Abstract: Researchers worldwide with information about the Kirsten ras (Ki-ras) tumour genotype and outcome of patients with colorectal cancer were invited to provide that data in a schematized format for inclusion in a collaborative database called RASCAL (The Kirsten ras incolorectal-cancer collaborative group). Our results from 2721 such patients have been presented previously and for the first time in any common cancer, showed conclusively that different gene mutations have different impacts on outcome, even when the mutations occur at the same site on the genome. To explore the effect of Ki-ras mutations at different stages of colorectal cancer, more patients were recruited to the database, which was reanalysed when information on 4268 patients from 42 centres in 21 countries had been entered. After predetermined exclusion criteria were applied, data on 3439 patients were entered into a multivariate analysis. This found that of the 12 possible mutations on codons 12 and 13 of Kirsten ras, only one mutation on codon 12, glycine to valine, found in 8.6% of all patients, had a statistically significant impact on failure-free survival (P=0.004, HR 1.3) and overall survival (P=0.008, HR 1.29). This mutation appeared to have a greater impact on outcome in Dukes' C cancers (failure-free survival, P=0.008, HR 1.5, overall survival P=0.02, HR 1.45) than in Dukes' B tumours (failure-free survival, P=0.46, HR 1.12, overall survival P=0.36, HR 1.15). Ki-ras mutations may occur early in the development of pre-cancerous adenomas in the colon and rectum. However, this collaborative study suggests that not only is the presence of a codon 12 glycine to valine mutation important for cancer progression but also that it may predispose to more aggressive biological behaviour in patients with advanced colorectal cancer. ⌐ 2001 Cancer Research Campaign.

Abstract: Electron spin resonance spectroscopy (ESR), Fourier transform infrared spectroscopy (FTIR), and monitoring of pyridine (py) and boric acid trimethyl ester (BATE) adsorption for determining surface acidity and basicity, respectively, were used to carry out further characterization of mixed ZnO/TiO2 polycrystalline solids prepared by different methods. Moreover, the powders were tested in a batch photoreactor for a probe reaction, i.e., 4-nitrophenol photodegradation in aqueous medium. ESR results indicated the presence of signals attributable to Zn+ species in ZnO/TiO2 (anatase) solids, while in ZnO/TiO2 (rutile) samples the presence of zinc induced only the formation of signals probably due to Ti3+ centers. FT-IR spectra showed no significant differences of the surface hydroxylation degree of the various photocatalysts, whereas the surface acidic properties generally decrease by increasing the amount of ZnO. Abstract: The photocatalytic degradation of the anthraquinonic dye Acid Blue 80 in aqueous solutions containing TiO2 dispersions has been investigated. The process has been monitored by following either the disappearance of the dye (via HPLC) and the formation of its end-products (via IC, GC, and TOC analysis). Although a relatively fast decolorization of the solutions has been observed, the mineralization is slower, and the presence of residual organic compounds was evidenced even after long term irradiation, confirming the relevant stability of anthraquinone derivatives. The identification of various unstable intermedi ates formed after low irradiation times was performed by HPLC-MS, allowing us to give insight into the early steps of the degradation process which mainly involve C−N bonds breaking and substrate hydroxylation. Complete and relatively fast mineralization of the substrate was achieved by irradiating the semiconductor dispersions in the presence of added K2S2O8.

Abstract: SUMMARY New seismic reflection profiles of the Italian deep crust project CROP provide new insights on the structure of the Ionian sea. In spite of the Apennines and Hellenides Neogene subduction zones, two conjugate passive continental margins are preserved at the margins of the Ionian sea, along the Malta escarpment to the southwest and the Apulian escarpment to the northeast. The Ionian sea is likely to be a remnant of the Mesozoic Tethys Ocean, confined by these two conjugate passive continental margins. The transition from continental to oceanic crust appears sharper to the northeast than to the southwest. The basin between southeast Sicily and southwest Puglia was about 330 km wide and suggests a low spreading rate. The inferred oceanic ridge should have been flattened by thermal cooling and buried by later sediments. Based on stratigraphic and structural constraints to the north in the Apennines belt, the ocean continued to the northwest. This palaeogeography is supported by the seismicity of the Apennines slab underneath the southern Tyrrhenian sea, which implies downgoing oceanic lithosphere. The adjacent absence or paucity of deep seismicity does not imply absence of subduction, but rather it can be interpreted as due to the more ductile behaviour of the subducted continental lithosphere. Surprisingly, we note that where the oceanic inherited basin is subducting underneath the Apennines, in the hangingwall of the subduction hinge there are outcropping slices of continental crystalline basement previously deformed by the Alpine orogen.

Abstract: The tropical green alga Caulerpa taxifolia in the Mediterranean has spread steadily since its introduction in 1984. At the end of 2000, approximately 131 km2 of benthos had been colonized in 103 independent areas along 191 km of coastline in six countries (Spain, France, Monaco, Italy, Croatia and Tunisia). Large regions neighboring the invaded areas appear favorable to further colonization, and there is thus no reason to believe that spreading will slow down in the years to come.

