Abstract: Fetal loss in animals and humans is frequently associated with inflammatory conditions. D6 is a promiscuous chemokine receptor with decoy function, expressed in lymphatic endothelium, that recognizes and targets to degradation most inflammatory CC chemokines. Here, we report that D6 is expressed in placenta on invading extravillous trophoblasts and on the apical side of syncytiotrophoblast cells, at the very interface between maternal blood and fetus. Exposure of D6−/− pregnant mice to LPS or antiphospholipid autoantibodies results in higher levels of inflammatory CC chemokines and increased leukocyte infiltrate in placenta, causing an increased rate of fetal loss, which is prevented by blocking inflammatory chemokines. Thus, the promiscuous decoy receptor for inflammatory CC chemokines D6 plays a nonredundant role in the protection against fetal loss caused by systemic inflammation and antiphospholipid antibodies.

Abstract: Impairment of ribosomal biogenesis can activate the p53 protein independently of DNA damage. The ability of ribosomal proteins L5, L11, L23, L26, or S7 to bind Mdm2 and inhibit its ubiquitin ligase activity has been suggested as a critical step in p53 activation under these conditions. Here, we report that L5 and L11 are particularly important for this response. Whereas several other newly synthesized ribosomal proteins are degraded by proteasomes upon inhibition of Pol I activity by actinomycin D, L5 and L11 accumulate in the ribosome-free fraction where they bind to Mdm2. This selective accumulation of free L5 and L11 is due to their mutual protection from proteasomal degradation. Furthermore, the endogenous, newly synthesized L5 and L11 continue to be imported into nucleoli even after nucleolar disruption and colocalize with Mdm2, p53, and promyelocytic leukemia protein. This suggests that the disrupted nucleoli may provide a platform for L5- and L11-dependent p53 activation, implying a role for the nucleolus in p53 activation by ribosomal biogenesis stress. These findings may have important implications with respect to understanding the pathogenesis of diseases caused by impaired ribosome biogenesis.

Abstract: In well-being research the term happiness is often used as synonymous with life satisfaction. However, little is known about lay people’s understanding of happiness. Building on the available literature, this study explored lay definitions of happiness across nations and cultural dimensions, analyzing their components and relationship with participants’ demographic features. Participants were 2799 adults (age range=30-60, 50% women) living in urban areas of Argentina, Brazil, Croatia, Hungary, India, Italy, Mexico, New Zealand, Norway, Portugal, South Africa and United States. They completed the Eudaimonic and Hedonic Happiness Investigation (EHHI), reporting, among other information, their own definition of happiness. Answers comprised definitions referring to a broad range of life domains, covering both the contextual-social sphere and the psychological sphere. Across countries and with little variation by age and gender, inner harmony predominated among psychological definitions, and family and social relationships among contextual definitions. Whereas relationships are widely acknowledged as basic happiness components, inner harmony is substantially neglected. Nevertheless, its cross-national primacy, together with relations, is consistent with the view of an ontological interconnectedness characterizing living systems, shared by several conceptual frameworks across disciplines and cultures. At the methodological level, these findings suggest the potential of a bottom-up, mixed method approach to contextualize psychological dimensions within culture and lay understanding.

Abstract: Compact autonomous power sources are one of the prerequisites for the development of wireless sensor networks. In this work vibration energy harvesting via piezoelectric resonant bimorph beams is studied. The available analytical approaches for the modeling of the coupled electromechanical behavior are critically evaluated and compared with a finite element (FEM) numerical model. The latter is applied to analyze thoroughly the stress and strain states, as well as to evaluate the resulting voltage and charge distributions in the piezoelectric layers. The aim of increasing the specific power generated per unit of scavenger volume is pursued by optimizing the shape of the scavengers. Two optimized trapezoidal configurations are hence identified and analyzed. An experimental set-up for the validation of the proposed numerical model and of the obtained optimized structures is developed. Results of a preliminary experimental assessment, confirming the improved performances of optimized scavengers, are finally given.
