Abstract: This paper proposes a regression model where the response is beta distributed using a parameterization of the beta law that is indexed by mean and dispersion parameters. The proposed model is useful for situations where the variable of interest is continuous and restricted to the interval (0, 1) and is related to other variables through a regression structure. The regression parameters of the beta regression model are interpretable in terms of the mean of the response and, when the logit link is used, of an odds ratio, unlike the parameters of a linear regression that employs a transformed response. Estimation is performed by maximum likelihood. We provide closed-form expressions for the score function, for Fisher's information matrix and its inverse. Hypothesis testing is performed using approximations obtained from the asymptotic normality of the maximum likelihood estimator. Some diagnostic measures are introduced. Finally, practical applications that employ real data are presented and discussed.

Abstract: Multimodality approach is the preferred treatment strategy for distal rectal cancer, including radical surgery, radiotherapy and chemotherapy. A significant proportion of patients managed by surgery, performed according to established oncological principles, appear to benefit from chemoradiation (CRT) therapy either pre- or postoperatively in terms of survival and recurrence rates. Preoperative CRT may be associated with less acute toxicity, greater tumor response/sensitivity, and higher rates of sphincter-saving procedures when compared with postoperative course.1,2 Furthermore, tumor downstaging may lead to complete clinical response (defined as absence of residual primary tumor clinically detectable) or complete pathologic response (defined as absence of viable tumor cells after full pathologic examination of the resected specimen, pT0N0M0). These situations may be observed in 10% to 30% of patients treated by neoadjuvant CRT and may be referred as stage 0 disease.3–8 Surgical resection of the rectum may be associated with significant morbidity and mortality, and in these patients, with significant rates of stoma construction.9 Moreover, surgical resection may not lead to increased overall and disease-free survival in these patients. For this reason, it has been our policy to carefully follow these patients with complete clinical response assessed after 8 weeks of CRT completion by clinical, endoscopic, and radiologic studies without immediate surgery. Patients considered with incomplete clinical response are referred to radical surgery. Surprisingly, up to 7% of these patients may present complete pathologic response (pT0N0M0) without tumor cells during pathologic examination, despite incomplete clinical response characterized by a residual rectal ulcer.8 To determine the benefit of surgical resection in patients with stage 0 rectal cancer treated by preoperative CRT therapy, we compared long-term results of a group of patients with incomplete clinical response followed by radical surgery versus a group of patients with complete clinical response not operated on.

Abstract: Heavy smoke from forest fires in the Amazon was observed to reduce cloud droplet size and so delay the onset of precipitation from 1.5 kilometers above cloud base in pristine clouds to more than 5 kilometers in polluted clouds and more than 7 kilometers in pyro-clouds. Suppression of low-level rainout and aerosol washout allows transport of water and smoke to upper levels,where the clouds appear “smoking” as they detrain much of the pollution. Elevating the onset of precipitation allows invigoration of the updrafts,causing intense thunderstorms,large hail,and greater likelihood for overshooting cloud tops into the stratosphere. There,detrained pollutants and water vapor would have profound radiative impacts on the climate system. The invigorated storms release the latent heat higher in the atmosphere. This should substantially affect the regional and global circulation systems. Together,these processes affect the water cycle,the pollution burden of the atmosphere,and the dynamics of atmospheric circulation. Several hundred thousand deforestation and agricultural fires burn in Amazonia during the dry season each year, covering vast areas with dense smoke (1, 2). The smoke’s radiative impact suppresses surface heating and evaporation and stabilizes the lower troposphere. In turn, this suppresses the formation of convective clouds and precipitation and thus slows down the hydrological cycle (3). The microphysical effects of the aerosols on clouds and precipitation are no less important but have until now only been inferred from modeling and satellite observations. Convective clouds forming in smoky air show substantially reduced droplet size compared to that of similar clouds in clean air (4), with a mean satelliteretrieved effective droplet radius of 9 m in smoky clouds compared to 14 mi n clean clouds (5). This reduction of cloud droplet size by smoke is associated with an inhibition of the onset of precipitation radar echoes up to heights of 6.5 km, compared to 3 km in smoke-free clouds (6, 7). Abstract: Detailed organic analysis of natural aerosols from the Amazonian rain forest showed considerable quantities of previously unobserved polar organic compounds, which were identified as a mixture of two diastereoisomeric 2-methyltetrols: 2-methylthreitol and 2-methylerythritol. These polyols, which have the isoprene skeleton, can be explained by OH radical-initiated photooxidation of isoprene. They have low vapor pressure, allowing them to condense onto preexisting particles. It is estimated that photooxidation of isoprene results in an annual global production of about 2 teragrams of the polyols, a substantial fraction of the Intergovernmental Panel on Climate Change estimate of between 8 and 40 teragrams per year of secondary organic aerosol from biogenic sources.

