary complexes with other ligands or “accessory” proteins and display altered binding and/or signaling properties in relation to the binary agonist-receptor complex. Allosteric sites on GPCRs represent novel drug targets because allosteric modulators possess a number of theoretical advantages over classic orthosteric ligands, such as a ceiling level to the allosteric effect and a potential for greater GPCR subtype-selectivity. Because of the noncompetitive nature of allosteric phenomena, the detection and quantification of such effects often relies on a combination of equilibrium binding, nonequilibrium kinetic, and functional signaling assays. This review discusses the development and properties of allosteric receptor models for GPCRs and the detection and quantification of allosteric effects. Moreover, we provide an overview of the current knowledge regarding the location of possible allosteric sites on GPCRs and candidate endogenous allosteric modulators. Finally, we discuss the potential for allosteric effects arising from the formation of GPCR oligomers or GPCRs complexed with accessory cellular proteins. It is proposed that the study of allosteric phenomena will become of progressively greater import to the drug discovery process due to the advent of newer and more sensitive GPCR screening technologies.

Abstract: This study investigated the nature of dyadic interaction in an adult ESL classroom. The study was longitudinal, classroom based, and examined the nature of interaction between 10 pairs of adult ESL students over a range of language tasks and over time (a semester). Four distinct patterns of dyadic interaction were found. These patterns are distinguishable in terms of equality and mutuality (Damon & Phelps, 1989). More importantly, the findings suggest that certain patterns of dyadic interaction are more conducive than others to language learning. These findings are explained by reference to Vygotsky’s theory of cognitive development.

Abstract: We argue that collaboration can act as a source of change in institutional fields through the generation of “proto-institutions”: new practices, rules, and technologies that transcend a particular collaborative relationship and may become new institutions if they diffuse sufficiently. A four-year study of the collaborative activities of a small nongovernmental organization in Palestine suggests that collaborations that are both highly embedded and have highly involved partners are the most likely to generate proto-institutions.

Abstract: Insecticide resistance is one of the most widespread genetic changes caused by human activity, but we still understand little about the origins and spread of resistant alleles in global populations of insects. Here, via microarray analysis of all P450s in Drosophila melanogaster, we show that DDT-R, a gene conferring resistance to DDT, is associated with overtranscription of a single cytochrome P450 gene, Cyp6g1. Transgenic analysis of Cyp6g1 shows that overtranscription of this gene alone is both necessary and sufficient for resistance. Resistance and up-regulation in Drosophila populations are associated with a single Cyp6g1 allele that has spread globally. This allele is characterized by the insertion of an Accord transposable element into the 5' end of the Cyp6g1 gene.

Abstract: . This paper focuses on the benefits that organizations may achieve from their investment in enterprise systems (ES). It proposes an ES benefit framework for summarizing benefits in the years after ES implementation. Based on an analysis of the features of enterprise systems, on the literature on information technology (IT) value, on data from 233 enterprise systems vendor-reported stories published on the Web and on interviews with managers of 34 organizations using ES, the framework provides a detailed list of benefits that have reportedly been acquired through ES implementation. This list of benefits is consolidated into five benefits dimensions: operational, managerial, strategic, IT infrastructure and organizational, and illustrated using perceived net benefit flow (PNBF) graphs. In a detailed example, the paper shows how the framework has been applied to the identification of benefits in a longitudinal case study of four organizations.

Abstract: Uncertainty is pervasive in ecology where the difficulties of dealing with sources of uncertainty are exacerbated by variation in the system itself. Attempts at classifying uncertainty in ecology have, for the most part, focused exclusively on epistemic uncertainty. In this paper we classify uncertainty into two main categories: epistemic uncertainty (uncertainty in determinate facts) and linguistic uncertainty (uncertainty in language). We provide a classification of sources of uncertainty under the two main categories and demonstrate how each impacts on applications in ecology and conservation biology. In particular, we demonstrate the importance of recognizing the effect of linguistic uncertainty, in addition to epistemic uncertainty, in ecological applications. The significance to ecology and conservation biology of developing a clear understanding of the various types of uncertainty, how they arise and how they might best be dealt with is highlighted. Finally, we discuss the various general strategies for dealing with each type of uncertainty and offer suggestions for treating compounding uncertainty from a range of sources.

Abstract: The culturability of bacteria in the bulk soil of an Australian pasture was investigated by using nutrient broth at 1/100 of its normal concentration (dilute nutrient broth [DNB]) as the growth medium. Three-tube most-probable-number serial dilution culture resulted in a mean viable count that was only 1.4% of the mean microscopically determined total cell count. Plate counts with DNB solidified with agar and with gellan gum resulted in viable counts that were 5.2 and 7.5% of the mean microscopically determined total cell count, respectively. Prior homogenization of the soil sample with an ultrasonic probe increased the viable count obtained by using DNB solidified with gellan gum to 14.1% of the mean microscopically determined cell count. A microscopic examination of the cell aggregates that remained after sonication revealed that the potential CFU count was only 70.4% of the total cell count, due to cells occurring as pairs or in clumps of three or more cells. Staining with SYTO 9 plus propidium iodide indicated that 91.3% of the cells in sonicated soil samples were potentially viable. Together, these findings suggest that the maximum achievable CFU count may be as low as 64.3% of the total cell count. Thirty isolates obtained from plate counting experiments performed with DNB as the growth medium were identified by comparative analysis of partial 16S rRNA gene sequences. A large proportion of these isolates represent the first known isolates of globally distributed groups of soil bacteria belonging to novel lineages within the divisions Actinobacteria, Acidobacteria, Proteobacteria, and Verrucomicrobia.

