Abstract: In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. For example, a key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process versus those that measure flux through the autophagy pathway (i.e., the complete process including the amount and rate of cargo sequestered and degraded). In particular, a block in macroautophagy that results in autophagosome accumulation must be differentiated from stimuli that increase autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. It is worth emphasizing here that lysosomal digestion is a stage of autophagy and evaluating its competence is a crucial part of the evaluation of autophagic flux, or complete autophagy. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. Along these lines, because of the potential for pleiotropic effects due to blocking autophagy through genetic manipulation, it is imperative to target by gene knockout or RNA interference more than one autophagy-related protein. In addition, some individual Atg proteins, or groups of proteins, are involved in other cellular pathways implying that not all Atg proteins can be used as a specific marker for an autophagic process. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular assays, we hope to encourage technical innovation in the field.

Abstract: Geant4 is a software toolkit for the simulation of the passage of particles through matter. It is used by a large number of experiments and projects in a variety of application domains, including high energy physics, astrophysics and space science, medical physics and radiation protection. Over the past several years, major changes have been made to the toolkit in order to accommodate the needs of these user communities, and to efficiently exploit the growth of computing power made available by advances in technology. The adaptation of Geant4 to multithreading, advances in physics, detector modeling and visualization, extensions to the toolkit, including biasing and reverse Monte Carlo, and tools for physics and release validation are discussed here.

Abstract: The main objective of this two-part state-of-the-art paper called “Recent Advances in the Design, Modeling, and Control of Multiphase Machines” is to present latest contributions in the multiphase machines' field. The first part of this paper focuses on the recent progress in the design, modeling, and control, whereas the drive is in healthy operation. This second part presents relevant contributions in two not analyzed fields. The first is in relation with the use of the additional degrees of freedom of multiphase machines and the exploitation of their fault-tolerant capabilities without adding extra hardware. The second one analyzes multiphase generation, particularly in grid-connected wind energy conversion systems and stand-alone applications. Recent progresses are shown and open challenges and future research directions are discussed.

Abstract: The cost reduction of power-electronic devices, the increase in their reliability, efficiency, and power capability, and lower development times, together with more demanding application requirements, has driven the development of several new inverter topologies recently introduced in the industry, particularly medium-voltage converters. New more complex inverter topologies and new application fields come along with additional control challenges, such as voltage imbalances, power-quality issues, higher efficiency needs, and fault-tolerant operation, which necessarily requires the parallel development of modulation schemes. Therefore, recently, there have been significant advances in the field of modulation of dc/ac converters, which conceptually has been dominated during the last several decades almost exclusively by classic pulse-width modulation (PWM) methods. This paper aims to concentrate and discuss the latest developments on this exciting technology, to provide insight on where the state-of-the-art stands today, and analyze the trends and challenges driving its future.

Abstract: A test oracle determines whether a test execution reveals a fault, often by comparing the observed program output to the expected output. This is not always practical, for example when a program's input-output relation is complex and difficult to capture formally. Metamorphic testing provides an alternative, where correctness is not determined by checking an individual concrete output, but by applying a transformation to a test input and observing how the program output “morphs” into a different one as a result. Since the introduction of such metamorphic relations in 1998, many contributions on metamorphic testing have been made, and the technique has seen successful applications in a variety of domains, ranging from web services to computer graphics. This article provides a comprehensive survey on metamorphic testing: It summarises the research results and application areas, and analyses common practice in empirical studies of metamorphic testing as well as the main open challenges.

Abstract: The increasing penetration of renewable energy sources (RES) poses a major challenge to the operation of the electricity grid owing to the intermittent nature of their power output. The ability of utility-scale battery energy storage systems (BESS) to provide grid support and smooth the output of RES in combination with their decrease in cost has fueled research interest in this technology over the last couple of years. Power electronics (PE) is the key enabling technology for connecting utility-scale BESS to the medium-voltage grid. PE ensure energy is delivered while complying with grid codes and dispatch orders. Simultaneously, the PE must regulate the operating point of the batteries, thus for instance preventing overcharge of batteries. This paper presents a comprehensive review of PE topologies for utility BESS that have been proposed either within industry or the academic literature. Moreover, a comparison of the presently most commercially viable topologies is conducted in terms of estimated power conversion efficiency and relative cost.