Abstract: In the framework of the ESO Large Programme "First Stars", very high-quality spectra of some 70 very metal-poor dwarfs and giants were obtained with the ESO VLT and UVES spectrograph. These stars are likely to have descended from the first generation(s) of stars formed after the Big Bang, and their detailed composition provides constraints on issues such as the nature of the first supernovae, the efficiency of mixing processes in the early Galaxy, the formation and evolution of the halo of the Galaxy, and the possible sources of reionization of the Universe. This paper presents the abundance analysis of an homogeneous sample of 35 giants selected from the HK survey of Beers et al. (1992, 1999), emphasizing stars of extremely low metallicity: 30 of our 35 stars are in the range −4.1 < (Fe/H) < −2. 7, and 22 stars have (Fe/H) < −3.0. Our new VLT/UVES spectra, at a resolving power of R ∼ 45 000 and with signal-to-noise ratios of 100-200 per pixel over the wavelength range 330-1000 nm, are greatly superior to those of the classic studies of McWilliam et al. (1995) and Ryan et al. (1996). The immediate objective of the work is to determine precise, comprehensive, and homogeneous element abundances for this large sample of the most metal-poor giants presently known. In the analysis we combine the spectral line modeling code "Turbospectrum" with OSMARCS model atmospheres, which treat continuum scattering correctly and thus allow proper inter- pretation of the blue regions of the spectra, where scattering becomes important relative to continuous absorption ( λ< 400 nm). We obtain detailed information on the trends of elemental abundance ratios and the star-to-star scatter around those trends, en- abling us to separate the relative contributions of cosmic scatter and observational/analysis errors. Abundances of 17 elements from C to Zn have been measured in all stars, including K and Zn, which have not previously been detected in stars with (Fe/H) < −3.0. Among the key results, we discuss the oxygen abundance (from the forbidden (OI) line), the different and sometimes complex trends of the abundance ratios with metallicity, the very tight relationship between the abundances of certain elements (e.g., Fe and Cr), and the high (Zn/Fe) ratio in the most metal-poor stars. Within the error bars, the trends of the abundance ratios with metallicity are consistent with those found in earlier literature, but in many cases the scatter around the average trends is much smaller than found in earlier studies, which were limited to lower-quality spectra. We find that the cosmic scatter in several element ratios may be as low as 0.05 dex. The evolution of the abundance trends and scatter with declining metallicity provides strong constraints on the yields of the first supernovae and their mixing into the early ISM. The abundance ratios found in our sample do not match the predicted yields from pair-instability hypernovae, but are consistent with element production by super- novae with progenitor masses up to 100 M� . Abstract: A landmark review of studies published prior to 1989 on socioeconomic status (SES) and obesity supported the view that obesity in the developing world would be essentially a disease of the socioeconomic elite. The present review, on studies conducted in adult populations from developing countries, published between 1989 and 2003, shows a different scenario for the relationship between SES and obesity. Although more studies are necessary to clarify the exact nature of this relationship, particularly among men, three main conclusions emerge from the studies reviewed: 1. Obesity in the developing world can no longer be considered solely a disease of groups with higher SES. 2. The burden of obesity in each developing country tends to shift towards the groups with lower SES as the country's gross national product (GNP) increases. 3. The shift of obesity towards women with low SES apparently occurs at an earlier stage of economic development than it does for men. The crossover to higher rates of obesity among women of low SES is found at a GNP per capita of about US$ 2500, the mid-point value for lower-middle-income economies. The results of this review reinforce the urgent need to: include obesity prevention as a relevant topic on the public health agenda in developing countries; improve the access of all social classes in these countries to reliable information on the determinants and consequences of obesity; and design and implement consistent public actions on the physical, economic, and sociocultural environment that make healthier choices concerning diet and physical activity feasible for all. A significant step in this direction was taken with the approval of the Global Strategy on Diet, Physical Activity and Health by the World Health Assembly in May 2004.

