Abstract: Phase contrast, ultraviolet microscopy, and freeze-etching were used to determine the amount of exocellular polymer surrounding unfixed cells of four genera of bacteria: Azotobacter vinelandii, Zoogloea ramigera, Leuconostoc mesenteroides, and an acid-tolerant, floc-forming Bacillus species. Thin-sectional electron microscopy was employed to measure the effectiveness of a modified ruthenium red staining method. The results obtained with this modification of ruthenium red staining technique were compared to results obtained when previously proposed ruthenium red methods of fixation were employed. The results of these relations were then compared to the amounts of exocellular material as determined with phase-contrast microscopy, ultraviolet microscopy, and freeze-etching. The data obtained indicate that improved fixation of exocellular polymer is achieved when cells are pretreated with ruthenium red as described herein. In addition, the modified methods also reveal cytological detail not apparent when other methods of ruthenium fixation are employed.

Major fatty acids were myristic, palmitic, palimitoleic, stearic, oleic, linoleic, linolenic and arachidonic acids.

Abstract: 1. A procedure for the purification of lactate dehydrogenase (E.C. 1.1.1.27) from the tapeworm,Hymenolepis diminuta, is described. 2. The 128-fold purified enzyme exhibits a specific activity of 106 units/mg of protein and was adjudged to be homogeneous by rechromatography, sedimentation velocity and sedimentation equilibrium ultracentrifugation, and a variety of electrophoretic techniques. 3. Only one form of the enzyme is present in the tapeworm as demonstrated by electrophoresis on cellulose acetate, polyacrylamide-gel and isoelectric focusing. 4. The molecular weight of the native enzyme is 141,000 and exhibits a sedimentation coefficient of 6·89 × 10-13 sec. The enzyme is a tetramer composed of subunits of molecular weight 36,000. However, under low protein concentrations (0·1 mg/ml) on analytical gel filtration, the enzyme dissociates to the dimeric state and exhibits a molecular weight of 75,000. 5. Inhibitor studies are consistent with an active center which may be catalytically and structurally similar to the enzyme from vertebrates. 6. The enzyme is kinetically similar to vertebrate heart LDH. The Km for pyruvate was 0·17 mM and for lactate was 6·3 mM. TheH. diminuta lactate dehydrogenase also forms an abortive ternary complex with coenzyme and substrate. 7. The data are discussed in regard to the physiology of the parasite and the host.

Abstract: SYNOPSIS. Some of the cytologic features of Euglena gracilis Klebs, Euglena gracilis var. bacillaris Gross (colorless), and Astasia longa Pringsheim were compared in an electron-microscope study. E. gracilis was readily differentiated from the colorless forms by the presence of chloroplasts. The 2 colorless forms were structurally similar; however, it was possible to differentiate bleached Euglena from Astasia longa by the presence of irregular plate-like aggregates of paramylum in the former; similar forms of paramylum were not seen in chloroplast-containing E. gracilis or in A. longa.

Abstract: SUMMARY Endogenous levels of indole-3-acetic acid were mesaured in synchronous cultures of Chlorella pyrenoidosa (TX-7-11-05). The cultures were synchronized by alternating light:dark periods of 15:9 hr at a temperature of 40 ± 1 C. After 2 synchronous cycles the cultures were exposed to a low light treatment of 350 ± 100 ft-c. The time to incipient cell division under these conditions was 6 hr and 15 min. Samples were taken at 3 sampling periods during the low light treatment period:low light 0 hr (LL0); low light 3 hr (LL3); and low light 6:15 hr (LL6:15). The algal extracts were analyzed by a fluorometric procedure which measured the indole-α-pyrone product formed by the action of the trifluoracetic acid-acetic anhydride reagent with IAA. The IAA levels increased gradually from the autospore stage (5.19 μg × 10−4/mg dry wt) to the adolescent stage (7.13 μg × 10−4/mg dry wt) and more rapidly when approaching the ripened adult stage (14.55 μg × 10−4/mg dry wt). The mean percentage increase from autospore to adolescent was 36.9%, and from adolescent to ripened adult 104.6%. The total percentage increase from autospore to adult was 180.3%. Levels of IAA were 2 times higher just prior to division than in the autospore stage.

Abstract: The structure of the cell wall, the arrangement of the cyst exine, and the origin and distribution of intine vesicles in Azotobacter vinelandii ATCC 12837 were examined by freeze-etching and conventional electron microscopic techniques. In the vegetative organism the cell wall appears to have a woven texture which disappears during cyst formation. The exine is composed of two different types of material: the outer layer is a fibrous, amorphous layer, and the numerous inner layers form the basic hexagonal structures which unite to form the cyst coat. The presence of intine vesicles in the encysting organism was confirmed in frozen-etched cells. The appearance of frozen-etched cells and cysts and the distribution of capsular material indicate that extracellular polysaccharide is an important factor in cyst formation.

Abstract: Jansen and Garvey (1972) found that the Personal Orientation Inventory developed hy Shostrom (1966) differentiated between clergymen rated high and low in clinical and counseling competence by their supervisor upon completion of a program of clinical pastoral education. High-rated clergymen had mean POI scale scores which were generally above those of Shosuom's adult sample but somewhat lower than those of the self-actualized normative group. Clergymen rated low in clinical and counseling prowess had mean scale scores which tended to approximate those of Shostrom's non-self-actualized group. Webster and Stewart (1969) compared the POI scores of clergymen with those of young adults in New Zealand. Clergymen scored significantly higher than the young adults on four scales of the POI and lower on one scale. As a group, the clergymen appeared to be less able to see the opposites of life as meaningfully related. The present study compared clergymen entering clinical training with Shostrom's (1966) normal adult normative sample. Ss were 93 clergymen who completed the clinical uainlng program at Willmar State Hospiml between June, 1967, and June, 1972. Their mean age was 33 pr. The sample was comprised of 68% Roman Catholic and 32% Protestant Ss. All Ss completed the POI during the first week of clinical training. Student's t test was used to test for differences between the mean scores of the clergymen and those of the normal adult group reported in the I972 supplement to the test manual.

Abstract: Twelve right-handed and 12 left-handed boys and girls were given grip strength tests for the preferred and nonpreferred hands in the right-, left-, and symmetro-tonic neck postures. Expectations were that right-handers would differ from left-handers in response to the tonic neck reflex in relation to preferred and nonpreferred grip strength. Results supported the conclusion that the left tonic neck reflex in the right-handers tested had more facilitative and inhibitory powers than the right tonic neck reflex and accounted for the difference between right-handers and left-handers in the manifestation of dominant strength of the preferred hand.