Abstract: Research on stabilization of coupled hyperbolic PDEs has been dominated by the focus on pairs of counter-convecting (“heterodirectional”) transport PDEs with distributed local coupling and with controls at one or both boundaries. A recent extension allows stabilization using only one control for a system containing an arbitrary number of coupled transport PDEs that convect at different speeds against the direction of the PDE whose boundary is actuated. In this paper we present a solution to the fully general case, in which the number of PDEs in either direction is arbitrary, and where actuation is applied on only one boundary (to all the PDEs that convect downstream from that boundary). To solve this general problem, we solve, as a special case, the problem of control of coupled “homodirectional” hyperbolic linear PDEs, where multiple transport PDEs convect in the same direction with arbitrary local coupling. Our approach is based on PDE backstepping and yields solutions to stabilization, by both full-state and observer-based output feedback, and trajectory tracking problems.

Abstract: Escherichia coli is one of the most-studied microorganisms worldwide but its characteristics are continually changing. Extraintestinal E. coli infections, such as urinary tract infections and neonatal sepsis, represent a huge public health problem. They are caused mainly by specialized extraintestinal pathogenic E. coli (ExPEC) strains that can innocuously colonize human hosts but can also cause disease upon entering a normally sterile body site. The virulence capability of such strains is determined by a combination of distinctive accessory traits, called virulence factors, in conjunction with their distinctive phylogenetic background. It is conceivable that by developing interventions against the most successful ExPEC lineages or their key virulence/colonization factors the associated burden of disease and health care costs could foreseeably be reduced in the future. On the other hand, one important problem worldwide is the increase of antimicrobial resistance shown by bacteria. As underscored in the last WHO global report, within a wide range of infectious agents including E. coli, antimicrobial resistance has reached an extremely worrisome situation that ‘threatens the achievements of modern medicine’. In the present review, an update of the knowledge about the pathogenicity, antimicrobial resistance and clinical aspects of this ‘old friend’ was presented.

Abstract: The use of multivalent carbohydrate compounds to block cell-surface lectin receptors is a promising strategy to inhibit the entry of pathogens into cells and could lead to the discovery of novel antiviral agents. One of the main problems with this approach, however, is that it is difficult to make compounds of an adequate size and multivalency to mimic natural systems such as viruses. Hexakis adducts of [60]fullerene are useful building blocks in this regard because they maintain a globular shape at the same time as allowing control over the size and multivalency. Here we report water-soluble tridecafullerenes decorated with 120 peripheral carbohydrate subunits, so-called 'superballs', that can be synthesized efficiently from hexakis adducts of [60]fullerene in one step by using copper-catalysed azide–alkyne cycloaddition click chemistry. Infection assays show that these superballs are potent inhibitors of cell infection by an artificial Ebola virus with half-maximum inhibitory concentrations in the subnanomolar range.

Abstract: Although the concept of a multiphase drive system dates back to the middle of the 20th century, the initial pace of development was rather slow, as witnessed by the first two surveys of the area published in the beginning of this century. However, considerably new developments have resulted in the last decade of the 20th century and the beginning of this century, leading to an authoritative survey of the asymmetrical six-phase drive control and subsequently of the review of the complete area. This also initiated the organization and subsequent publication of the first IEEE Transactions on Industrial Electronics "Special Section on Multiphase Machines and Drives" in May 2008, which commenced with another survey paper, and that contained 12 original research papers. Since the publication of this Special Section in May 2008, the level of interest and pace of developments in the area have further accelerated and substantial new knowledge has been generatedwith an ever-increasing number of published research papers and reported new industrial applications. Such a trend has been emphasized in a recent paper. It therefore seemed appropriate to revisit the area and organize this Special Section as a sequel to the first one. The call for the Special Section papers resulted in 51 submissions, almost twice as many as the total back in 2008, thus confirming a substantial growth of the area. Indeed, the amount of new knowledge acquired since the publication of the first Special Section in 2008 has meant that it was not possible to provide a complete and thorough survey of the field in a single review paper.