Abstract: Polycrystalline ZnO/TiO2 solids have been prepared with four different methods using home prepared TiO2 (anatase) or TiO2 (rutile) as supports and Zn(NO3)2·6H2O or Zn(CH3COO)2·2H2O as precursors for ZnO. The bulk and surface properties of the samples were investigated by means of TG-DTA, XRD, TEM, SEM-EDAX, XPS, BET surface area determination, and porosity measurements. XRD and TEM results indicate that no significant defect structures exist in any of the samples. The ZnO crystallinity and its enrichment on the surface of TiO2 particles were dependent on the preparation method. The surface areas generally decrease by increasing the amount of ZnO except when ZnO from Zn(CH3COO)2·2H2O was supported on TiO2 (rutile). The samples prepared from Zn(CH3COO)2·2H2O were more porous than those prepared from Zn(NO3)2·6H2O. This was confirmed by BET surface area determinations and SEM observations. Abstract: Contribution of Vgamma9/Vdelta2 T lymphocytes to immune protection against Mycobacterium tuberculosis is still a matter of debate. It was reported earlier that Vgamma9/Vdelta2 T lymphocytes kill macrophages harboring live M. tuberculosis through a granule-dependent mechanism that results in killing of intracellular bacilli. This study found that Vgamma9/Vdelta2 T lymphocytes reduce the viability of both extracellular and intracellular M. tuberculosis. Granulysin and perforin, both detected in Vgamma9/Vdelta2 T lymphocytes, play a major role, which indicates that Vgamma9/Vdelta2 T lymphocytes directly contribute to a protective host response against M. tuberculosis infection.

Abstract: Photocatalytic reactions by transition-metal (V, Cr, Fe, Co, Cu, Mo, or W) loaded TiO2 (M-TiO2) powders suspended in aqueous solutions of methanol, (S)-lysine (Lys), or acetic acid were investigated. The photoactivities of various samples were compared with the rate constant (kr) of recombination of photoexcited electrons and positive holes determined by femtosecond pump–probe diffuse reflection spectroscopy (PP-DRS). As a general trend, increased loading decreased the rate of formation of the main products (H2 , pipecolinic acid (PCA), and CO2) under UV (>300 nm) irradiation, and the effect became more intense on increasing the loading. In PP-DRS, these M-TiO2 gave similar decays of absorption at 620 nm arising from excitation by a 310 nm pulse (<100 fs). The second-order rate constant (kr) markedly increased with loading, even at a low level (0.3%) and further increased with an increase in loading up to 5%. The photocatalytic activity of platinized M-TiO2 for H2 and PCA production under deaerated conditions depended strongly on kr, but the relation between kr and the rate of CO2 production by unplatinized M-TiO2 under aerated conditions was ambiguous; other factor(s) might control the rate of the latter. These different kr dependences of photoactivity on the reaction kinetics governed by e−–h+ recombination were attributed to the presence of O2 and Pt deposits. A simple kinetic model to explain the overall rate of these photocatalytic reactions is proposed, and the effect of recombination kinetics on photoactivity is discussed.

We analytically obtain the condition for the noise enhanced stability effect and verify it by numerical simulations.

Abstract: This study aimed to investigate whether transcranial magnetic stimulation (TMS) can induce selective working memory (WM) deficits of visual-object versus visual-spatial information in normal humans. Thirty-five healthy subjects performed two computerized visual n-back tasks, in which they were required to memorize spatial locations or abstract patterns. In a first series of experiments, unilateral or bilateral TMS was delivered on posterior parietal and middle temporal regions of both hemispheres after various delays during the WM task. Bilateral temporal TMS increased reaction times (RTs) in the visual-object, whereas bilateral parietal TMS selectively increased RTs in the visual-spatial WM task. These effects were evident at a delay of 300 ms. Response accuracy was not affected by bilateral or unilateral TMS of either cortical region. In a second group of experiments, bilateral TMS was applied over the superior frontal gyrus (SFG) or the dorsolateral prefrontal cortex (DLPFC). TMS of the SFG selectively increased RTs in the visual-spatial WM task, whereas TMS of the DLPFC interfered with both WM tasks, in terms of both accuracy and RTs. These effects were evident when TMS was applied after a delay of 600 ms, but not one of 300 ms. These findings confirm the segregation of WM buffers for object and spatial information in the posterior cortical regions. In the frontal cortex, the DLPFC appears to be necessary for WM computations regardless of the stimulus material.

Abstract: Up-regulation of proapoptotic genes has been reported in heart failure and myocardial infarction. To determine whether caspase genes can affect cardiac function, a transgenic mouse was generated. Cardiac tissue-specific overexpression of the proapoptotic gene Caspase3 was induced by using the rat promoter of alpha-myosin heavy chain, a model that may represent a unique tool for investigating new molecules and antiapoptotic therapeutic strategies. Cardiac-specific Caspase3 expression induced transient depression of cardiac function and abnormal nuclear and myofibrillar ultrastructural damage. When subjected to myocardial ischemia-reperfusion injury, Caspase3 transgenic mice showed increased infarct size and a pronounced susceptibility to die. In this report, we document an unexpected property of the proapoptotic gene caspase3 on cardiac contractility. Despite inducing ultrastructural damage, Caspase3 does not trigger a full apoptotic response in the cardiomyocyte. We also implicate Caspase3 in determining myocardial infarct size after ischemia-reperfusion injury, because its cardiomyocyte-specific overexpression increases infarct size.