Abstract: Incompletely infiltrated collagen fibrils in acid-etched dentin are susceptible to degradation. We hypothesize that degradation can occur in the absence of bacteria. Partially demineralized collagen matrices (DCMs) prepared from human dentin were stored in artificial saliva. Control specimens were stored in artificial saliva containing proteolytic enzyme inhibitors, or pure mineral oil. We retrieved them at 24 hrs, 90 and 250 days to examine the extent of degradation of DCM. In the 24-hour experimental and 90- and 250-day control specimens, we observed 5- to 6-microm-thick layers of DCM containing banded collagen fibrils. DCMs were almost completely destroyed in the 250-day experimental specimens, but not when incubated with enzyme inhibitors or mineral oil. Functional enzyme analysis of dentin powder revealed low levels of collagenolytic activity that was inhibited by protease inhibitors or 0.2% chlorhexidine. We hypothesize that collagen degradation occurred over time, via host-derived matrix metalloproteinases that are released slowly over time.

Abstract: Motor neuron diseases (MNDs) are a group of neurodegenerative disorders with involvement of upper and/or lower motor neurons, such as amyotrophic lateral sclerosis (ALS), spinal muscular atrophy (SMA), progressive bulbar palsy, and primary lateral sclerosis. Recently, we have mapped a new locus for an atypical form of ALS/MND (atypical amyotrophic lateral sclerosis [ALS8]) at 20q13.3 in a large white Brazilian family. Here, we report the finding of a novel missense mutation in the vesicle-associated membrane protein/synaptobrevin-associated membrane protein B (VAPB) gene in patients from this family. Subsequently, the same mutation was identified in patients from six additional kindreds but with different clinical courses, such as ALS8, late-onset SMA, and typical severe ALS with rapid progression. Although it was not possible to link all these families, haplotype analysis suggests a founder effect. Members of the vesicle-associated proteins are intracellular membrane proteins that can associate with microtubules and that have been shown to have a function in membrane transport. These data suggest that clinically variable MNDs may be caused by a dysfunction in intracellular membrane trafficking.

Abstract: Construction of the first stage of the Pierre Auger Observatory has begun. The aim of the Observatory is to collect unprecedented information about cosmic rays above 1018 eV. The first phase of the project, the construction and operation of a prototype system, known as the engineering array, has now been completed. It has allowed all of the sub-systems that will be used in the full instrument to be tested under field conditions. In this paper, the properties and performance of these sub-systems are described and their success illustrated with descriptions of some of the events recorded thus far. © 2003 Elsevier B.V. Abstract: The tropics are the main source of the atmosphere's sensible and latent heat, and water vapour, and are therefore important for reconstructions of past climate1. But long, accurately dated records of southern tropical palaeoclimate, which would allow the establishment of climatic connections to distant regions, have not been available. Here we present a 210,000-year (210-kyr) record of wet periods in tropical northeastern Brazil—a region that is currently semi-arid. The record is obtained from speleothems and travertine deposits that are accurately dated using the U/Th method. We find wet periods that are synchronous with periods of weak East Asian summer monsoons2, cold periods in Greenland3, Heinrich events in the North Atlantic4 and periods of decreased river runoff to the Cariaco basin5. We infer that the wet periods may be explained with a southward displacement of the Intertropical Convergence Zone. This widespread synchroneity of climate anomalies suggests a relatively rapid global reorganization of the ocean–atmosphere system. We conclude that the wet periods probably affected rainforest distribution, as plant fossils show that forest expansion occurred during these intermittent wet intervals, and opened a forest corridor6,7,8 between the Amazonian and Atlantic rainforests.

