Abstract: The National Institute of Mental Health strategic plan for advancing psychiatric neuroscience calls for an acceleration of discovery and the delineation of developmental trajectories for risk and resilience across the lifespan. To attain these objectives, sufficiently powered datasets with broad and deep phenotypic characterization, state-of-the-art neuroimaging, and genetic samples must be generated and made openly available to the scientific community. The enhanced Nathan Kline Institute Rockland Sample (NKI-RS) is a response to this need. NKI-RS is an ongoing, institutionally-centered endeavor aimed at creating a large-scale (N>1000), deeply phenotyped, community-ascertained, lifespan sample (ages 6-85 years old) with advanced neuroimaging and genetics. These data will be publically shared, openly and prospectively (i.e., on a weekly basis). Herein, we describe the conceptual basis of the NKI-RS, including study design, sampling considerations, and steps to synchronize phenotypic and neuroimaging assessment. Additionally, we describe our process for sharing the data with the scientific community while protecting participant confidentiality, maintaining an adequate database, and certifying data integrity. The pilot phase of the NKI-RS, including challenges in recruiting, characterizing, imaging, and sharing data, is discussed while also explaining how this experience informed the final design of the enhanced NKI-RS. It is our hope that familiarity with the conceptual underpinnings of the enhanced NKI-RS will facilitate harmonization with future data collection efforts aimed at advancing psychiatric neuroscience and nosology.

Abstract: BackgroundChronic obstructive pulmonary disease (COPD) is thought to result from an accelerated decline in forced expiratory volume in 1 second (FEV1) over time. Yet it is possible that a normal decline in FEV1 could also lead to COPD in persons whose maximally attained FEV1 is less than population norms. MethodsWe stratified participants in three independent cohorts (the Framingham Offspring Cohort, the Copenhagen City Heart Study, and the Lovelace Smokers Cohort) according to lung function (FEV1 ≥80% or <80% of the predicted value) at cohort inception (mean age of patients, approximately 40 years) and the presence or absence of COPD at the last study visit. We then determined the rate of decline in FEV1 over time among the participants according to their FEV1 at cohort inception and COPD status at study end. Abstract: Autophagy is a newly recognized innate defense mechanism, acting as a cell-autonomous system for elimination of intracellular pathogens. The signals and signalling pathways inducing autophagy in response to pathogen invasion are presently not known. Here we show that autophagy is controlled by recognizing conserved pathogen-associated molecular patterns (PAMPs). We screened a PAMP library for effects on autophagy in RAW 264.7 macrophages and found that several prototype Toll-like receptor (TLR) ligands induced autophagy. Single-stranded RNA and TLR7 generated the most potent effects. Induction of autophagy via TLR7 depended on MyD88 expression. Stimulation of autophagy with TLR7 ligands was functional in eliminating intracellular microbes, even when the target pathogen was normally not associated with TLR7 signalling. These findings link two innate immunity defense systems, TLR signalling and autophagy, provide a potential molecular mechanism for induction of autophagy in response to pathogen invasion, and show that the newly recognized ability of TLR ligands to stimulate autophagy can be used to treat intracellular pathogens.

Abstract: The need for sound ecological science has escalated alongside the rise of the information age and “big data” across all sectors of society. Big data generally refer to massive volumes of data not readily handled by the usual data tools and practices and present unprecedented opportunities for advancing science and inform- ing resource management through data-intensive approaches. The era of big data need not be propelled only by “big science” – the term used to describe large-scale efforts that have had mixed success in the individual-driven culture of ecology. Collectively, ecologists already have big data to bolster the scientific effort – a large volume of distributed, high-value information – but many simply fail to contribute. We encourage ecologists to join the larger scientific community in global initiatives to address major scientific and societal problems by bringing their distributed data to the table and harnessing its collective power. The scientists who contribute such information will be at the forefront of socially relevant science – but will they be ecologists?

Abstract: We present the first results from a very deep Chandra X-ray observation of the core of the Perseus cluster of galaxies. A pressure map reveals a clear thick band of high pressure around the inner radio bubbles. The gas in the band must be expanding outward and the sharp front to it is identified as a shock front, yet we see no temperature jump across it; indeed there is more soft emission behind the shock than in front of it. We conclude that in this inner region either thermal conduction operates efficiently or the co-existing relativistic plasma seen as the radio mini-halo is mediating the shock. If common, isothermal shocks in cluster cores mean that we cannot diagnose the expansion speed of radio bubbles from temperature measurements alone. They can at times expand more rapidly than currently assumed without producing significant regions of hot gas. Bubbles may also be significantly more energetic. The pressure ripples found in earlier images are identified as isothermal sound waves. A simple estimate based on their amplitude confirms that they can be an effective distributed heat source able to balance radiative cooling.We see multiphase gas with about 10{sup 9}M{sub {circle_dot}} at a temperature of about 0.5 keV. Much, but not all, of this cooler gas is spatially associated with the optical filamentary nebula around the central galaxy, NGC1275. A residual cooling flow of about 50M{sub {circle_dot}} yr{sup -1} may be taking place. A channel is found in the pressure map along the path of the bubbles, with indications found of outer bubbles. The channel connects in the S with a curious cold front.