Abstract: Background The cause and pathogenesis of chronic urticaria are still poorly understood. IgE-independent reactions, are common in adult patients with chronic urticaria, who have daily spontaneous occurrence of weals. H1-receptor antagonists (antihistamines) are the major class of therapeutic agents used in the management of urticaria and angioedema. Nevertheless, chronic urticaria is often difficult to treat and may not be controlled by antihistamines alone. It has been postulated that mediators other than histamine, such as kinins, prostaglandin and leukotrienes, may be responsible for some of the symptoms in urticaria which are not controlled by antihistamines. In this study, which was randomized double-blind, placebo-controlled, we compare the clinical efficacy and safety of montelukast (MT) 10 mg given once a day and cetirizine (CET) 10 mg given once a day with placebo (PLA), in the treatment of patients with chronic urticaria who have positive challenge to acetylsalicylic acid (ASA) and/or food additives. Patients and methods A group of 51 patients, ranging in age from 15 to 71 years, with chronic urticaria and positive challenge to food additives and/or ASA, participated in this study for a period of 4 weeks, starting from a 3-day run-in. The assessment of the efficacy was based on scores of daily urticaria symptoms. Results MT significantly increased the percentage of symptom-free days for hive and itch. Analysis of frequency distribution of urticaria scores for each symptom gave similar results (MT vs. CET and MT vs. PLA, P < 0.001). The interference with sleep due to their skin condition was also lower in the group treated with MT (P < 0.001). In addition, the median number of days without the rescue medication was significantly higher in the MT group (24 days) than both the CET and the PLA groups (18 days, P < 0.001, and 20 days, P < 0.001, respectively). Finally, a low incidence of adverse events was observed in this study. Conclusion The results of this comparative study demonstrate that montelukast orally administered once a day is very effective for the treatment of cutaneous symptoms in patients with chronic urticaria due to food additives and/or ASA.

Abstract: We present the broadband X-ray spectrum of Cyg X-1 in the low/hard state as observed by the instruments on board BeppoSAX. The spectrum spans from 0.1 to 200 keV, allowing the total accretion luminosity to be observed rather than extrapolated, corresponding to ~2% of the Eddington limit for a 10 M☉ black hole. The broad bandpass allows us to determine the continuum shape with great accuracy. Simple models of Compton upscattering of seed photons from the accretion disk do not adequately match the spectrum. At low energies an additional continuum component is required, giving a complex soft excess which extends up to ~4 keV, in line with previous results from ASCA. Moreover, we clearly detect a reflected component from the accretion disk that is smeared, probably because of relativistic and Doppler effects. The reflecting material is not strongly ionized and does not subtend a large solid angle as seen from the corona (Ω/2π ~ 0.1-0.3). The estimated inner radius of the disk, which depends on the inclination of the system, is most probably between 10 and 70 gravitational radii (Rg). An unsmeared reprocessed component, probably originating from the companion star or the outer disk, could also be present. In this case, the inner radius of the disk, as inferred from the smeared reflection, is smaller, between 6Rg and 20Rg.

Abstract: We present the results of a BeppoSAX observation of the Z source GX 349+2 covering the energy range 0.1-200 keV. The presence of flares in the light curve indicates that the source was in the flaring branch during the BeppoSAX observation. We accumulated energy spectra separately for the nonflaring intervals and for the flares. In both cases, the continuum is well described by a soft blackbody (kTBB ~ 0.5 keV) and a Comptonized spectrum corresponding to electron temperature kTe ~ 2.7 keV, optical depth τ ~ 10 (for a spherical geometry), and seed-photon temperature kTW ~ 1 keV. All temperatures tend to increase during the flares. In the nonflaring emission, a hard tail dominates the spectrum above 30 keV. This can be fit by a power law with photon index ~2, contributing ~2% of the total source luminosity over the BeppoSAX energy range. A comparison with hard tails that are detected in some soft states of black hole binaries suggests that a similar mechanism could originate these components in black hole and neutron star systems.

Abstract: 1. Marine fouling affects most man-made surfaces temporarily or permanently immersed in the sea, resulting in significant (or substantial) economic costs. Intense research is aimed at preventing or reducing fouling. 2. The most widespread solution to avoid fouling formation is to make surfaces unsuitable for settlers, coating them with antifouling (AF) paints containing toxic compounds. Most AF agents (e.g. tributyltin, (TBT)) have undesirable effects on non-target species, including commercially important organisms. 3. To date, the use of TBT in AF paints has been restricted (but not prohibited) in a number of countries and new biocides are in use. 4. The environmental problems posed to marine systems by AF technologies are here briefly reviewed. 5. New approaches focusing on alternatives to the use of biocidal AF paints are also considered and discussed. Copyright © 2001 John Wiley & Sons, Ltd.