Abstract: Central neurons regenerate axons if a permissive environment is provided; after spinal cord injury, however, inhibitory molecules are present that make the local environment nonpermissive. A promising new strategy for inducing neurons to overcome inhibitory signals is to activate cAMP signaling. Here we show thatcAMP levels fall in the rostral spinal cord, sensorimotor cortex and brainstem after spinal cord contusion. Inhibition of cAMP hydrolysis by the phosphodiesterase IV inhibitor rolipram prevents this decrease and when combined with Schwann cell grafts promotes significant supraspinal and proprioceptive axon sparing and myelination. Furthermore, combining rolipram with an injection of db-cAMP near the graft not only prevents the drop in cAMP levels but increases them above those in uninjured controls. This further enhances axonal sparing and myelination, promotes growth of serotonergic fibers into and beyond grafts, and significantly improves locomotion. These findings show that cAMP levels are key for protection, growth and myelination of injured CNS axons in vivo and recovery of function.

Abstract: The characteristics of intensity, geographical location, and persistence of the South Atlantic convergence zone (SACZ) during the austral summer are investigated. Intensity and spatial features of the SACZ are identified by performing a factor analysis of structural properties of outgoing longwave radiation (OLR) data. The first two leading factors explain 65% of the total variance of structural properties and characterize the SACZ according to intensity and location (oceanic versus continental). An index is constructed based on the magnitude of the factor scores to identify intense (weak) and oceanic (continental) SACZ. The intense SACZ category is associated with negative OLR anomalies over a large area of tropical South America, extending from the western Amazon to the Atlantic Ocean. The weak SACZ category is observed with positive OLR anomalies over tropical South America and negative OLR anomalies over southeastern South America. Abstract: The centrality dependence of transverse momentum distributions and yields for ${\ensuremath{\pi}}^{\ifmmode\pm\else\textpm\fi{}},{K}^{\ifmmode\pm\else\textpm\fi{}},p$, and $\overline{p}$ in $\text{Au}+\text{Au}$ collisions at $\sqrt{{s}_{NN}}=200\phantom{\rule{0.3em}{0ex}}\text{GeV}$ at midrapidity are measured by the PHENIX experiment at the Relativistic Heavy Ion Collider. We observe a clear particle mass dependence of the shapes of transverse momentum spectra in central collisions below $\ensuremath{\sim}2\phantom{\rule{0.3em}{0ex}}\text{GeV}∕c$ in ${p}_{T}$. Both mean transverse momenta and particle yields per participant pair increase from peripheral to midcentral and saturate at the most central collisions for all particle species. We also measure particle ratios of ${\ensuremath{\pi}}^{\ensuremath{-}}∕{\ensuremath{\pi}}^{+}$, ${K}^{\ensuremath{-}}∕{K}^{+}$, $\overline{p}∕p$, $K∕\ensuremath{\pi}$, $p∕\ensuremath{\pi}$, and $\overline{p}∕\ensuremath{\pi}$ as a function of ${p}_{T}$ and collision centrality. The ratios of equal mass particle yields are independent of ${p}_{T}$ and centrality within the experimental uncertainties. In central collisions at intermediate transverse momenta $\ensuremath{\sim}1.5--4.5\phantom{\rule{0.3em}{0ex}}\text{GeV}∕c$, proton and antiproton yields constitute a significant fraction of the charged hadron production and show a scaling behavior different from that of pions.