Abstract: Cell-surface receptors are the targets for more than 60% of current drugs. Traditionally, optimizing the interaction of lead molecules with the binding site for the endogenous agonist (orthosteric site) has been viewed as the best means of achieving selectivity of action. However, recent developments have highlighted the fact that drugs can interact with binding sites on the receptor molecule that are distinct from the orthosteric site, known as allosteric sites. Allosteric modulators could offer several advantages over orthosteric ligands, including greater selectivity and saturability of their effect.

Abstract: The active left-slip Altyn Tagh fault defines the northern edge of the Tibetan plateau. To determine its deformation history we conducted integrated research on Cenozoic stratigraphic sections in the southern part of the Tarim Basin. Fission-track ages of detrital apatites, existing biostratigraphic data, and magnetostratigraphic analysis were used to establish chronostratigraphy, whereas composition of sandstone and coarse clastic sedimentary rocks was used to determine the unroofing history of the source region. Much of the detrital grains in our measured sections can be correlated with uplifted sides of major thrusts or transpressional faults, implying a temporal link between sedimentation and deformation. The results of our studies, together with existing stratigraphic data from the Qaidam Basin and the Hexi Corridor, suggest that crustal thickening in northern Tibet began prior to 46 Ma for the western Kunlun Shan thrust belt, at ca. 49 Ma for the Qimen Tagh and North Qaidam thrust systems bounding the north and south margins of the Qaidam Basin, and prior to ca. 33 Ma for the Nan Shan thrust belt. These ages suggest that deformation front reached northern Tibet only ∼10 ± 5 m.y. after the initial collision of India with Asia at 65–55 Ma. Because the aforementioned thrust systems are either termination structures or branching faults of the Altyn Tagh left-slip system, the Altyn Tagh fault must have been active since ca. 49 Ma. The Altyn Tagh Range between the Tarim Basin and the Altyn Tagh fault has been a long-lived topographic high since at least the early Oligocene or possibly late Eocene. This range has shed sediments into both the Tarim and Qaidam Basins while being offset by the Altyn Tagh fault. Its continuous motion has made the range act as a sliding door, which eventually closed the outlets of westward-flowing drainages in the Qaidam Basin. This process has caused large amounts of Oligocene–Miocene sediments to be trapped in the Qaidam Basin. The estimated total slip of 470 ± 70 km and the initiation age of 49 Ma yield an average slip rate along the Altyn Tagh fault of 9 ± 2 mm/yr, remarkably similar to the rates determined by GPS (Global Positioning System) surveys. This result implies that geologic deformation rates are steady state over millions of years during continental collision.

Abstract: Fe-rich metapelitic granulites of the Musgrave Block, central Australia, contain several symplectic and coronal reaction textures that post-date a peak S2 metamorphic assemblage involving garnet, sillimanite, spinel, ilmenite, K-feldspar and quartz. The earliest reaction textures involve spinel- and quartz-bearing symplectites that enclose garnet and to a lesser extent sillimanite. The symplectic spinel and quartz are in places separated by later garnet and/or sillimanite coronas. The metamorphic effects of a later, D3, event are restricted to zones of moderate to high strain where a metamorphic assemblage of garnet, sillimanite, K-feldspar, magnetite, ilmenite, quartz and biotite is preserved. Quantitative mineral equilibria calculations in the system K2O–FeO–MgO–Al2O3–SiO2–H2O–TiO2–Fe2O3 (KFMASHTO) using Thermocalc 3.0 and the accompanying internally consistent dataset provide important constraints on the influence of TiO2 and Fe2O3 on biotite-bearing and spinel-bearing equilibria, respectively. Biotite-bearing equilibria are shifted to higher temperatures and spinel-bearing equilibria to higher pressures and lower temperatures in comparison to the equivalent equilibria in K2O–FeO–MgO–Al2O3–SiO2–H2O (KFMASH). The sequence of reaction textures involving spinel is consistent with a D2 P–T path that involved a small amount of decompression followed predominantly by cooling within a single mineral assemblage stability field. Thus, the reaction textures reflect changes in modal proportions within an equilibrium assemblage rather than the crossing of a univariant reaction. The D3 metamorphic assemblage is consistent with lower temperatures than those inferred for D2.

Abstract: OBJECTIVE: To test the effectiveness of, and explore interactions between, three interventions to prevent falls among older people. DESIGN: A randomised controlled trial with a full factorial design. SETTING: Urban community in Melbourne, Australia. PARTICIPANTS: 1090 aged 70 years and over and living at home. Most were Australian born and rated their health as good to excellent; just over half lived alone. INTERVENTIONS: Three interventions (group based exercise, home hazard management, and vision improvement) delivered to eight groups defined by the presence or absence of each intervention. MAIN OUTCOME MEASURE: Time to first fall ascertained by an 18 month falls calendar and analysed with survival analysis techniques. Changes to targeted risk factors were assessed by using measures of quadriceps strength, balance, vision, and number of hazards in the home. RESULTS: The rate ratio for exercise was 0.82 (95% confidence interval 0.70 to 0.97, P=0.02), and a significant effect (P<0.05) was observed for the combinations of interventions that involved exercise. Balance measures improved significantly among the exercise group. Neither home hazard management nor treatment of poor vision showed a significant effect. The strongest effect was observed for all three interventions combined (rate ratio 0.67 (0.51 to 0.88, P=0.004)), producing an estimated 14.0% reduction in the annual fall rate. The number of people needed to be treated to prevent one fall a year ranged from 32 for home hazard management to 7 for all three interventions combined. CONCLUSIONS: Group based exercise was the most potent single intervention tested, and the reduction in falls among this group seems to have been associated with improved balance. Falls were further reduced by the addition of home hazard management or reduced vision management, or both of these. Cost effectiveness is yet to be examined. These findings are most applicable to Australian born adults aged 70-84 years living at home who rate their health as good.