Abstract: Soil and water losses due to agricultural mismanagement are high and non-sustainable in many orchards. An experiment was set up with rainfall simulation at 78 mm h–1 over 1 hour on 20 paired plots of 2 m2 (bare and straw covered) in new persimmon plantations in Eastern Spain. Effects of straw cover on the control of soil and water losses were assessed. An addition of 60% straw cover (75 g m–2) resulted in delayed ponding and runoff generation and consequently reduced water losses from 60% to 13% of total rainfall. The straw cover reduced raindrop impact and thus sediment detachment from 1014 to 47 g plot–1 h–1. The erosion rate was reduced from 5.1 to 0.2 Mg ha–1 h–1. The straw mulch was found to be extremely efficient in reducing soil erosion rates.

Abstract: Evaluation of the environmental impact caused by construction materials frequently presents such obstacles as the mismatch between the construction project location and where the LCA database was made, lack of transparency, and/or the unsuitability of the data to the building project conditions, thereby making it necessary to establish a state-of-the-art review for researchers in order to facilitate selection between the wide variety of databases available. A review of existent LCA databases containing data for building materials has been performed. A list of features and criteria for their evaluation is developed, and subsequently applied in order to compare the various databases. Their methodology, documentation, data quality and comprehensiveness are thereby analysed. Despite the existence of a considerable number of databases, only a few contain data on construction materials. Some projects have been abandoned and several more can be considered incomplete. However, GaBi Database and Ecoinvent stand out for their integrity, usability and dedicated resources. A starting point in the selection of an LCA database for construction materials is provided. With all the information gathered herein, researchers are equipped to make a well-founded choice, and the selection process is certainly improved.

Abstract: One of the most attractive features of multiphase machines is the fault-tolerant capability due to the higher number of phases. Different postfault control strategies based on hysteresis, proportional integral (PI)-resonant, and predictive techniques have been recently proposed. They all proved their capabilities to withstand fault situations and to preserve the fundamental component of the air-gap field, while achieving minimum losses, maximum torque per ampere, and reducing torque vibrations. Nonetheless, due to their recent introduction, no thorough study has yet appeared comparing the performance of these controllers. In this paper, two open-phase fault-tolerant control schemes are experimentally compared in a real five-phase induction machine. The controllers being compared are based on PI-resonant and predictive control techniques, respectively. The experiments include pre- and postfault situations. Obtained results show that both control methods offer nearly the same performance. When compared, predictive control provides faster control response and superior performance at low-speed operation but is found to be less resilient to fault detection delays and to have higher current ripple. Regarding the controller implementation, it is shown that the transition from prefault to postfault operation involves modeling the nonlinear effects observed when an open-phase fault occurs for the predictive controller and proper retuning of the PI trackers for the PI-resonant controller, to ensure postfault operation.

Abstract: The production of liquid biofuels to blend with gasoline is of worldwide importance to secure the energy supply while reducing the use of fossil fuels, supporting the development of rural technology with knowledge-based jobs and mitigating greenhouse gas emissions. Today, engineering for plant construction is accessible and new processes using agricultural residues and municipal solid wastes have reached a good degree of maturity and high conversion yields (almost 90% of polysaccharides are converted into monosaccharides ready for fermentation). For the complete success of the 2G technology, it is still necessary to overcome a number of limitations that prevent a first-of-a-kind plant from operating at nominal capacity. We also claim that the triumph of 2G technology requires the development of favourable logistics to guarantee biomass supply and make all actors (farmers, investors, industrial entrepreneurs, government, others) aware that success relies on agreement advances. The growth of ethanol production for 2020 seems to be secured with a number of 2G plants, but public/private investments are still necessary to enable 2G technology to move on ahead from its very early stages to a more mature consolidated technology.

Abstract: The reliable generation of micron-sized droplets is an important process for various applications in droplet-based microfluidics. The generated droplets work as a self-contained reaction platform in droplet-based lab-on-a-chip systems. With the maturity of this platform technology, sophisticated and delicate control of the droplet generation process is needed to address increasingly complex applications. This review presents the state of the art of active droplet generation concepts, which are categorized according to the nature of the induced energy. At the liquid/liquid interface, an energy imbalance leads to instability and droplet breakup.