Abstract: Background Cleft lip or palate (or the two in combination) is a common birth defect that results from a mixture of genetic and environmental factors. We searched for a specific genetic factor contributing to this complex trait by examining large numbers of affected patients and families and evaluating a specific candidate gene. Methods We identified the gene that encodes interferon regulatory factor 6 (IRF6) as a candidate gene on the basis of its involvement in an autosomal dominant form of cleft lip and palate, Van der Woude's syndrome. A single-nucleotide polymorphism in this gene results in either a valine or an isoleucine at amino acid position 274 (V274I). We carried out transmission-disequilibrium testing for V274I in 8003 individual subjects in 1968 families derived from 10 populations with ancestry in Asia, Europe, and South America, haplotype and linkage analyses, and case–control analyses, and determined the risk of cleft lip or palate that is associated with genetic variation in IRF6. Abstract: Owing to the potential role of the tick Amblyomma cooperi in the enzootic cycle of Rickettsia rickettsii, the etiologic agent of Brazilian spotted fever (BSF), this study evaluated infection by Rickettsia species in A. cooperi ticks collected from an area in Brazil where BSF is endemic. Among a total of 40 A. cooperi adult ticks collected in an area of BSF endemicity in the state of Sao Paulo, PCR analysis detected DNA of Rickettsia bellii in 16 ticks (40%), and 3 other ticks (7.5%) were positive for a previously unidentified spotted-fever-group (SFG) rickettsia. Cultivation in Vero cell cultures by the shell vial technique with individual A. cooperi ticks resulted in two isolates of R. bellii and one isolate genotypically characterized as an SFG rickettsia. The two R. bellii isolates were established in Vero cell cultures in the laboratory and were confirmed to be R. bellii by molecular analysis of the gltA and 17-kDa protein-encoding genes and by electron microscopic analysis. The SFG rickettsial isolate could not be stably passaged in cell culture in the laboratory, but molecular analysis of early passages suggested that it was closely related to Rickettsia parkeri, Rickettsia africae, and Rickettsia sibirica. These results do not support the role of A. cooperi in the ecology of R. rickettsii in the area studied, but they add two more species of rickettsiae to the poorly developed list of species occurring in ticks in South America.

Abstract: Aim of the study was to compare the trimming and non-trimming variants of the microtensile technique with the 'micro' push-out test in the ability to measure accurately the bond strength of fiber posts luted inside root canals. In 15 endodontically treated teeth (Group A), fiber posts were cemented with Excite DSC in combination with Variolink II (Ivoclar-Vivadent). In 15 roots RelyX Unicem (3M-ESPE) was used for fiber post luting (Group B). Within each group, the bond strength of cemented fiber posts was assessed with the trimming and non-trimming microtensile technique, as well as with the push-out test. The great number of premature failures (16.9% in Group A, 27.5% in Group B) and the finding of high standard deviation values make questionable the reliability of the trimming microtensile technique. With the non-trimming microtensile technique, only five sticks were obtained from a total of six roots. The remaining specimens failed prematurely during the cutting phase. With the push-out test no premature failure occurred, the variability of the data distribution was acceptable, and regional differences in bond strength among root levels could be assessed. Relatively low values of bond strength were, in general, recorded for luted fiber posts. In conclusion, when measuring the bond strength of luted fiber posts, the push-out test appears to be more dependable than the microtensile technique.

