TL;DR: Recon 2, a community-driven, consensus 'metabolic reconstruction', is described, which is the most comprehensive representation of human metabolism that is applicable to computational modeling and has improved topological and functional features.
Abstract: Multiple models of human metabolism have been reconstructed, but each represents only a subset of our knowledge. Here we describe Recon 2, a community-driven, consensus 'metabolic reconstruction', which is the most comprehensive representation of human metabolism that is applicable to computational modeling. Compared with its predecessors, the reconstruction has improved topological and functional features, including ~2× more reactions and ~1.7× more unique metabolites. Using Recon 2 we predicted changes in metabolite biomarkers for 49 inborn errors of metabolism with 77% accuracy when compared to experimental data. Mapping metabolomic data and drug information onto Recon 2 demonstrates its potential for integrating and analyzing diverse data types. Using protein expression data, we automatically generated a compendium of 65 cell type–specific models, providing a basis for manual curation or investigation of cell-specific metabolic properties. Recon 2 will facilitate many future biomedical studies and is freely available at http://humanmetabolism.org/.

TL;DR: It is shown that zebrafish golden mutants share these melanosomal changes and that golden encodes a putative cation exchanger slc24a5 (nckx5) that localizes to an intracellular membrane, likely the melanosome or its precursor.
Abstract: Lighter variations of pigmentation in humans are associated with diminished number, size, and density of melanosomes, the pigmented organelles of melanocytes. Here we show that zebrafish golden mutants share these melanosomal changes and that golden encodes a putative cation exchanger slc24a5 (nckx5) that localizes to an intracellular membrane, likely the melanosome or its precursor. The human ortholog is highly similar in sequence and functional in zebrafish. The evolutionarily conserved ancestral allele of a human coding polymorphism predominates in African and East Asian populations. In contrast, the variant allele is nearly fixed in European populations, is associated with a substantial reduction in regional heterozygosity, and correlates with lighter skin pigmentation in admixed populations, suggesting a key role for the SLC24A5 gene in human pigmentation.

TL;DR: In this article, the authors focus on the most commonly used estimate of reliability, internal reliability, which is used for interpreting study effects and test results, and propose a method to estimate the reliability of test results.
Abstract: Although often ignored, reliability is critical when interpreting study effects and test results. Accordingly, this article focuses on the most commonly used estimate of reliability, internal consi...

TL;DR: This paper examined the effect of international experience on service firms' selection of foreign markets and entry modes and found that as their experience increases and becomes geographically more diversified, service firms tend to choose markets that are culturally less similar to their home country.
Abstract: The paper examines the effect of international experience on service firms' selection of foreign markets and entry modes. The investigation utilizes survey date from 151 United States-based service firms. Results on market selection suggest that, as their experience increases and becomes geographically more diversified, service firms tend to choose markets that are culturally less similar to their home country. On entry mode choice, the paper departs from traditional linear conceptualizations and hypothesizes a U-shaped relationship between experience and propensity for integrated entry modes. Results generally support the hypothesis. The paper explains these findings and describes how service firms resemble and differ from manufacturing firms in their foreign market entry behavior.

TL;DR: In this article, the notion of syncretic rent-seeking behavior is proposed to explain how firms generate economic rents through competition and cooperation, and a four-cell typology of rentseeking strategic behaviors is developed.
Abstract: In this article, the notion of syncretic rent-seeking behavior is proposed to explain how firms generate economic rents through competition and cooperation Previously, competition and cooperation have been viewed largely as opposite ends of a single continuum By conceptualizing competition and cooperation as distinct but interrelated dimensions, we develop a four-cell typology of rent-seeking strategic behaviors Propositions linking syncretic rent-seeking behavior, organizational competencies, and business performance are offered to guide future theory development and research efforts
