Abstract: Consensus linkage maps are important tools in crop genomics. We have assembled a high-density tetraploid wheat consensus map by integrating 13 data sets from independent biparental populations involving durum wheat cultivars (Triticum turgidum ssp. durum), cultivated emmer (T. turgidum ssp. dicoccum) and their ancestor (wild emmer, T. turgidum ssp. dicoccoides). The consensus map harboured 30 144 markers (including 26 626 SNPs and 791 SSRs) half of which were present in at least two component maps. The final map spanned 2631 cM of all 14 durum wheat chromosomes and, differently from the individual component maps, all markers fell within the 14 linkage groups. Marker density per genetic distance unit peaked at centromeric regions, likely due to a combination of low recombination rate in the centromeric regions and even gene distribution along the chromosomes. Comparisons with bread wheat indicated fewer regions with recombination suppression, making this consensus map valuable for mapping in the A and B genomes of both durum and bread wheat. Sequence similarity analysis allowed us to relate mapped gene-derived SNPs to chromosome-specific transcripts. Dense patterns of homeologous relationships have been established between the A- and B-genome maps and between nonsyntenic homeologous chromosome regions as well, the latter tracing to ancient translocation events. The gene-based homeologous relationships are valuable to infer the map location of homeologs of target loci/QTLs. Because most SNP and SSR markers were previously mapped in bread wheat, this consensus map will facilitate a more effective integration and exploitation of genes and QTL for wheat breeding purposes.

Abstract: MUTATIONS in the Schizosaccharotnyces pombe cdc8 gene impair cytokinesis1. Here we clone cdc8+ and find that it encodes a novel tropomyosin. Gene disruption results in lethal arrest of the cell cycle, but spore germination, cell growth, DNA replication and mitosis are all unaffected. Haploid cdc8 gene disruptants are rescued by expression of a fibroblast tropomyosin complementary DNA. Immunofluorescence microscopy of wild type and cdc8 gene distruptants indicates that cdc8 tropomyosin is present in two distinct cellular distributions: in dispersed patches, and during cytokinesis as a transient medial band. Collectively these results indicate that cdc8 tropomyosin has a specialized role which, we suggest, is to form part of the F-actin contractile ring at cytokinesis. These results establish the basis for further genetic studies of cytokinesis and of contractile protein function in S. pombe.

Abstract: Climate warming is predicted to reduce the extent of ice cover in the Arctic and, within the Hudson Bay region, the annual ice may be significantly decreased or entirely lost in the foreseeable future. The ringed seal (Phoca hispida), a key species that depends on sea ice, will likely be among the first marine mammals to show the negative effects of climatic warming. We used 639 ringed seals killed by Inuit hunters from western Hudson Bay (1991–1992, 1999–2001) to assess trends in recruitment relative to snow depth, snowfall, rainfall, temperature in April and May, North Atlantic Oscillation (NAO) from the previous winter, and timing of spring break-up. Snowfall and ringed seal recruitment varied from lower than average in the 1970s, to higher in 1980s and lower in 1990s. Prior to 1990, seal recruitment appeared to be related to timing of spring ice break-up which was correlated with the NAO. However, recent 1990–2001 environmental data indicate less snowfall, lower snow depth, and warmer temperatures in April and May when pups are born and nursed. Decreased snow depth, particularly below 32 cm, corresponded with a significant decrease in ringed seal recruitment as indicated by pups born and surviving to adults that were later harvested. Earlier spring break-up of sea ice together with snow trends suggest continued low pup survival in western Hudson Bay.