Abstract: Objectives: The conical beam computed tomography (CBCT) technique presents an innovation of tomographic imaging systems and subsequent volumetric image reconstruction for dentistry. When compared with other methods of tomographic imaging CBCT is characterized by rapid volumetric image acquisition from a single low radiation dose scan of the patient. The NewTom (NewTom 9000; Quantitative Radiology, Verona, Italy) is an example of such a CBCT machine dedicated to dental and maxillofacial imaging, particularly for surgical and/or prosthetics implant planning in the field of dentistry. The aim of this study was to evaluate the accuracy of the linear measurements obtained in CBCT images using a NewTom. Methods: Thirteen measurements were obtained in dry skulls (n=8) between internal and external anatomical sites using a caliper. These were considered as real measurements. Abstract: Molecular dynamics simulations of room temperature molten salts (ionic liquids) containing imidazolium cations have been performed. Ten different systems were simulated at 323 K by using united atom force fields, in which the anion size (F−, Cl−, Br−, and PF6−) and the length of the alkyl chain of 1-alkyl-3-methylimidazolium cations (1-methyl-, 1-ethyl-, 1-butyl-, and 1-octyl-) were systematically varied. It is shown that the resulting equilibrium structures account for the observed features of experimental static structure factors when available. A detailed analysis of the simultaneous effect of changing the anion and the alkyl chain on the preferential location of nearest-neighbor anions around the cations is provided. It is shown that regions above and below the imidazolium ring are the preferential ones in case of large anions. By increasing the length of the alkyl chain, nearest-neighbor anions are pushed away from the volume occupied by the flexible alkyl chain. Partial structure factors of 1-butyl-...

Abstract: We studied the time- and frequency-domain indexes of heart rate variability (HRV) in 653 patients without any evidence of heart disease relative to age, gender, heart rate, body mass index, and functional capacity. There was an inverse correlation of HRV with heart rate (p <0.001). HRV indexes decreased with increasing age, differed by gender, and were higher in patients with higher functional capacity. No correlation was noted between HRV and body mass index.

Abstract: OBJECTIVE: To update the social distribution of women's obesity in the developing world and, in particular, to identify the specific level of economic development at which, if any, women's obesity in the developing world starts to fuel inequities in health. DESIGN: Multilevel logistic regression analyses applied to anthropometric and socioeconomic data collected by nationally representative cross-sectional surveys conducted from 1992 to 2000 in 37 developing countries within a wide range of world regions and stages of economic development (gross national product (GNP) from US$190 to 4440 per capita). SUBJECTS: In total, 148 579 nonpregnant women aged 20–49 y. MEASUREMENTS: Body mass index to assess obesity status; quartiles of years of education to assess woman's socioeconomic status (SES), and GNP per capita to assess country's stage of economic development. RESULTS: Belonging to the lower SES group confers strong protection against obesity in low-income economies, but it is a systematic risk factor for the disease in upper-middle income developing economies. A multilevel logistic model—including an interaction term between the country's GNP and each woman's SES—indicates that obesity starts to fuel health inequities in the developing world when the GNP reaches a value of about US$2500 per capita. CONCLUSIONS: For most upper-middle income economies and part of the lower-middle income economies, obesity among adult women is already a relevant booster of health inequities and, in the absence of concerted national public actions to prevent obesity, economic growth will greatly expand the list of developing countries where this situation occurs.

Abstract: Objective To describe the prevalence of DSM-IV disorders and the pattern of comorbidity in a population-based sample of 7- to 14-year-old Brazilian schoolchildren. Method Random sampling of schools (stratified into private, public rural, and public urban) was followed by random sampling of pupils from school lists. In 2000–2001, a total of 1,251 children were assessed for DSM-IV diagnoses using the Development and Well-Being Assessment, a structured multiinformant assessment supplemented by verbatim reports reviewed by clinicians. Results The response rate was 83%. The overall prevalence of DSM-IV disorders was 12.7% (95% confidence interval = 9.8%–15.5%), with 3.5% of children being assigned as not otherwise specified rather than operationalized diagnoses. The overall prevalence of psychiatric disorder was significantly higher than in a British study with the same measures and diagnostic procedures (12.7% versus 9.7%, p = .02). Conclusions Approximately one in eight schoolchildren in the study area in the southeast of Brazil have psychiatric disorders involving a level of distress or social impairment likely to warrant treatment. The gulf between need and provision is currently vast.

Abstract: We present STAR measurements of the azimuthal anisotropy parameter v{sub 2} and the binary-collision scaled centrality ratio R{sub CP} for kaons and lambdas ({Lambda} + {bar {Lambda}}) at mid-rapidity in Au+Au collisions at {radical}s{sub NN} = 200 GeV. In combination, the v{sub 2} and R{sub CP} particle-type dependencies contradict expectations from partonic energy loss followed by standard fragmentation in vacuum. We establish p{sub T} {approx} 5 GeV/c as the value where the centrality dependent baryon enhancement ends. The K{sub S}{sup 0} and {Lambda} + {bar {Lambda}} v{sub 2} values are consistent with expectations of constituent-quark-number scaling from models of hadron formation by parton coalescence or recombination.

Abstract: Rutile is an important carrier of high field strength elements (HFSE; Zr, Nb, Mo, Sn, Sb, Hf, Ta, W). Its Zr content is buffered in systems with quartz and zircon as coexisting phases. The effects of temperature (T) and pressure (P) on the Zr content in rutile have been empirically calibrated in this study by analysing rutile–quartz–zircon assemblages of 31 metamorphic rocks spanning a T range from 430 to 1,100°C. Electron microprobe measurements show that Zr concentrations in rutile vary from 30 to 8,400 ppm across this temperature interval, correlating closely with metamorphic grade. An uncertainty in absolute T of ±50°C is inherited from T estimates of the natural samples used. A close approach to equilibrium of Zr distribution between zircon and rutile is suggested based on the high degree of reproducability of Zr contents in rutiles from different rock types from the same locality. At a given locality, the calculated range in T is mostly ±10°C, indicating the geological and analytical precision of the rutile thermometer. Possible applications of this new geothermometer are discussed covering the fields of ultrahigh temperature (UHT) granulites, sedimentary provenance studies and metamorphic field gradients.

Abstract: The objective of the present investigation was to compare the sensitivity of an electronic nociceptive mechanical paw test with classical mechanical tests to quantify the intensity variation of inflammatory nociception. The electronic pressure-meter test consists of inducing the hindpaw flexion reflex by poking the plantar region with a polypropylene pipette tip adapted to a hand-held force transducer. This method was compared with the classical von Frey filaments test and with the rat paw constant pressure test, a modification of the Randall and Selitto test developed by our group. When comparing the three methods, the electronic pressure-meter and the rat paw constant pressure test, but not the von Frey filaments test, detected time vs treatment interactions in prostaglandin E2 (PGE2)-induced hypernociception. Both methods also detected the PGE2-induced hypernociception in dose- (50-400 ng/paw) and time- (1-4 h) dependent manners, and time vs treatment interactions induced by carrageenin (25-400 microg/paw). Furthermore, the electronic pressure-meter test was more sensitive at early times, whereas the constant pressure test was more sensitive at later times. Moreover, the electronic pressure-meter test detected the dose-dependent antinociceptive effect of local indomethacin (30-300 microg/paw) and dipyrone (80-320 microg/paw) on carrageenin- (200 microg/paw) and PGE2- (100 ng/paw) induced hypernociception, respectively, and also detected the ineffectiveness of indomethacin (300 microg) on the effect of PGE2. Our results show that the electronic pressure-meter provides a sensitive, objective and quantitative mechanical nociceptive test that could be useful to characterize new nociceptive inflammatory mediators and also to evaluate new peripheral analgesic substances.

Abstract: Deficient activities of multiple steroidogenic enzymes have been reported without and with Antley-Bixler syndrome (ABS), but mutations of corresponding cytochrome P450 enzymes have not been found. We identified mutations in POR, encoding P450 oxidoreductase, the obligate electron donor for these enzymes, in a woman with amenorrhea and three children with ABS, even though knock-out of POR is embryonically lethal in mice. Mutations of POR also affect drug-metabolizing P450 enzymes, explaining the association of ABS with maternal fluconazole ingestion.
